    Notes: Summary Leptin is an adipocyte-derived blood-borne satiety factor that acts on its cognate leptin receptor (Ob-R) in the hypothalamus, thereby regulating food intake and energy expenditure. To explore whether mutations in the Ob-R gene cause obesity in humans, we have searched for mutations in the gene for Ob-Rb, a biologically active receptor isoform, in obese Japanese subjects. We have also examined associations between such mutants and obesity in the Japanese. Genomic DNAs were used as templates in polymerase chain reaction (PCR) with primers selected to amplify exons 2 to 20 of the human Ob-Rb gene. Direct sequence analysis of the PCR products revealed 7 nucleotide sequence variants (Lys109Arg, Gln223Arg, Ser343Ser, Ser492Thr, Lys656Asn, Ala976Asp, and Pro1019Pro) in the Ob-Rb coding region from 17 obese Japanese subjects with a family history of obesity (BMI 39.3 ± 8.4 kg/m2). No missense and nonsense mutations were found such as those in Zucker fatty (fa/fa) rats and Koletsky (fa k /fa k) rats. Nucleotide substitutions occurred at relatively high frequencies at codons 109, 223, 976, and 1019 (79, 91, 100, and 85 %, respectively). Allele frequency of each variant determined by PCR-RFLP and PCR-single strand conformation polymorphism analyses showed no significant differences between 47 obese (BMI 35.1 ± 6.5 kg/m2) and 68 non-obese (BMI 21.6 ± 2.2 kg/m2) subjects. The present study represents the first report of sequence variants of the Ob-Rb gene in the Japanese and provides evidence against either obesity-causing mutations or association of sequence variants with obesity in obese Japanese subjects.     Notes: Abstract Aims/hypothesis. Previous studies show that inflammatory cytokines play a part in the development of insulin resistance. Thiazolidinediones were developed as insulin-sensitizing drugs and are ligands for the peroxisome proliferator-activated receptorγ (PPARγ). We hypothesized that the anti-diabetic mechanism of thiazolidinediones depends on the quantity of PPARγ in the insulin resistant state in which inflammatory cytokines play a part. Methods. We isolated rat PPARγ1 and γ2 cDNAs and examined effects of various cytokines and thiazolidinediones on PPARγ mRNA expression in rat mature adipocytes. Results. Various inflammatory cytokines, such as tumour necrosis factor-α (TNF-α), interleukin-1α (IL-1α), IL-1β, IL-6 and leukaemia inhibitory factor decreased PPARγ mRNA expression. In addition, hydrogen peroxide, lysophosphatidylcholine or phorbol 12-myristate 13-acetate also decreased the expression of PPARγ. The suppression of PPARγ mRNA expression caused by 10 nmol/l of TNF-α was reversed 60 % and 55 % by treatment with 10–4 mol/l of troglitazone and 10–4 mol/l of pioglitazone, respectively. The suppression of glucose transporter 4 mRNA expression caused by TNF-α was also reversed by thiazolidinediones. Associated with the change of PPARγ mRNA expression, troglitazone improved glucose uptake suppressed by TNF-α. Conclusion/interpretation. Our study suggests that inflammatory cytokines could be factors that regulate PPARγ expression for possible modulation of insulin resistance. In addition, we speculate that the regulation of PPARγ mRNA expression may contribute to the anti-diabetic mechanism of thiazolidinediones. [Diabetologia (1999) 42: 702–710]

    Notes: Abstract  We cloned the 5’ upstream region of IL-15 genomic DNA and examined promoter activity in macrophages stimulated with lipopolysaccharide(LPS). The 1.2 kilobase (kb) fragment of the 5’ upstream region contained binding elements for LPS-inducible transcription factors such as NFIL-6 or NF-κB. Determined by luciferase assay following transient transfection in the J774A.1 macrophage cell line, the 1.2 kb of the 5’ upstream region exhibited high promoter activity in response to LPS, while promoter activity was significantly reduced by the 5’ deletion of 313 base pairs containing the NF-κB binding motif. Nuclear protein prepared from LPS-stimulated macrophages formed a complex with the NF-κB binding sequence of the IL-15 promoter. Taken together, the binding of nuclear protein to the NF-κB binding site is required for transcriptional activation of the IL-15 gene in LPS-stimulated macrophages.

    Notes: Summary We report on the biexciton formation of the pseudodirect exciton consisting of the AlAs-X z electron and the GaAs-Γ heavy hole in (GaAs) m /(AlAs) m type-II superlattices withm=10, 12, and 13 monolayers. The photoluminescence lineshape of the pseudodirect exciton exhibits a doublet feature having an energy separation of ∼3 meV at the excitation power of the order of mW/cm2 in all the samples, and the low-energy band grows superlinearly. The transient profiles of the doublet photoluminescence band early indicate the biexciton formation: the delay of the rise of the biexciton photoluminescence on the low-energy side and its shorter decay time in comparison with the exciton photoluminescence.

    Notes: Both theoretical and experimental electron-impact total ionization cross sections of CF4, C2F6, and C3F8 are presented. The experimental cross sections have been measured as a function of incident electron energy T from threshold to 3 keV. A parallel plate condenser type apparatus was used. The molecular polarizability for C3F8 was empirically estimated to be α=10.6 Å3±0.8 Å3. Theoretical cross sections calculated from the binary-encounter-Bethe (BEB) method, which combines a modified form of the Mott cross section and the Bethe cross section, are compared with the experimental cross sections. The BEB cross sections calculated from correlated molecular wave functions with theoretical estimates for multiple ionization are about 10% higher than the experimental data at the peak for CF4, while they are in excellent agreement with the experimental data for C2F6 and C3F8. Our analysis shows that the BEB theory implicitly includes part of neutral dissociation, such as CF4→CF3+F, and hence tends to be an upper limit to the total ionization cross section. We found that the difference between our best theory for CF4 and the present experimental cross section exhibits a remarkable similarity to the shape of the recently measured cross section for neutral dissociation, though there is no a priori reason for the similarity. Owing to the large number of bound electrons, the correlation included in our wave functions for C2F6 and C3F8 is more limited than for CF4. Hence, we believe that for these two molecules the calculated cross sections are lower than the true BEB values, in spite of the apparent excellent agreement between the theory and the experiment. © 1999 American Institute of Physics.

    Notes: Hydrodynamic instabilities, such as the Rayleigh–Taylor (R–T) instability, play a critical role in inertial confinement fusion as they finally cause fuel-pusher mixing that potentially quenches thermonuclear ignition. Good understanding of the instabilities is necessary to limit the mixing within a tolerable level. A series of experiments has been conducted on the GEKKO XII laser facility [C. Yamanaka et al., IEEE J. Quantum Electron. QE-17, 1639 (1981)] to measure hydrodynamic instabilities in planar foils directly irradiated by 0.53 μm laser light. It has been found that (1) the imprint is reasonably explained by an imprint model based on the equation of motion with the pressure perturbation smoothed by the cloudy-day effect, and (2) the experimental R–T growth rate is significantly reduced from the classical growth rate due probably to ablative stabilization enhanced by nonlocal heat transport. © 1997 American Institute of Physics.
