    Notes: Abstract  Platinum-containing drugs enter the cell slowly and have a poor tissue penetration. Increasing the permeability of the cell membrane might increase the intracellular drug concentration. Digitonin, a detergent that increases cell permeability by binding to cholesterol molecules in the cell membrane, can increase cisplatin accumulation and reduce tumour growth in vitro. The aim of this study was to determine whether digitonin could increase the efficacy of carboplatin (CBDCA) in vivo. In LH rats, a hepatoma was implanted in the liver. At 7 days after implantation, digitonin (or saline in the control group) was infused via the hepatic artery and, 10 min later, CBDCA was injected. Biopsies from the tumour and liver parenchyma were obtained after 1 h. The concentration of platinum measured in the liver tumours was higher in the digitonin group than in the control groups. In the liver parenchyma the concentrations were of the same magnitude. Measured with the 133Xe-clearance technique, digitonin did not alter the tumour blood flow. Digitonin enhanced the tumour-growth-retarding effect of CBDCA given intra-aterially at 5 mg/kg but not at 25 mg/kg. No increase in toxicity was observed for digitonin given together with CBDCA at 5 mg/kg. Systemic administration of CBDCA was not influenced by digitonin. These findings demonstrate that pretreatment with digitonin increases the tumour uptake of CBDCA and potentiates the cytotoxic effect of CBDCA.

    Notes: Summary The shear orientation of a micellar hexagonal liquid crystalline phase was investigated by small-angle neutron scattering. The hexagonal phase in the quiescent state showed a symmetrical scattering pattern typical of a polydomain structure. Enhanced scattering along the flow direction was observed during shear and the anisotropy of scattering intensity became stronger with increasing shear rate. The anisotropic scattering pattern corresponds to an orientation perpendicular to the flow direction and can be interpreted as a log-rolling state. The oriented sample did not relax after cessation of shear. The results from small-angle neutron scattering confirm data obtained previously from rheo-small angle light scattering measurements and are discussed in comparison to shear alignment of lyotropic liquid crystalline polymer solutions.

    Notes: Abstract  The analysis of the interaction of micelles formed by a blockcopolymer is given by means of small-angle X-ray (SAXS) and small-angle neutron scattering (SANS). The blockcopolymer consists of poly(styrene) and poly(ethylene oxide) (molecular weight of each block: 1000 g/mol) and forms well-defined micelles (weight-association number: 400, weight-average diameter: 15.4 nm) in water. The internal structure has been studied previously (Macromolecules 29:4006 (1996)) by SAXS. There it has been shown that the micelles are spherical objects. The structure factor S(q) as a function of the scattering vector q (q=(4π/λ) sin (θ/2); λ: wavelength of the radiation in the medium; θ: scattering angle) can be extracted from both sets of small-angle scattering data (SANS: q≤0.4 nm-1; SAXS: q≤0.6 nm-1). It is shown that particle interaction in the present system can be described by assuming soft interaction which is modeled by a square-step potential.

    Notes: Abstract Protein digestibility by gastrointestinal homogenates of Dicentrarchus labrax, was investigated in 5 feeds in relation to their amino acids composition and their ability to sustain growth of D. labrax post larval juveniles. Gastric proteolysis was found to contribute 〈25% of the total proteolytic capacity. Correlation was found between intestinal proteolytic capacity and the feed protein content of three pairs of amino acids. The sum of basic amino acids, arginine + lysine in the feed protein exhibited a positive correlation with susceptibility to proteolysis. The sum of the acid amino acids glycine and proline showed a negative correlation. Comparison of the intestinal proteolytic enzymatic activities in 3 growth stages of this species revealed similar profiles. Post larval and large juveniles had similar proteolytic capacities when compared on the basis of equal activity on an artificial substrate of Trypsin. Specific growth rates of post larval fish fed on these 5 feeds correlated positively with their protein digestibility as measured with large juvenile's intestinal homogenates. The results suggest that in this species, the efficiency of feeds in sustaining growth at the post larval stage can be estimated by measuring the feed protein digestibility by the proteolytic system of larger sized juveniles.

    Notes: Abstract: We have investigated the simple shear flow behavior of wormlike micelles using small-angle neutron scattering and mechanical measurements. Ternary surfactant solutions made of cetylpyridinium chloride, hexanol and brine (0.2 M NaCl) and hereafter abbreviated as CPCl-Hex were studied in the concentrated regime, . In a preliminary report (Berret et al. [#!ref16!#]), the discontinuity of slope observed in the shear stress versus shear rate curve was interpreted in terms of first-order phase transition between an isotropic state and a shear-induced nematic state ( transition). At the transition rate, , the solution exhibits a macroscopic phase separation into viscous and fluid layers (inhomogeneous shear flow). Above a second characteristic shear rate, the flow becomes homogeneous again, the sheared solution being nematic only. The neutron patterns obtained in the two-state inhomogeneous region have been re-examined. Based on a consistent analysis of both orientational and translational degrees of freedom related to the wormlike micelles, we emphasize new features for the transition. In the present paper, the shear rate variations of the relative proportions of each phase in the two-state region, as well as the viscosity ratio between isotropic and nematic phases are derived. We demonstrate in addition that slightly above the transition rate, the shear induced nematic phase is already strongly oriented, with an order parameter P 2 = 0.65. The orientational state is that of a nematic flow-oriented monodomain. Finally, from the locations of the neutron scattering maxima for each isotropic and nematic contributions, we evaluate the concentrations for each phase and and derived a dynamical phase diagram of CPCl-Hex, in terms of the stress versus and . According to the classification by Schmitt et al. [#!ref22!#], the transition observed in CPCl-Hex micellar solutions could result from a positive flow-concentration coupling, in agreement with the observed monotonically increasing shear stress in the two-phase region.

    Notes: Abstract: We study the behavior under flow of soft spherical micelles forming a fcc phase at high volume fraction. Due to the size (300 Å) of the elementary objects, the structure can be investigated through X-rays and neutron scattering, at rest and under flow in a Couette cell. Using scattering in two perpendicular directions, different mechanisms of flow are identified. At intermediate shear (around 100 s-1) the system exhibits the so called layer sliding mechanism where two dimensional hexagonal compact planes of spheres align themselves with the Couette cell walls. At lower shear rate, the fcc structure is locally preserved, and the flow is mediated by defects between crystallites. At high shear rate, we observe the melting of the structure and a liquid-like structure factor. Moreover, we were able to use the existence of the layer sliding regime to generate a fcc monocrystal by annealing the satcking faults between the decorrelated planes created by the layer sliding.
