    Notes: Summary In spontaneously diabetic GK rats, insulin secretion from pancreatic beta cells in response to glucose is selectively impaired, probably due to deficient intracellular metabolism of glucose and impaired closure of KATP channels during glucose stimulation. By using electrically permeabilized islets of GK rats, we explored the functional modulations in exocytotic steps distal to the rise in [Ca2 + ]i in the diabetic condition. At 30 nmol/l Ca2 + (basal conditions) insulin release was similar between GK and non-diabetic control Wistar rats. In response to 3.0 μmol/l Ca2 + (maximum stimulatory conditions), insulin release was significantly augmented in permeabilized GK islets (p 〈 0.01). Raising glucose concentrations from 2.8 to 16.7 mmol/l further augmented insulin release induced by 3.0 μmol/l Ca2 + from permeabilized control islets(p 〈 0.001), but had no effect on that from permeabilized GK islets. The stimulatory effect of glucose on insulin release from permeabilized control islets was partly inhibited by 2,4-dinitrophenol, an inhibitor of mitochondrial oxidative phosphorylation (p 〈 0.01). The hyperresponse to Ca2 + in GK islets may play a physiologically compensatory role on the putative functional impairment both in [Ca2 + ]i rise and energy state in response to glucose in diabetic β cells, and may explain the relative preservation of insulin release induced by non-glucose depolarizing stimuli, such as arginine, from pancreatic islets in non-insulin-dependent diabetes mellitus.     Notes: Abstract Objective: Dynamic intrinsic PEEP (PEEPi-dyn) is the airway pressure required to overcome expiratory flow and is considered to represent the lowest regional PEEPi. However, there are few data to validate this assumption. We investigated if PEEPi-dyn represents the lowest PEEPi. Setting: The animal laboratory at the Osaka University Medical School. Measurements and results: We compared static PEEPi (PEEPi-stat) and PEEPi-dyn in healthy animals. Five adult white rabbits (2.77 ± 0.05 kg) were anesthetized, tracheostomized, and intubated with several different sizes of endotracheal tubes (ETT) (2.0, 2.5, 3.0, 3.5, or 4.0 mm i. d.). The animals were paralyzed and ventilated (Siemens Servo 900C). Baseline ventilator settings were at a rate of 50/min, inspiratory:expiratory (I:E) ratio of 2:1 or 4:1, and minute ventilation was manipulated to create 3 or 5 cm H2O PEEPi-stat. PEEPi-stat was measured using the expiratory hold button of the ventilator. PEEPi-dyn showed large variations. In all ventilator settings, PEEPi-dyn was higher than PEEPi-stat (p 〈 0.001). The larger the ETT, the higher the PEEPi-dyn at an I:E ratio of 2:1 (p 〈 0.05). The higher the minute ventilation, the greater the difference between PEEPi-stat and PEEPi-dyn. The tidal volume and the difference showed a significant correlation (r 2 = 0.514, p 〈 0.001). Conclusions: The value of PEEPi-dyn was dependent on ventilatory settings, and PEEPi-dyn does not necessarily represent the lowest regional PEEPi within the lungs.

    Notes: 1. ATP-sensitive K+ channels (KATP) are activated either by decreased intracellular ATP content or ATP/ADP ratio during ischaemia. We examined the role of a cerebral KATP in arterial pressure regulation during acute cerebral ischaemia using SHR and WKY rats. Thirteen week old male SHR or WKY rats were anaesthetized with urethane, and arterial pressure and heart rate were recorded under an artificial ventilation.2. Intracerebroventricular (i.c.v.) injections of glibenclamide, a specific inhibitor of KATP, elicited dose-dependent vasopressor responses in WKY with bilateral ligation of carotid arteries, whereas it caused smaller vasopressor responses in SHR than WKY.3. Systemic administration of AVP V1 receptor antagonist, OPC-21268, abolished the vasopressor responses of i.c.v. injections of glibenclamide in WKY but not in SHR.4. Intracerebroventricular injections of glibenclamide caused both the increase in plasma concentration of AVP and the decrease in pituitary AVP content in WKY with bilateral ligation of carotid arteries, whereas it elicited no significant change in plasma and pituitary concentration of AVP in SHR with bilateral ligation of carotid arteries.5. Cerebral KATP may play a role in the protection of excess hypertension by inhibiting AVP release from the pituitary glands during acute ischaemia in WKY, but this mechanism might not work in SHR during acute cerebral ischaemia.

    Notes: Fine powders of Sm2(Fe0.9Co0.1)17Nx (x=∼3) with particle sizes (d) around 1 μm as coated with zinc metal produced via the photodecomposition of diethylzinc [Zn(C2H5)2], which still provided high remanence (Br) and coercivity (Hcj) values of ∼1.43 T and ∼0.85 MAm−1, were molded to compression-type resin bonded Zn/Sm2(Fe0.9Co0.1)17Nx magnets with density values of ∼6.33 g cm−3. By optimizing the preparation conditions such as grinding, surface coating, and molding for them, the highest maximum energy product of (BH)max=186 kJm−3 for Hcj=0.73 MAm−1 was recorded among all kinds of the Sm–Fe–N based magnets reported to date. Furthermore, the excellent aging behavior of the bonded Zn/Sm2(Fe0.9Co0.1)17Nx magnets was observed after standing in air at ∼393 K and the thermal coefficient for permanent magnet was evaluated to be α(Br)=−0.04% K−1. © 1999 American Institute of Physics.

    Notes: Abstract. The BAT2 gene lies within the class III region of the major histocompatibility complex. We investigated the frequency of the BAT2 microsatellite alleles (BAT2) in 74 young-onset insulin-dependent diabetes mellitus (IDDM) patients, 51 adult-onset IDDM patients, and 85 normal control subjects, and assessed the associations among these BAT2 alleles, TNFa microsatellite alleles (TNFa), and HLA-DRB1 alleles. The frequency of the BAT2.9 allele was significantly increased in the young-onset IDDM patients (12.8 vs 4.1%, Pc=0.04896), whereas the frequency of BAT2.12 allele was significantly decreased in young-onset IDDM patients (0.0 vs 11.8%, Pc=0.00002) compared with control subjects. The BAT2.9 allele was strongly associated with TNFa9 in the young-onset IDDM patients, although no association was found between the BAT2.9 and HLA-DRB1 alleles. The BAT2.12 allele was strongly associated with TNFa13, and with DRB1*1502 in control subjects. These results suggest that the BAT2 microsatellite polymorphism is associated with the age-at-onset of IDDM and possibly with the inflammatory process of pancreatic β-cell destruction during the development of IDDM. However, this association is not independent of TNFa polymorphisms.

    Notes: Abstract The subcellular localization and developmental changes of aconitase (EC 4.2.1.3) isoforms were studied in pumpkin tissues by anion-exchange chromatography. In etiolated pumpkin cotyledons three aconitase isoforms (denoted Aco I, II, and III, respectively) were found, and the first to be eluted (Aco I) represented the major part of the total aconitase activity in the tissue. Aconitase I gradually disappeared during the greening of cotyledons. In hypocotyls, roots and mature leaves Aco I was barely detectable, though the two other isoforms were present. In pumpkin mitochondria purified by differential or density gradient centrifugation, a high amount of Aco II was present, whereas the other isoforms were detected in low quantities. Since aconitase has never been clearly detected in glyoxysomal fractions, we conclude that the first and the third aconitase isoforms are cytosolic, while the second one represents the mitochondrial aconitase. Moreover, the first aconitase isoform is present in appreciable amounts only in etiolated cotyledons, so that it should represent the aconitase isoform involved in the glyoxylate cycle.

    Notes: Abstract We monitored the serial changes of natural killer cell (NK) activity in eight recipients of living-related liver transplantation. The HLA types of all eight patients were haplotypically identical with those of their donors. Tacrolimus and methylprednisolone were used for immunosuppression. The NK activity before transplantation was 24.1 ± 20.2 % which is surprisingly low when compared with the value for normal individuals (67.7 ± 13.2 %, P 〈 0.01) or a liver dysfunction group (49.4 ± 21.9 %, P 〈 0.05). Serial changes in NK activity revealed a minimum of 6.1 ± 3.6 % 1 week after transplantation, gradually increasing to 49.2 ± 12.5 % at 2 months after transplantation. These results suggest that the diseased liver might play an important role in the suppression of NK activity.

    Notes: Abstract • Background: Menkes' disease may be due to a lack or deficiency of copper in various organs. The macular mouse is known as a model for Menkes' disease. We examined melanin granules in the retinal pigment epithelium and the activity of cytochrome oxidase, a copper-containing enzyme, in the retinas of macular mice by electron microscopy. • Methods: In the retinas of hemizygote macular mice we demonstrated cytochemically (oxidative polymerization of diaminobenzidine to an osmophilic reaction product) the activity of cytochrome oxidase. The distribution of melanin granules in the retinal pigment epithelium related to the activity of another copper-containing enzyme, tyrosinase, was also studied. Stereological methods were applied to obtain quantitative data. • Results: In the retinal photoreceptor inner segments of the macular mouse, the mitochondria were more numerous than in normal littermates and they appeared swollen. There were fewer melanin granules in the retinal pigment epithelium of macular mice than in that of normal littermates. The cytochrome oxidase activity was significantly lower in the macular mice than in the controls. • Conclusion: Macular mice have lower activity of cytochrome oxidase and fewer melanin granules than do normal mice. Both changes may be related to copper deficiency. These results correspond to the retinal changes seen in patients with Menkes' disease.
