    Notes: The naturally occurring enantiomer of cocaine, (–)-cocaine, has been previously labeled with 11C on the N-methyl group and used in conjunction with positron emission tomography to show that cocaine is rapidly taken up in the striata of human and baboon brain. In the present study, the behaviorally inactive (+)-cocaine was similarly labeled, with a view to its use for measuring the nonspecific binding of cocaine. No brain uptake was seen, although transport of cocaine into the brain is not expected to be stereoselective. The explanation for the lack of uptake was determined to be very rapid metabolism of (+)-cocaine in the blood. By 30s after administration of labeled (+)-cocaine, it was undetectable in plasma. In vitro studies demonstrated that (+)-cocaine is 50% debenzoylated to (+)-ecgonine methyl ester within 5 s of exposure to baboon plasma but not to washed erythrocytes. The hydrolysis of (–)-cocaine is at least 1,000 times slower. Serum butyrylcholinesterase (EC 3.1.1.8) appears to be responsible for this hydrolysis, as evidenced by its inhibition by physostigmine and catalysis by commercially available pseudocholinesterase from horse and human blood.

    Description / Table of Contents: Resume On examine ici les contraintes secondaires causées par les directions de cisaillement dans des joints de parois vitrées à base d'élastomères. A partir des courbes de déformation tracées pour deux joints fabriqués par Dow Corning, on a obtenu des valeurs de module de Young et de coefficient de Poisson tout à fait typiques. On calcule la distribution des contraintes dans un joint de 10 mm×10 mm×50 mm soumis à un cisaillement transversal en utilisant les propriétés de rigidité initiale dans un modèle analytique établi suivant la méthode de l'élément fini. On observe de légères différences entre les modèles linéaire et non linéaire, car le joint semble répondre de façon presque linéaire aux petits déplacements considérés dans cette étude. Le modèle analytique indique que la composante de traction σ22 est maximale à un niveau proche de l'interface d'adhérence et des coins du joint. A cet endroit du joint adhésif, la composante d'adhérence σ22 est environ deux fois plus importante que la contrainte de cisaillement, ce qui explique pourquoi les joints de parois vitrées ont tendance à la rupture dans des essais de cisaillement à proximité de la surface du support.
    Notes: Abstract The paper investigates the secondary stresses induced by shear movements in elastomeric structural glazing sealants. Typical values of Young's modulus and Poisson's ratio are derived from load-displacement curves measured for two structural glazing sealants (Dow Corning products). Using the primary stiffness properties in an analytical model based on the finite-element method, the stress distribution in a 10 mm×10 mm×50 mm joint subjected to transversal shear is calculated. Little difference is observed between the linear and non-linear models, as the sealant seems to respond nearly linearly to the small displacements considered in this study. The analytical model shows that the tension component σ22 exhibits a maximum at a level close to the adhesion interface and close to the joint edges. At this location in the adhesive joint, the tension component σ22 is approximately twice as large as the shear stress τ, which explains why structural glazing sealants tend to fail in shear tests close to the substrate surface.

    Notes: Abstract The cystic fibrosis transmembrane conductance regulator (CFTR) gene of 110 cystic fibrosis (CF) patients from the south-west of Germany was screened for 12 different mutations. This analysis resulted in an identification of 79% of all CF mutations and a complete genotype in 66% of the families. The most common mutation found was ΔF508 (67%). Another 5 mutations accounted for a further 12.5% (4% G542X; 3% R553X; 3% N1303K; 2% 1717-1 G→A; 0.5% G551D) whereas 6 mutations (R117H, A455E, ΔI507, S549I, S549N, and R1162X) were not found. Fifty-four (49%) patients were AF508 homozygotes and 18 (16.5%) were compound heterozygotes for ΔF508 and one of the rarer mutations. These frequencies differ slightly from those found in the north of Germany and considerably from those reported from the south of Europe, which seems to be consistent with a north to south decline of the relative abundance of ΔF508. Two patients, age 6 and 25 years, were compound heterozygotes for G542X and N1303K. The clinical features of the 6 year old were characterised by severe gastrointestinal and as yet only mild pulmonary complications whereas the 25 year old manifested severe pulmonary and gastrointestinal symptoms indicating that the N1303K mutation of the C-terminal CFTR nucleotide binding fold significantly impairs protein function in both the pancreas and the lungs.

    Notes: Summary It has been shown previously that verapamil and other calcium antagonists and calmodulin inhibitors can reverse multidrug resistance. We compared the potency of the dihydropyridine derivatives (4R)-3-[3-(4,4-diphenyl-l-piperadinyl)-propyl]-5-methyl-1,4-dihydro-2,6-dimethyl-4-(3-nitrophenyl)-pyridine-3,5-dicarboxylate-hydrochloride (B859-35), a metabolite of B859-35, niguldipine and (R)-nitrendipine to that of (RS)-verapamil in reversing multidrug resistance. The accumulation of the fluorescent dye rhodamine 123, which is transported by the P-glycoprotein, was determined by a flow cytometer. Multidrug-resistant human HeLa KB-8-5 and Walker rat carcinoma cells were incubated in the presence and in absence of the drugs indicated above. We found that 0.1 μM B859-35 increases the accumulation of rhodamine 123 in multidrug-resistant KB-8-5 and Walker cells more effectively than 1 μM (RS)-verapamil. In sensitive KB-3-1 cells addition of the drugs had no significant influence on the accumulation of rhodamine 123. In KB-8-5 cells, 10 nM Adriamycin caused a reduction of cell growth to 85% compared to untreated controls (=100%). If 1 μM B859-35, B859-35 metabolite, niguldipine, verapamil or (R)-nitrendipine was added to 10 nM Adriamycin, growth reduction compared with untreated controls increased to 12%, 11%, 23%, 63%, and 82% respectively. The effect of 0.1 μM B859-35 was a reduction in proliferation to 38%, that of 0.1 μM verapamil to 72%. These data illustrate that B859-35, a compound with antitumor activity in several tumors, is at least ten times more potent than racemic verapamil in reversing multidrug resistance.
