    Notes: The high-repetition switching capabilities of a recently developed symmetric Mach–Zehnder (SMZ) all-optical switch are experimentally investigated. Using a series of four control pulses at intervals of 100 or 25 ps, nearly full switching is achieved at the corresponding intervals. The results experimentally show that the repetition rate, as well as the switching speed, of the SMZ all-optical switch is not restricted by the slow relaxation of the utilized high-efficiency band-filling effect. © 1995 American Institute of Physics.

    Notes: We propose a polarization-discriminating Mach-Zehnder (PDMZ) all-optical switch. The switching speed of this PDMZ all-optical switch is not limited by the slow relaxation time of highly efficient incoherent nonlinearities. We demonstrate a squarelike modulation characteristic, which is necessary in most switching applications, at a switching speed (on-off time) of 40 ps. We also demonstrate ultrafast switching at a detector limited speed of ∼8 ps. © 1995 American Institute of Physics.

    Notes: Polarization of 8×8 vertical cavity surface emitting laser (VCSEL) arrays is completely controlled. These index-guided VCSELs have a 6×5 μm rectangular poststructure consisting of DBR mirrors. All 64 VCSELs emit fundamental single-mode and linearly polarized light with a polarization angle deviation of only 2.9°. Their light output characteristics are almost the same as those of conventional 6×6 μm polarization-uncontrolled VCSELs. © 1995 American Institute of Physics.

    Notes: 1. Effects of anti-Parkinsonian drugs on neurobehavioural changes induced by bilateral lesions of dopaminergic neurons were investigated in rats.2. Dopaminergic neurons in rats were lesioned bilaterally by injection of 6-hydroxydopamine (6-OHDA; 8 μg) into the medial forebrain bundle at the level of the posterolateral hypothalamus. As a result, a decrease in locomotor activity and marked catalepsy and prolongation of grasping time were observed.3. Levodopa, talipexole, bromocriptine and theophylline dose-dependently antagonized the decrease in locomotor activity induced by bilateral 6-OHDA lesions. These drugs also showed antagonistic effects on the appearance of catalepsy and prolongation of grasping time induced by bilateral 6-OHDA lesions. In contrast, trihexyphenidyl showed no antagonizing effect on the neurobehavioural changes induced by 6-OHDA lesions at any concentration tested.4. Combined treatment with levodopa and talipexole antagonized the neurobehavioural changes induced by bilateral 6-OHDA lesions, whereas no marked changes were observed when either drug was administered separately. The same findings were noted with the simultaneous use of either levodopa (2 mg/kg) and theophylline (2 mg/kg) or talipexole (0.005 mg/kg) and theophylline (2 mg/kg).5. These results indicate that this model may be useful for estimating the effects of drugs in the treatment of Parkinson's disease.

    Notes: Time-resolved X-ray diffraction of muscle has demanded ever-increasing flux into small sample volumes with low beam divergence. Results are reported of static and time-resolved small-angle X-ray diffraction studies on muscle fibers using a hard X-ray undulator installed in the Tristan main ring at KEK, Tsukuba, Japan, as an innovative source of synchrotron radiation more intense and better collimated than that available with the Photon Factory bending-magnet beamline. Static studies used the low divergence of the source to obtain detailed high-quality diffraction patterns of stable muscle states. The diffraction patterns from live skeletal muscles showed the numerous (over 100) meridional reflections. The well collimated beam from the undulator made it possible to clearly resolve, with an angular resolution of ca 700 nm, the closely spaced diffraction peaks arising from the two halves of the thick filaments centred on the M lines in a sarcomere, in addition, the diffraction peaks from the thin filaments on opposite sides of the Z bands could be resolved with an angular resolution of ca 1000 nm. The detailed structure of the meridional pattern defines the nature of the molecular packing in the thick and thin filaments. Time-resolved experiments using a focusing mirror aimed to prove cross-bridge states in striated muscle fibers by collecting X-ray diffraction data at a 0.185 ms time resolution from sinusoidally oscillating chemically skinned rabbit muscle fibers during active contraction and in rigor. When sinusoidal length changes at 500 Hz with a peak-to-peak amplitude of 0.6% of the muscle length were applied to a small fiber bundle, the tension showed a simple elastic response during the length oscillation. In the active muscle the intensity of the 14.5 nm myosin-based meridional reflection changed out of phase with the tension change during the oscillating length change. In contrast, in the rigor muscle it occurred in phase with the tension change. The high time-resolved experiments provide an insight into the coupling between conformational changes and force generation of the actomyosin cross-bridges. These studies provide a preview of the expected gains for muscle studies from the more widespread use of undulator radiation at third-generation synchrotron sources.

    Notes: Abstract Chronic granulomatous disease (CGD) is a group of disorders characterized by the failure of phagocytes to produce superoxide. One-third of the cases of CGD in the USA and Europe results from defects in the gene encoding p47 phox , a cytoplasmic component of NADPH oxidase for superoxide generation. In this study, we constructed the bicistronic retrovirus vector Ha-MDR-IRES-p47, which carries cDNAs for a human multi-drug-resistance gene (MDR1) and p47 phox . The amphotropic retroviral producer cells were generated, and the supernatant of the producer cells was used to transduce Epstein-Barr virus-transformed B (EBV-B) cells, established from B cells of p47 phox -deficient CGD patients, as an in vitro model of gene therapy for p47 phox -deficient CGD. The transduced cells expressed both P-glycoprotein and p47 phox protein, and the expression levels were increased after appropriate vincristine selection. The levels of superoxide production in the vincristine-selected cells were increased to a level similar to normal EBV-B cells. This result suggests that it is possible to achieve 100% correction of the CGD defect in p47 phox -deficient EBV-B cells by using the bicistronic vector. This strategy could be employed not only in vitro, but also in vivo, in the gene therapy of a number of inherited diseases.

This paper reports a field study on the effects of soil moisture stress on the rooting habits, transpiration rate, and xylem anatomy of two sorghum cultivars, Tabat (drought susceptible) and Gadambalia (drought tolerant). Two levels of water stress, -0.02 MPa (wet) and -0.75 MPa (dry), were applied. Tabat had a higher root length density (RLD), higher late metaxylem (LMX) vessels per nodal root, higher leaf area, and higher transpiration rate than Gadambalia. In Tabat, soil moisture stress reduced RLD by 30%, nodal roots by 31%, number of LMX vessels in the root by 42%, leaf area by 13%, and transpiration rate by 11%. In Gadambalia soil moisture stress did not affect RLD at depths ≥0.2m, number of nodal roots, or number of LMX vessels per nodal root. However, leaf area and transpiration rate were reduced by 3 and 11%, respectively. Under dry conditions, Gadambalia displayed a higher water extraction efficiency than Tabat throughout the profile (0-0.9 m). In Gadambalia, unlike Tabat, the stem was highly sclerified. A 1- to 3-cell thick layer of schlerenchyma was observed beneath the epidermis. The peripheral vascular bundles were surrounded with a 3- to 6-cell thick schlerenchyma sheath. However, in roots anatomical differences were less prominent. Drought tolerance in Gadambalia is associated with higher water extraction efficiency, fewer nodal roots per plant, fewer LMX vessels per nodal root, a smaller leaf area, and a well developed sclerenchyma.

    Notes: Abstract. A cattle genetic linkage map was constructed which covers more than 95 percent of the bovine genome at medium density. Seven hundred and forty six DNA polymorphisms were genotyped in cattle families which comprise 347 individuals in full sibling pedigrees. Seven hundred and three of the loci are linked to at least one other locus. All linkage groups are assigned to chromosomes, and all are orientated with regards to the centromere. There is little overall difference in the lengths of the bull and cow linkage maps although there are individual differences between maps of chromosomes. One hundred and sixty polymorphisms are in or near genes, and the resultant genome-wide comparative analyses indicate that while there is greater conservation of synteny between cattle and humans compared with mice, the conservation of gene order between cattle and humans is much less than would be expected from the conservation of synteny. This map provides a basis for high-resolution mapping of the bovine genome with physical resources such as Yeast and Bacterial Artificial Chromosomes as well as providing the underpinning for the interpolation of information from the Human Genome Project.
