    Notes: Abstract. The molecular diversity of inhibitor-resistant TEM (IRT) enzymes was explored using a strategy which involved DNA amplification by polymerase chain reaction (PCR), analysis of restriction fragment length polymorphism (RFLP), and direct nucleotide sequencing. The study of plasmid-borne genes from 27 strains, resistant to amoxicillin and β-lactamase-inhibitor combinations, identified mutations resulting in amino acid change at positions 69, 244, 275, and 276 known to be associated with the IRT phenotype and a mutation at nucleotide position 162 in the promoter region. These mutations were found to lie on two different gene sequences, described here as ``TEM-1B like'' and ``TEM-2 like'' restriction linkage groups. Further analysis, of nucleotide sequences of promoter and coding regions of the β-lactamases, confirmed that a given mutation causing IRT phenotype could be associated with two different gene sequence frameworks and two different causal mutations could lie on identical gene sequence framework. These data argue in favor of convergent phenotypic evolution of IRT enzymes under the selective pressure imposed by the intensive clinical use of β-lactam–β-lactamase inhibitor combinations.

    Notes: Using microdiffraction techniques at the Advanced Photon Source, we have found evidence for local molecular alignment within the mesoscopic fibers of DNA–cationic membrane complexes, a system originally conceived as gene delivery vectors. Furthermore, these mesoscopic structures can be manipulated by using lithographically patterned microchannel arrays, so that the optical axes of individual fibers are aligned parallel to the channels. These observations suggest that DNA–membrane complexes may have additional technological applications, such as active electrophoretic media and templates for the fabrication of semiconductor superlattices and nanoporous materials with tunable pore sizes. © 1998 American Institute of Physics.

    Notes: Abstract: A synthetic peptide (25 amino acids) corresponding to a specific portion of the third intracytoplasmic loop of the rat serotonin 5-HT1B/1Dβ receptor was coupled to keyhole limpet hemocyanin and injected monthly into rabbits. Anti-peptide antibodies were detected by enzyme-linked immunosorbent assay and characterized by immunoprecipitation of the 5-HT1B/1Dβ receptor in CHAPS-solubilized extracts from rat striatal membranes. Up to 60% of solubilized striatal serotonin-O-carboxymethylglycyl[125I]iodotyrosinamide ([125I]GTI; a selective 5-HT1B/1D radioligand) binding sites were immunoprecipitated and subsequently pharmacologically identified as 5-HT1B receptors. The remaining 40% of [125I]GTI binding sites were shown to be 5-HT1D receptors. In addition, these antibodies were successfully used in immunofluorescence experiments to detect the 5-HT1B/1Dβ, but not the 5-HT1D/1Dα, receptor in transiently transfected LLC-PK1 cells. Immunoautoradiographic experiments performed with brain sections from the rat, mouse, and guinea pig showed that the substantia nigra and globus pallidus contained the highest densities of 5-HT1Dβ receptor-like immunoreactivity. Comparison of the regional distribution of immunolabeling with that of the specific binding of [125I]GTI in the brain of these species further confirmed that the anti-peptide antibodies selectively recognized only the 5-HT1Dβ component of [125I]GTI specific receptor binding sites.

    Notes: Results are presented on development of processes for fabricating linear and circular Bragg–Fresnel lenses (BFLs) on Si and III–V compound material substrates, and on x-ray characterization of linear BFLs at the Advanced Photon Source (APS). Processes were developed for fabricating long (zone length 〉5 mm) linear BFLs on Si with enhanced capability for focusing high-energy x rays. By stitching together 20 sequentially exposed 400-μm-long linear BFLs, we were able to fabricate 8-mm-long linear BFLs with 0.5-μm finest zone width. BFLs were also fabricated on III–V compound semiconductor substrates GaAs and InP, with improved process control due to the substantially reduced zone thickness required (∼50% less than Si). Reduction of the zone aspect ratio (thickness/width) lessens the demand on the process technology, and may lead to higher lens resolution and pattern transfer accuracy. A process was explored to fabricate BFLs on a GaAs/AlGaAs heterostructure incorporating a built-in "etch stop" layer to ensure uniform zone thickness. Experimental characterization of the focusing properties of a field-stitched 8-mm-long linear BFL on Si (zone aperture=150 μm) was conducted at APS undulator beamline 1-ID-C using 10-keV x rays. Based on measured focal plane intensity distribution, the focus was estimated to be 1.2 μm, comparable to the geometrically demagnified source size of ∼1 μm. Lens efficiency was estimated to be ∼30%. Work is underway to incorporate the BFL-microprobe in x-ray microdiffraction and fluorescence microscopy experiments to study spatially confined complex materials. © 1998 American Institute of Physics.

    Notes: New treatments for chronic wounds require carefully performed clinical trials with significant endpoints. Total wound closure is the only endpoint currently accepted by the Food and Drug Administration. This study describes a scale that measures ease of wound closure and applies it to a four-arm prospectively randomized, blinded pressure ulcer trial of recombinant human platelet-derived growth factor-BB. Following validation of interrater reliability, 83 evaluable subjects' photographs were given a weekly ease of closure score by four raters blinded to treatment. The change of ease of closure score was correlated with the change of wound area and volume. Each ease of closure score was given a procedural cost. Results showed ease of closure did not directly correlate with either wound area or volume, suggesting that it was measuring additional information. The mean change in ease of closure score was 6 for subjects treated with 100 μg recombinant human platelet-derived growth factor-BB daily; 5 for those treated with 300 μg growth factor daily or 100 μg recombinant human platelet-derived growth factor-BB bid; and 4 for those treated with placebo. The cost savings ranged from $7200 for the group receiving 100 μg recombinant human platelet-derived growth factor-BB daily to $6300 for the controls. Outcomes in all 4 groups were significantly improved from their starting evaluation (p 〈 0.001). Based on this study, ease of closure is a verifiable endpoint that can be related to cost efficiency and may be a measure of efficacy.

    Notes: A panel of public health practitioners sponsored by the Health Resources and Services Administration met December 6–8, 1994, to examine current roles and responsibilities for dental public health workers and to recommend changes in education and training to meet challenges posed by an evolving health care system. Overall, at least the same number, if not more, dental public health personnel will be needed in the future. While some new roles were identified, the panel felt that only small numbers of personnel will be needed to fill these new roles. Not all of these roles necessarily require a dental degree. The panel felt that a need exists for more academicians for dental schools, schools of public health, dental public health residencies, and dental hygiene programs; oral epidemiologists and health services researchers; health educators; and specialists in utilization review/outcomes assessment, dental informatics, nutrition, program evaluation, and prevention. To meet these personnel needs: (1) dental public health residency programs should be structured to meet the educational needs of working public health dentists with MPH degrees through on-the-job residency programs; (2) the standards for advanced specialty education programs in dental public health should be made sufficiently flexible to include dentists who have advanced education and the requisite core public health courses; (3) flexible MPH degree programs must be available because of the rising debt of dental students and the decreased numbers of graduating dentists; (4) loan repayment should be available for dentists who have pursued public health training and are working in state or local health departments; and (5) standards for advanced education in dental public health should be developed for dental hygienists.

    Notes: Abstract The production of β-lactamase in Streptomyces cacaoi, which contains two β-lactamase-encoding genes, blaL and blaU, is inducible by β-lactam compounds. The two genes have been cloned independently in S. lividans TK24, a β-lactamase-negative species. The blaU clone did not respond to the presence of β-lactams, whereas the blaL clone appeared to be inducible in S. lividans. The latter clone contains two open reading frames, blaA and blaB, located just upstream of but transcribed divergently from blaL, which were shown to be required for the production as well as the induction of BlaL. The deduced BlaA protein belongs to the LysR family of transcription regulators. In order to examine the role of BlaA in regulation, we here report on overexpression of a GST-BlaA fusion protein in Escherichia coli and its use for antibody preparation. The GST-BlaA fusion protein was partially purified and bandshift assays showed that it bound the 197-bp blaL-blaA intergenic region. The BlaA DNA binding-site was further restricted to a 30-bp sequence containing a T-N11-A motif, a characteristic of LysR-type promoters. Another T-N11-A motif upstream of the blaU gene was also shown to bind BlaA. The affinities of these two T-N11-A motifs in BlaA binding were comparable. A plasmid bearing the blaU structural gene and the blaA-blaB regulatory region was constructed and shown to confer on an S. lividans host the capacity to produce inducible β-lactamase. It can thus be concluded that the S. cacaoiblaL and blaU genes are controlled by the same regulatory system.
