    Notes: Abstract  In macaque monkeys, aspiration but not excitotoxic lesions of the medial temporal lobe limbic structures, the amygdala and hippocampus, produce a severe impairment in visual recognition memory. Furthermore, certain ventromedial cortical regions, namely the rhinal (i.e., entorhinal and perirhinal) cortex, are now known to be critical for visual recognition memory. Because the route taken by temporal cortical efferent fibers, especially perirhinal efferents, passes nearby the amygdala, it is possible that inadvertent damage to these fibers is produced by the aspirative but not the excitotoxic process, thereby accounting at least in part for the different behavioral outcomes of the two types of lesion. To test this idea, we assessed the integrity of the rhinal corticothalamic projection system after aspiration lesions of the amygdala. Three rhesus monkeys with unilateral amygdala removals received bilaterally symmetrical injections of a retrograde fluorescent tracer into the medial portion of the mediodorsal nucleus of the thalamus. Retrogradely labeled cells were identified using conventional fluorescence microscopy techniques. In all three cases, the rhinal cortex of the intact hemispheres contained moderate numbers of retrogradely labeled cells. By contrast, the rhinal cortex of the amygdalectomized hemispheres consistently contained few retrogradely labeled cells, and a direct comparison of the two hemispheres showed this difference to be statistically significant. A similar asymmetric pattern was observed for area TE but not for the cortex lining the dorsal bank of the superior temporal sulcus, nor for the rostral cingulate motor area, which was examined as a control. The results indicate that aspiration lesions of the amygdala not only remove the cell bodies of the amygdala, as intended, but also inadvertently disrupt projection fibers arising from cells in the rhinal cortex and area TE that pass nearby or through the amygdala en route to the thalamus. Behavioral studies examining the effects of aspiration lesions of the amygdala in nonhuman primates need to take these findings into consideration.

    Notes: Laboratory trials have demonstrated the efficacy of nicotine replacement in smoking cessation bur absolute success races are low. For many, nicotine gum is hard to use and transdermal nicotine is slow-acting and passive. A new, faster-acting nicotine nasal spray (NNS) can provide easily self-administered relief from cigarette withdrawal. The NNS was tested for safely and efficacy in smoking cessation. Two hundred and fifty-five smokers were randomized to NNS or a piperine placebo. Drug use was limited to 8–32 doses/day for 6 months. Subjects were tested while smoking and at post-cessation daily (week 1) with follow-up at weeks 2, 3, 6 and at 3 months, 6 months and 1 year. Continuous abstinence analyses (CO ≤8 ppm.; no slips) showed that NNS significantly enhanced success rates over placebo overall (p 〈 0.001) and at all test intervals. Differences at key intervals between active and placebo were: 63% vs. 40% (day 5), 51% vs. 30% (week 3), 43% vs. 20% (6 weeks), 34% vs. 13% (3 months), 25% vs. 10% (6 months) and 18% vs. 8% (1 year). Side effects were common but tolerable. Cotinine measures showed that replacement of nicotine approximated 30% of smoking levels. Hazard functions revealed relapse risks peaked at day 1, day 5 and 3 weeks for strict abstinence. It is concluded NNS is safe, efficacious and a viable alternative treatment for smoking cessation.

    Notes: The significance of changes in cytokinin content during early fruit growth was examined in the kiwifruit (Actinidia deliciosa var. deliciosa cv. Hayward). Fruit growth was modified by the reduction of seed number or by the application of the synthetic phenylurea cytokinin N-(2-chloro-4-pyridyl)-N-phenylurea (CPPU). The influence of these treatments on cell division was monitored by flow cytometry and changes in the endogenous cytokinins were measured at days 10 and 20 after anthesis, using high-performance liquid chromatography and radioimmunoassay. Total cytokinin levels appeared not to be limiting growth since the highest total cytokinin concentration was detected in unpollinated fruit, which abscised by day 25 after anthesis. However, compared with control fruit which had the highest concentration of zeatin (Z) 10 days post anthesis, Z levels were low in unpollinated fruit. It is hypothesised that an increase in Z is the critical change in cytokinin metabolism required for the initiation of cell division and fruit growth. The synthetic cytokinin CPPU promoted fruit development, but there was a decrease in the endogenous cytokinin concentration. Zeatin was not detected in CPPU-treated fruit. Cell division was reduced in unpollinated fruitlets but there was no significant difference (P 〉 0.05) between the other treatments. Differences in final fruit size appeared to be due to cell expansion.

    Notes: Abstract In a group of heavy smokers, overnight abstinence from smoking facilitated the perception of briefly presented smoking words. Subjects in the nicotineabstinent condition accurately identified significantly more smoking-related words than food-related or neutral words. However, a group tested in a non-abstinent condition showed no significant differences in ability to identify the three different word types. Smokers deprived of cigarettes were also significantly better able to categorize smoking words than non-abstinent subjects. These results demonstrate an abstinence-based facilitation of processing smoking-related stimuli at the semantic level, consistent with the hypothesis that smoking-related concepts are activated, or primed, during deprivation from nicotine.

    Notes: Abstract We previously observed a marked increase ingastrointestinal toxicity of rac -flurbiprofen comparedto the therapeutically equivalent dose of the Senantiomer. This paper quantitates these observations and examines the mechanism by which thisparadoxical toxicity occurs. We have evaluated the ulcerscores, mucosal neutrophil infiltration, byimmunostaining of CD11/18 antigen, and mucosalneutrophil activity by myeloperoxidase measurement at two doselevels of (R)-, (S)-, and rac-flurbiprofen, administeredover 30 days. Dose-response for intestinal ulcerproduction was observed for rac- and (S)-flurbiprofen; animals given (R)flurbiprofen exhibited noulcers. Yet rac-flurbiprofen proved to be twice asulcerogenic as (S)-flurbiprofen. The mechanism of theexacerbation of gastrointestinal toxicity of(S)flurbiprofen by the noncyclooxygenase inhibiting(R)-flurbiprofen is believed to be associated with itseffect on ICAM-1 up-regulation. This is followed byneutrophil adhesiveness to ICAM-1 via the LFA-1 antigenon its surface and the extravasation ofneutrophils into the tissue. We also examined the effectof high dose (R)-flurbiprofen vs vehicle over 15 days inanimals in which ulcers had been produced by treatment with (S)-flurbiprofen for the previous 15 days.(R)-flurbiprofen did not sustain induced ulcers. Theresults of this study suggest that human studies beconducted to determine if enhanced gastrointestinal toxicity occurs in man. This is at issue sincerac compounds of this class are available over thecounter and others may be introduced.

    Notes: Abstract Sexual harassment has been a difficult area of legal interpretation. Legal frameworks used in other areas of Title VII of the Civil Rights Act have not been used in the development of standards by which to judge on the job harassment. Specific difficulties analyzed are the status of group versus individual protection, the notion that harassment is a correctable violation, problems of defining a hostile environment and determining harm, adverse effects on third parties, harassment without sexuality, and same sex harassment. Disagreements between the Circuit Courts and the EEOC are highlighted and effects on individuals and organizations are discussed. Suggestions for organization policy are made.
