    Notes: Summary Mitochondrial DNA is exclusively maternally inherited. We recently found the prevalence of diabetic patients with an A to G transition at position 3243 of leucine tRNA (3243 base pair (bp) mutation) to be nearly 1 % in randomly selected Japanese subjects. Here, we report the higher prevalence of diabetic patients with the 3243 bp mutation in a specific Japanese population of women attending a diabetic pregnancy clinic. Of 102 patients with non-insulin-dependent diabetes mellitus 6 (5.9 %) were positive for the mutation, 1 (8.3 %) of 12 patients with gestational diabetes and 2 (5.9 %) out of 34 borderline diabetic patients. In contrast, none of 64 patients (0 %) with insulin-dependent diabetes mellitus had the 3243 bp mutation. Moreover, there was a difference in the prevalence of spontaneous abortions between patients with and without this mutation (27.3 vs 12.4 %). Among nine probands with the mutation, four had a history of one spontaneous abortion (p = 0.0518) and two had a history of two abortions (p = 0.0479). Two probands had a spontaneous abortion even while under strict diabetic metabolic control. The 3243 bp mutation thus may cause spontaneous abortion during pregnancy.     Notes: Abstract A new system for high-speed and continuous 3D measurement is presented. It is based on the slit-ray projection method. The key component is the image plane constructed by a position-sensitive device (PSD) array that is horizontally un segmented and linear, but vertically numbered. All PSD row elements are attached to their respective analog signal processors. The analog signals are multiplexed and digitized by multiplexers and A/D converters at appropriate sampling intervals and stored in memory. By virtue of this configuration of the image plane, basis data for 3D measurement are acquired during only one scan of the slit ray at high speed in the form of memory addresses and data. Moreover, if we use large capacity and/or dual port memories, we can continuously obtain 3D range data for as many scenes as needed. The trial system has been implemented with a newly designed 128-channel PSD array and has verified our method, enabling us to obtain successive 3D range data at video rates with an error within±0.3%.

    Notes: Abstract Objective: This study was performed to investigate whether it is possible to use saliva instead of blood usually used for therapeutic drug monitoring (TDM) of disopyramide. Methods: Six healthy male volunteers ingested 200 mg of disopyramide base, and the disopyramide concentrations in saliva and plasma (total and unbound) were determined by the HPLC. Results: Disopyramide concentration-time profiles for the saliva were nearly equal to those for the plasma unbound concentrations. A large variation for absorption time of the drug was observed among the subjects. Disopyramide concentrations (Cs) in saliva did not correlated well with plasma total concentrations (Cp), r = 0.799, but did well with unbound concentrations (Cpu), r = 0.969, for the 3–12 h period on the elimination phase. The mean ratio of disopyramide concentrations in the saliva against the plasma unbound concentrations was almost constant (1.02(0.10), CV = 9.7%) for the period. The pharmacokinetic parameters (tmax, t1/2, AUC, AUMC and MRT values) for disopyramide calculated from the saliva data were nearly equal to those from the unbound data. Conclusion: Disopyramide concentrations in saliva correlated well with plasma unbound concentrations on the elimination phase.

    Notes: A new crystal form of PHO4 DNA-binding domain-DNA complex was obtained by the truncation of the N-terminal segment. This diffracts to a higher resolution than any forms reported previously. The present crystal belongs to orthorhombic space group P212121, with unit-cell dimensions a=53.51, b=68.30, c= 108.77 Å. The crystal contains one protein dimer-DNA complex in the asymmetric unit.

    Notes: Nanometer size polarized domains were written in a PbZr1−xTixO3 (PZT) thin film using an atomic force microscope (AFM) and the relationship between the polarized domain and the grain of the film was investigated. The polarized domain was formed by applying a pulse voltage to the ferroelectric PZT thin film through a conductive AFM tip. The polarized domain structure was observed by imaging the piezoelectric-induced surface vibration by an AFM with an ac voltage applied between the tip and the bottom electrode of a sample. The polarized domains with a diameter of 50 nm were written within a single grain. © 1996 American Institute of Physics.

    Notes: Summary In normal human skin, immtmoreactive thrombomodulin (TM) is expressed in a strict differentiation related pattern. solely in suprabasal spinous layer keratinocytes. To evaluate the polential application of TM as a differentiation marker for keratinocyte-derived skin tumours, we have studied immunohistopathological, biochemical and functional TM activities in various skin tumours. Immunoreactive, full sized and enzymatically active TM was expressed in keratinocyte-derived skin tumours (squamous cell carcinoma, seborrhoeic keratosis and partly Bowen's disease), as well as normal epidermal keratinoeytes and endothelial cells. However, no TM was detected in basal cell carcinotnas, senile keratosis or non-squanious epithelial tumours such as malignant melanoma, naevus pigmentosus and Paget's disease. Interestingly, decreased expression was observed in verruca vulgaris. These findings suggested that differentiation-dependent TM expression was restricted to epithelial skin tumours and undetectable on neural crest derived tumours. TM is a differentiation marker for spinous layer keratinocytes and is a useful tool in histopathological study of epithelial tumours.

    Notes: Abstract— The factors affecting the fatigue strength of nitrided titanium were clarified. The fatigue strength depended strongly on the fracture strength of the compound layer formed on the surface by nitriding. We found a Hall-Petch relationship between the fatigue strength of nitrided titanium and the grain size. The findings indicated that the reduction in the fatigue strength by nitriding results from both the formation of the compound layer possessing low fracture strength and grain growth occurring from ordinary nitriding. Furthermore, low-temperature nitriding (620°C, 24 h) was proposed to suppress grain growth. This treatment method improved not only the wear resistance and the corrosion resistance but also the fatigue strength of titanium.

    Notes: Changes in cytosolic proteins of the olfactory system (olfactory epithelium, olfactory nerve and olfactory bulb) and the telencephalon of masu salmon Oncorhynchus masou were analysed by two-dimensional polyacrylamide gel electrophoresis during parr-smolt transformation; parr, pre-smolt and full-smolt stages. In the olfactory system, several protein spots appeared and disappeared in the course of smolting. One protein spot in particular with an estimated molecular weight of 27 kDa and isoelectric point of 5.6 (M27) disappeared in common with the olfactory system during smolting. The disappearance of M27 was also observed in the telencephalon. These proteins, which appeared and disappeared, may reflect the changes in olfactory function during smolting. Simultaneously, the present study confirmed a salmonid olfactory specific protein of 24 kDa (N24), which existed in the olfactory system but not in the telencephalon, as a single neutral protein spot in masu salmon.

    Notes: Summary We investigated whether lysine transport would be subject to adaptive regulation in Caco-2 human intestinal cells. The activity of Lys transport in Caco-2 cells decreased with increasing incubation time with lOmM Lys. Among the two systems involved in Lys transport, the system b0,+ component was greatly decreased by incubating cells ith lOmM Lys, whereas the system y+ component did not change. These results suggest that system b0,+ mainly contributes to the adaptive regulation of Lys transport in Caco-2 cells.

    Notes: Abstract This study was undertaken to investigate whether the level of C-reactive protein (CRP) in the serum of dogs undergoes physiological variation, using 10 normal Beagle dogs (5 males and 5 females), 1–2 years old, maintained in a healthy condition in a controlled environment. The CRP concentration in the sera collected seven times each day at intervals of approximately 3 h ranged from 0.8 to 16.4 µg/ml (mean 5.06±3.60) in one experiment and from 0.8 to 14.0 µg/ml (mean 4.50±2.80) in a second experiment. On examining the 24-h variations in the concentration of CRP in serum, neither consistent changes nor a definite pattern of circadian rhythm was detected. During 28 days observation, only very slight changes, which seemed attributable to analytical errors, were seen in any of the dogs, except one. The concentration of CRP in the serum during the 28 days ranged from 0.8 to 22.6 µg/ml (mean 3.65±1.40). The concentrations underwent no significant variations in individual dogs, but significant differences were found between the dogs (p〈0.01).
