Notes: Abstract Aims/hypothesis. Although increased polyol pathway activity has been implicated in the pathogenesis of diabetic microangiopathy, the relation with diabetic macroangiopathy remains unclear. Galactose feeding is known to stimulate the polyol pathway and to develop abnormalites similar to those in diabetic microangiopathy. Our study was conducted to investigate whether an activation of polyol pathway by long-term treatment with galactose produced morphological changes in coronary arteries of dogs and the effect of an aldose reductase inhibitor, epalrestat, was also studied.¶Methods. Dogs received either normal chow or chow containing 30 % galactose with or without epalrestat given orally (20 or 50 mg · kg–1). After 44 months, morphometric analyses of coronary arteries were carried out and the galactitol contents in aortas were measured.¶Results. The ratio of areas of the intimal layer to those of the medial layer, an indicator of intimal thickening, was statistically significantly increased in galactose-fed dogs compared with control dogs. Galactose-fed dogs had a remarkable accumulation of galactitol in their aortas. These morphological and biochemical deficits were reduced by treatment with epalrestat.¶Conclusion/interpretation. This report morphologically shows diabetes-like macrovascular abnormalities in galactosaemic animals, suggesting that polyol pathway hyperactivity is closely related to the development of diabetic macroangiopathy, which could be prevented by aldose reductase inhibition. Notes: Abstract  Analytical instruments used for measurements of air and water pollution are calibrated by using reference materials such as standard gases and standard solutions. In Japan, since the middle of the 1970s, those reference materials which are traceable to the national standards maintained at national research institutes have been supplied to users by reference material producers. In order to establish the primary standards and to secure the traceability from the working standards to the national ones, various analytical methods such as coulometric, titrimetric and gravimetric analyses for purity determination and highly sensitive atomic spectrometry for trace analysis have been developed as the primary methods and reference methods. The Japanese Measurement Law, revised in 1992, has introduced a new traceability system in which a public organization, a "designated calibration body", can also prepare and maintain the national standards under the advice and instruction of national research institutes. The designated calibration body can provide calibration services to reference material producers (accredited calibration bodies) by using the national standards. The reference materials supplied in conformity with the traceability system include standard gases, pH standard solutions, metal standard solutions and non-metal ion standard solutions.

Notes: In the Fock–Bargmann space, the complete harmonic-oscillator basis for the three-cluster system is constructed. The indices of the reduction U(6)&supuline;U(2)×U(3) are chosen as the quantum numbers. The Pauli-forbidden states are eliminated by the orthogonal transformation of basis functions. The basis states are obtained in terms of hypergeometric functions and the spherical Wigner functions. Their simple form allows one to solve the problem of calculating the matrix elements of the microscopic Hamiltonian needed for the study of three-cluster systems within the algebraic version of RGM. © 1995 American Institute of Physics.

Notes: The addition of chondroitin sulphate proteoglycans (CSPGs), purified from the rat brain, to the culture medium of PC12D cells inhibited their proliferation and neurite outgrowth. Therefore, we investigated the effects of several extracellular components on the inhibitory actions of CSPGs on PC12D cells, as well as their immunocytochemical distribution in the rat embryo to determine whether the findings in vitro could be reproduced in vivo. Coating of the substratum with polylysine was necessary for the appearance of the inhibitory effects of brain CSPGs on PC12D cells. The additional pretreatment of polylysine-coated dishes with laminin or fibronectin promoted the outgrowth of neurites from PC12D cells. Laminin and fibronectin, but not collagen (types I and IV) and CELL-TAK (cell adhesion molecules), prevented the inhibitory effects of brain CSPGs in a concentration-dependent manner. Doses producing 50% reduction by laminin (or fibronectin) of the CSPG effects were 1.5 (or 25) μg/ml for neurite outgrowth and 2.2 (or 28) μg/ml for proliferation. The ratio of dish-attached CSPGs to laminin necessary for 50% reduction was about ∼50:l (wt/wt). Laminin from any source had the same effect. Brain CSPGs also obviously impeded the growth of fibres from dorsal root ganglion explants and primary cultured dorsal root ganglion neurons. Neurocan (a major CSPG in the brain)-like immunoreactivity was detected in the boundary caps and roof plate in the rat embryo at 13.5 days of gestation, when DRG neurons were extending their axons to the neural tube. The distributions of laminin and tenascin appeared, respectively, to be slightly and considerably different from that of neurocan. These results suggest that brain CSPGs can generate barriers to the growth of axons from the dorsal root ganglion and that the ratio of CSPGs to laminin may be important in regulating such growth.

Notes: A homoeologous quantitative trait locus to that of eps5L on barley chromosome 5H was identified in a syntenic region of wheat chromosome 5A. Wheat single chromosome recombinant lines (SCRs) were developed from a cross between ‘Chinese Spring’(‘Cappelle-Desprez’ 5A) and ‘Chinese Spring’(Triticum spelta 5A), these were grown together with the parental controls under different vernalization and photoperiod regimes. The variation for ear emergence time accelerated heading induced by the T. spelta segment indicated an effect associated with the Xcdo412-Xbcd9 interval. Since no differences between the SCRs and controls in responses to vernalization and photoperiod treatments were detected, this effect was identified as an earliness per se gene, Q Eetocs-5 A.2, which may be homoeologous to the eps5L quantitative trait locus of barley. Xbcd926 has been found to be closely linked to the rice flowering time quantitative trait loci, QHd9a or FLTQ2, on chromosome 9, suggesting possible relationships among the quantitative trait loci across wheat, barley and rice genomes.

Notes: Immunohistochemical identification of myoepithelial cells using α-smooth muscle actin provides little information about the nature of solid or quasi-solid portions of epithelial hyperplasia and ductal carcinoma in situ (DCIS) because actin-rich myoepithelial cells are usually demonstrated only in the stromal–epithelial junction of both lesions. We studied the differential distribution of α-subunit (S100-α) and β-subunit (S100-β) of S100 protein in actin-negative areas of usual epithelial hyperplasia and DCIS by employing the streptavidin method with monospecific rabbit antibodies against each subunit. All usual epithelial hyperplasias (n=17) were composed of heterogeneous epithelial cell types; cells expressing S100-α and/or S100-β were intermingled with non-expressing cells, resulting in a mosaic-like pattern. On the contrary, DCIS (n=32) uniformly lacked immunoreactive S100-β; S100-α was diffusely expressed in 24 (68.8%) DCIS (three solid/comedo, 13 cribriform, four endocrine, one micropapillary, three papillary variants) and negative in the remaining eight (31.2%) DCIS (one cribriform, two micropapillary, four papillary and one apocrine variants). In conclusion, in contrast to usual epithelial hyperplasia that expresses both S100-α and S100-β in a heterogeneous pattern, DCIS can express only S100-α in a monotonous pattern, possibly signifying unidirectional differentiation toward secretory glandular epithelium.

Notes: To determine the importance of silkworm moth allergens, we tested 267 patients with allergic rhinitis with CAP-RAST®, a new assay system for detecting specific IgE in sera. We used the following allergen sources: house-dust mite, Candida, Altemaria, cat dander, orchard grass, ragweed, mugwort, Japanese hops, Japanese cedar, Japanese cypress, cotton, silk, larval chironomid midge, adult chironomid midge, and silkworm moth. As expected, the highest positive rate of reaction, as determined by CAP-RAST, was 73.8% for Japanese cedar, followed by 53.2% for mite and 50.6% for Japanese cypress. Although the positive rate for silkworm moth was not as high as for cedar pollen, one-third of patients had specific IgE against silkworm moth. The positive rate of reaction to silkworm moth was much higher than that to chironomid. It was interesting to note that the patients reported had not been documented as having frequent contact with silkworm moth allergen. Correlations between silkworm moth and silk, and silkworm moth and chironomid midge were found to be significant (P 〈0.001). Silkworm moth allergen showed the third highest reaction rate in patients with severe symptoms. These results suggest that silkworm moth allergen should be considered to be important in patients with allergic rhinitis in Japan.

Notes: The complex (2-cyanoethyl)bis(dimethylglyoximato)(triphenylphosphine)cobalt(III), [Co(C4H7N2O2)2(C3H4N){P(C6H5)3}], when it was crystallized rapidly from an aqueous methanol solution, has four crystal forms, three of which were obtained separately from different solvents in large amounts. The crystal structures of the four forms were determined by X-rays. The molecular structures in the four forms have different conformations around the Co—C and Co—P bonds. The packing energy calculation indicated that the four forms have approximately the same energy. The 2-cyanoethyl group of this complex was isomerized to the 1-cyanoethyl group when the powdered sample of the complex was irradiated with a xenon lamp. For three crystal forms prepared separately, the solid-state photoisomerization rates were measured from the change in the IR spectra, assuming first-order kinetics. A quantitative relationship between the rate constant and the size of the reaction cavity for the 2-cyanoethyl group has been obtained for the three crystal forms.
