    Notes: The use of pipeline feedback to ensure good control of material flow systems has been developed over the years on a pragmatic basis. More recently, the mechanism by which the improved control is achieved has been the subject of theoretical analysis. In turn, this has led to recommendations for good parameter settings which may be used with confidence when applied to a particular generic decision support system (DDS) known as the "to make" model. One consequence of utilizing pipeline feedback is the enhanced damping capability of this system. In our experience, many supply chains may be represented by the coupling together of a series of To-Make models. In this paper, we show that the use of supply chain feedback within each echelon greatly reduces the order amplification as it proceeds upstream from the market place. Using as an example a model of the Beer Game Supply chain, it is concluded that demand amplification is readily reduced by a factor of 2:1.

    Notes: Abstract Background: Recent studies have demonstrated the usefulness of gene-modified tumor cells for immunotherapy. Using the tumorigenic murine fibrosarcoma, MCA 106, we investigated the effects of localized interferon-γ (IFNg) secretion on tumorigenicity and on long-term memory. Methods: The murine IFNg (MuIFNg) gene was introduced into tumor cells. High and low IFNg-secreting clones were isolated. C57BL/6 mice were injected subcutaneously (s.c.) with either parental (P), high or low IFNg-secreting (H- or L-IFNg) cells, and tumor growth was assessed weekly. Spleens were harvested on different days postinjection (p.i.) to assess in vitro cytolytic activity. In parallel, tissues from injection sites were stained with macrophage-, CD4-, and CD8-detecting antibodies. Mice were injected s.c. with H-IFNg MCA106 tumor. After 150 days the animals were rechallenged s.c. with MCA106P in one leg and with irrelevant syngeneic tumor in the other. Results: Both P- and L-IFNg cells had similar growth, whereas the H-IFNg cells never grew. Only splenocytes from the H-IFNg animals showed in vitro CTL activity persisting until day 30 p.i. Histological data revealed a macrophage and CD4+ infiltrate much earlier in the H-IFNg group compared with the P group. Only the irrelevant, syngeneic tumor grew in animals previously injected with H-IFNg cells, whereas both P and irrelevant syngeneic tumors grew in controls. Conclusions: Transduction of MCA106 cells with the MuIFNg gene diminished in vivo tumorigenicity in proportion to the amount of IFNg secreted. Immunization with H-IFNg cells elicited a host response characterized by macrophages and CD4+ cells. Long-term tumor-specific memory was seen after immunization with H-IFNg cells.

    Notes: Abstract The responses of Saccharomyces cerevisiae towards the oxyanions tellurite, selenite and chromate were investigated in order to establish the involvement of the yeast vacuole in their detoxification. Three mutants of S. cerevisiae with defective vacuolar morphology and function were used; mutant JSR180D1 is devoid of any vacuolar-like structure while ScVatB and ScVatC are deficient in specific protein subunits of the vacuolar (V)-H -ATPase. All the mutant strains showed increased sensitivity to tellurite and chromate compared to their parental strains. Such sensitivity of the mutants was associated with increased accumu-lation of tellurium and chromium. These results indicate that accumulation of both tellurium and chromium occurred mainly in the cytosolic compartment of the cell, with detoxification influenced by the presence of a functionally-active vacuole which may play a role in compartmentation as well as regulation of the cytostolic compartment for optimal expression of a detoxification mechanism, e.g. reduction. In contrast, the vacuolar-lacking mutant, JSR180D1, and the defective V-H ATPase mutant ScVatB displayed lower selenium accu-mulation than their parental strains. Additionally, the mutant strain ScVatB displayed a higher tolerance to selenite than the parental strain. This result suggests that accumulation of selenium occurs mainly in the vacuolar compartment of the cell with tolerance depending on the ability of the cytosolic component to reduce selenite to elemental selenium, which might, in turn, be related to activity of the V-H -ATPase. These results are discussed in relation to vacuolar compartmentation and the significance of the vacuolar H -ATPase in cytosolic homeostasis of H both of which may affect the accumulation, reduction, and toler-ance to the tested metal(loids). © Rapid Science 1998

    Description / Table of Contents: Zusammenfassung.  Die Dissoziationskonstanten von 3-Phenylsulfonamidorhodanin (PSR) in 0.1 M KCl und 60% (v/v) Ethanol-Wasser wurden potentiometrisch ermittelt. Die stufenweisen Stabilitätskonstanten seiner Komplexe mit Mn2+, Co2+ Ni2+, Zn2+, La3+, Ce3+, Gd3+, UO2+ und Th4+ wurden bestimmt. Die Stabilitätsreihenfolge lautet Th4+ 〉 UO2+ 〉 Gd3+ 〉 La3+ 〉 Mn2+ 〈 Co2+〈 Ni2+ 〈 Cu2+ 〉 Zn2+. Die thermodynamischen Parameter von PSR und seinen Komplexen werden diskutiert. Der Dissoziationsprozeß verläuft nicht-spontan, endotherm und entropisch ungünstig. Die Komplexbildung ist spontan, exo- oder endotherm je nach Metall und entropisch begünstigt. Die Zusammensetzung der Komplexe wurde spektrophotometrisch bestimmt; es entstehen 1:1- und 1:2-Komplexe (Metall:Ligand)..
    Notes: Summary.  The dissociation constants of 3-phenylsulfonamidorhodanine (PSR) were determined potentiometrically in 0.1 M KCl and 20% (v/v) ethanol-water. The stepwise stability constants of the complexes of Mn2+, Co2+, Ni2+, Cu2+, Zn2+,La3+, Ce3+, Gd3+, UO2+ 2, and Th4+ with PSR were determined. The stabilities of the complexes were found as follows: Th4+ 〉 UO2+ 2 〉 Gd3+ 〉 Ce3+ 〉 La3+ 〉 Mn2+〈 Co2+ 〈 Ni2+ 〈 Cu2+ 〉 Zn2+. The thermodynamic parameters for PSR and its complexes were evaluated and discussed. The dissociation process is non-spontaneous, endothermic, and entropically unfavourable. The formation of the complexes was found to be spontaneous, exothermic or endothermic (depending on the metal), and entropically favourable. The stoichiometries of the complexes were determined spectrophotometrically and indicate the formation of 1:1 and 1:2 (metal:ligand) complexes..

    Notes: Abstract The purpose of this study was to evaluate the protective effect of a new endotoxin analogue, monophosphoryl lipid A (MLA) in a rabbit model of myocardial ischemia/reperfusion and to show if this protection was mediated via synthesis of 70 kDa heat shock protein (HSP 70). Three groups of New Zealand White rabbits underwent 30 min coronary occlusion, followed by 4 hours reperfusion. First group of rabbits (n = 6) were treated with 0.35 ml vehicle (40 % propylene glycol, 10 % ethanol in water). The second and third group of rabbits (n = 6–8) were treated with MLA (35 μg/kg, i.v.) 12 and 24 hours prior to ischemia and reperfusion. MLA treatment either 12 or 24 h prior to ischemia/reperfusion demonstrated significantly reduced infarct size (12.5 ± 1.7 and 14.7 ± 2.1% for 12 and 24 h) when compared with vehicle control (40.4 ± 8.6%, mean ± S.E.M, p 〈 0.05). No significant differences in the infarct size was observed between the 12 and 24 h MLA treated groups. The area at risk was not significantly different between the three groups. Baseline values of heart rate, systolic and diastolic blood pressure were not significantly different between the control and MLA treated groups. However, the systolic as well as diastolic blood pressure during reperfusion were significantly lower in rabbits treated with MLA. Western blot analysis of the protein extracts of the hearts (n = 2/group) demonstrated no increase in the expression of the inducible form of HSP 70 following treatment with MLA. We conclude that MLA has significant anti-infarct effect in rabbit which is not mediated by the cardioprotective protein HSP 70. The anti-infarct effect of this drug is superior to the reported protective effects of delayed ischemic or heat stress preconditioning. We hypothesize that the pharmacologic preconditioning afforded by MLA is accomplished via a unique pathway that bypasses the usual intracellular signaling pathways which lead to the myocardial protection with the expression of heat shock proteins.

    Notes: Abstract Patients with an augmented or diverted urinary system are considered higher risk recipients in view of increased technical problems and infective complications. We studied the long-term renal graft function and survival in patients with a pretransplant ileal conduit or ileal/caecocystoplasty. Between 1986 and 1997, 14 of 1253 (1.1 %) renal transplant recipients had their transplant ureters anastomosed into an abnormal urinary tract. These consisted of ten ileal conduits and four ileal/caecocystoplasties. Median follow up was 42 months (range 1–156). All ten ileal conduits were discharged with a functioning graft. There was one graft loss chronic rejection and one cardiac death. The median creatinine level was 130 mmol/l and 50 % have a urinary infection. All the patients with an ileal/caecocystoplasty were discharged with a functioning graft and these are still functioning; median creatinine of 132 mmol/l and 75 % have a urinary infection. One- and 3-year graft survival was 93 % and 86 %. We conclude that the long-term outcome of renal transplantation in carefully assessed patients with an abnormal urinary tract is satisfactory despite a high incidence of urinary infection.
