    Notizen: In this paper it is demonstrated that Raman spectroscopy can be used for in situ online monitoring of semiconductor layer growth. For this purpose an ultra-high-vacuum chamber was designed to suit molecular beam epitaxial growth as well as simultaneous optical measurements utilizing a multichannel detection Raman system. As a result Raman spectra can be taken while growth progresses. As shown by the example of InSb growth, they provide information on the interface chemistry, crystal quality of the growing layer, and the growth mode as well.

    Notizen: Abstract In order to identify DNA variants within the 5′-flanking region of the bovine κ-casein (κCn)-encoding gene, this area of the gene from 13 cows belonging to seven breeds (Holstein Friesian, Brown Swiss, German Simmental, Jersey, Galloway, Scottish Highland and Ceylon Dwarf Zebu) was analysed. For each individual, about 1 kb of the 5′-flanking region including exon I was amplified by polymerase chain reaction (PCR). The biotinylated PCR product was immobilized on magnetic beads followed by direct bidirectional sequencing using an automated DNA sequencer. Fifteen DNA variants were identified, some of which are located within potential regulatory sites and possibly involved in the expression of the κ-casein encoding gene.

    Notizen: Abstract For the detection of polymorphisms within the 5′-flanking region of the β-lactoglobulin (-LG) -encoding gene a nucleotide sequence containing 795 bp of the promoter and 59 bp of exon I was cloned and sequenced. After comparing the sequence from the DNA of 11 diverse cows (different breeds and milk-protein yields), 14 singlebp substitutions were identified within the 5′-flanking region and two in the 5′-untranslated region (5′-UTR) of exon I. Some of the variants are located in potential binding sites for trans-acting factors or in the 5′-UTR. A PCR-based RFLP analysis was performed, and the genotypes of an additional 60 cows were identified at five variable 5′-flanking sites. The results reveal three frequent combinations between the A and B alleles of the protein-coding region and the novel 5′-flanking DNA variants. This finding may explain the differences of the protein-variant-dependent β-LG synthesis (A〉B) observed in vivo. A sequence comparison of the bovine and ovine promoters reveals an homology of 92.8% and shows a higher degree of conservation between positions -600 and -300.

    Notizen: A new modification of a focusing SANS instrument where at least the first crystal is set for diffraction in the asymmetric transmission geometry is presented. Unlike an earlier version which employed both crystals in the symmetric Bragg-case geometry [Kulda & Mikula (1983). J. Appl. Cryst. 16, 498–504], the new modification permits one simultaneously to exploit the effects of natural wavelength focusing and space condensation of the diffracted beam due to asymmetric diffraction. This DBC instrument seems to be suitable for use with beams with a large cross section at the first crystal and for SANS experiments on samples with a small width (few mm).

    Notizen: Some experimental results are presented for an elastically bent perfect silicon crystal in a strongly asymmetric diffraction geometry as a neutron monochromator. The use of this unconventional geometry of the monochromator appears to be suitable for a wide (several centimetres) incident polychromatic beam, when, thanks to the spatial condensation of the diffracted neutrons (Fankuchen effect), a high monochromatic beam density may be obtained. Furthermore, when using focusing in real and in momentum space by adjusting an optimum bending radius, the intensity diffracted by a sample may be comparable even with the best mosaic monochromators such as highly oriented pyrolytic graphite (PG). A comparison is demonstrated on the rocking curves of a strongly mosaic Ni–Al(020) crystal obtained with the monochromatic beam from bent Si(111), Si(400) and from PG(002), Cu(220) mosaic monochromators.
