    Notes: Abstract: We synthesized a potent and selective antagonist radioligand for the neurokinin (NK)-1 receptor and characterized its binding to guinea pig striatal membranes. (R) - N - [2 - [Acetyl[3H3][(2 - methoxyphenyl) - methyl]amino] - 1 - (1H - indol - 3 - ylmethyl)ethyl][1,4′ - bipiperidine]-1′-acetamide ([3H]LY303870) binds to a single class of sites with an equilibrium KD of 0.22 nM and a Bmax of 723 fmol/mg of protein. Unlabeled LY303870 potently inhibited the binding with an IC50 of 0.56 nM, whereas the less active (S)-enantiomer (LY306155) was substantially less potent. The nonpeptide NK-1 antagonists (±)-CP96,345 and (±)-RP 67580 had IC50 values of 0.74 and 49 nM, respectively. Substance P (SP) was also a potent inhibitor with with an IC50 of 3.1 nM. The inhibition by SP could be separated into two components: a high-affinity component with a Ki of 0.53 nM and a lower-affinity component with a Ki of 155 nM. Addition of 100 µM guanylyl 5′-imidodiphosphate [Gpp(NH)p] in the incubation increased the relative amount of the low-affinity agonist state of the receptor. Consistent with the antagonist properties of LY303870, the dissociation rate of [3H]LY303870 was not changed by the presence of 100 µM Gpp(NH)p. The distribution of [3H]LY303870 binding sites in the guinea pig brain closely matched the distribution of NK-1 receptors labeled by [3H]SP. Therefore, [3H]LY303870 is a potent and selective antagonist radioligand for NK-1 receptors in guinea pig brain. In addition, regulation of NK-1 agonist affinity by guanine nucleotides is similar to that seen for monoaminergic receptors.

    Notes: Abstract: (R)-[3H]Tomoxetine is a radioligand that binds to the norepinephrine (NE) uptake site with high affinity but also binds to a second, lower-affinity site. The goal of the present study was to identify the nature of this low-affinity site by comparing the binding properties of (R)-[3H]tomoxetine with those of (R/S)-[3H]nisoxetine, a highly selective ligand for the NE uptake site. In homogenate binding studies, both radioligands bound to the NE uptake site with high affinity, whereas (R)-[3H]tomoxetine also bound to a second, lower-affinity site. The autoradiographic distribution of binding sites for both radioligands is consistent with the known distribution of NE-containing neurons. However, low levels of (R)-[3H]-tomoxetine binding were seen in the caudate-putamen, globus pallidus, olfactory tubercle, and zona reticulata of the substantia nigra, where (R/S)-[3H]nisoxetine binding was almost absent. In homogenates of the caudate-putamen, the NE uptake inhibitors desipramine and (R)-nisoxetine and the serotonin (5-HT) uptake inhibitor citalopram produced biphasic displacement curves. Autoradiographic studies using 10 nM (R)-nisoxetine to mask the binding of (R)-[3H]tomoxetine to the NE uptake site produced autoradiograms that were similar to those produced by [3H]citalopram. Therefore, (R)-[3H]tomoxetine binds to the NE uptake site with high affinity and the 5-HT uptake site with somewhat lower affinity.

    Notes: As a tool for screening large numbers of biological samples by means of amplification (e.g., Qβ or PCR) we have constructed a thermocycler that includes optionally a 96-channel or 960-channel glass fiber fluorimeter (combined with a cooled charge-coupled-device camera). We briefly describe the system integration of all components like liquid handling, thermostats, an x,y,z robot arm, and the glass fiber fluorimeter. The integrated glass fiber fluorimeter allows sensitive on-line measurements in 960 channels within 5 s. Two different screening procedures were carried out. In a first experiment PCR reactions were done in the presence of the known PCR inhibitor hematin and its suppressor transferrin. The system was used to titrate the suppressor with the inhibitor hematin in order to determine the maximum inhibitor concentration tolerated at a given suppressor concentration. We processed 96 PCR samples in parallel with 11 different concentration steps. In a second experiment the 960-channel glass fiber fluorimeter was used to monitor on line the amplification of the Qβ system in the presence or absence of an inhibitor (heparin). Since the doubling time of Qβ RNA variants is about 20 s, on-line detection is crucial for the experimental setup. The evolution of new RNA species adapted to high inhibitor concentrations could be proved by comparison of the fluorimetric signal and electrophoresis. © 1997 American Institute of Physics.

    Notes: Mean velocity and Reynolds-stress profiles were measured in the incompressible turbulent boundary layer with zero pressure gradient on the aerodynamically smooth sidewall of the German–Dutch windtunnel. Data were taken at Reynolds numbers Reδ2, based on momentum thickness δ2 of 2×104, 4×104, and 6×104 by means of four different types of hot-wire probes (three-wire probes, X wire, and normal-wire probes). There are also measurements of skin friction and of spectra. The data compare well with the few available other measurements, and all profiles show independence of Reynolds number in the outer region of the boundary layer when plotted against y/Δ, where Δ is the Rotta–Clauser length. © 1995 American Institute of Physics.

    Notes: The compound Ba3Ca1.18Nb1.82O9-delta (BCN18) has been discussed recently as a promising high-temperature proton conductor. Oxygen loss under very reducing conditions has been determined quantitatively using thermogravimetry. The thermodynamic parameters DeltaH°O= 2.58 ± 0.06 eV and DeltaS°O= 1.06 × 10-3 eV/K provide a suitable thermodynamic description for the equation O×O right arrow ½O2+V··O+ 2e´. Enthalpy and entropy of the hydrogen absorption reaction have been determined indirectly for BCN18: DeltaH°H= -0.8 eV and DeltaS°H= -6.6 × 10-4 eV/K. Hydrogen absorption is, therefore, an exothermic process. Valence changes where Nb5++e´ right arrow Nb4+ also have been taken into account under reducing conditions.

    Notes: Dimensional changes of the high-temperature proton conductor Ba3Ca1.18Nb1.82O9−δ, when exposed to D2O vapor, were investigated using length-change and precision density measurements. Such information is essential for possible applications of proton conductors in solid oxide fuel cells and humidity and hydrogen sensors. A linear increase of the sample lengths with increases in the deuterium content was observed. Comparison of the present D2O data with those that were previously obtained for H2O showed that there was a small isotope effect in the lattice expansion. The fact that the length-change-versus-hydrogen-isotope-concentration curves were almost isotope independent supported the validity of the take-up reaction H2O(g) + VOö+ OxO is an ⇄ 2OHO. In regard to the latter equation, it was found that small islands of silver, palladium, nickel, and platinum act as catalytic promoters of the reaction and lead, for given heavy-water-steam exposure conditions, to an increase of water absorption of up to 100%, when compared with samples without the catalytic surface layers.

    Notes: The water uptake of nominally 5%-Yb-doped strontium zirconate (SrZr0.95Yb0.05O3−α) was studied in situ by thermogravimetry at different temperatures and water vapor pressures. The process could be described in terms of an ideal solution behavior, showing a lower effective vacancy concentration than that expected from the nominal dopant content. The solution reaction was found to have ΔS°=−127 J/(mol·K) and ΔH°=−106 kJ/mol.

    Notes: Background and Objective Measurement of guanine in dust proved a good assessment of mite allergen exposure.Methods Exposure to mite allergens may lead to atopic inflictions. In a semi-natural test system the development of Dermatophagoides pteronyssinus (Trouessart) and Glycyphagus domesticus (De Geer), and the presence of their guanine excretion, was examined in a dustsoiled and mouldy environment. Mites were counted after heat-escape, and guanine was detected by means of capillary zone electrophoresis. For each species, 50 mites randomly taken, were inoculated on soiled test-surfaces of 10 × 10 cm. Rough wooden board, gypsum board, tufted carpet, and a self-made mattress representing wall surfaces and home-textiles, respectively, were used. Eight weeks after inoculation with mites only, the surfaces were all mould ridden, and mite and guanine measurements were taken. The Spearman rank correlation test and the Mann-Whitney U-test were used in statistical analysis. The confidence limit was set at 1%.Results Among the various test-surfaces, no differences were found regarding total mite numbers and amount of guanine present (P 〉 0.0l). For the dust-eating mite D. pteronyssinus, total mite numbers correlated with the amount of guanine present (P= 0.002) on all inoculated surfaces, indicating feeding on the protein-rich dust. For the mould devouring mite G. domesticus, however, no such correlation was found (P= 0.72). Apparently, they mainly consumed fungal carbohydrates during this experiment.Conclusion The allergological relevance of storage mites has been under discussion for the last 25 years. In humid homes, these mites will feed almost exclusively on fungi and may produce allergenic or irritating substances different from those arising on protein-rich laboratory media used in allergen extract production or present in carpets, bedding and furniture.
