Notes: Summary By using oligonucleotide hybridization, restriction endonuclease analysis and direct sequencing of amplified genomic DNA, we have been able to characterize 18 different mutations in the β-globin genes of 161 β thalassemia homozygotes and 107 β-thalassemia heterozygotes from Turkey (429 β-thalassemia chromosomes). Previous studies dealing with β-thalassemia in Mediterranean countries have shown that, in most Mediterranean populations, only a few mutations are prevalent. In contrast, β-thalassemia in Turkey does not seem to be associated with a few predominant mutations. The six most frequent alleles, IVS-I-110 (G→A), IVS-I-6(T→C), FSC-8 (-AA), IVS-I-1(G→A), -30(T→A) and FSC-5 (CT), account for only 69.3% of the disease genes; indeed, all 26 mutations assayed represent 85.8% of the disease genes, confirming the considerable molecular heterogeneity of β-thalassemia among Turks, and indicating the possible presence of rare, previously undefined, mutations in the population. Two mutations observed in this study, IVS-I-116 (T→G) and Cd44(-C), have not been reported in the Turkish population to date. Since preventive medical services, such as genetic counseling and prenatal diagnosis, are greatly improved by detailed knowledge of the molecular pathology of β thalassemia, we strongly believe that the presented data will facilitate the intended establishment of a prenatal diagnosis center, based on DNA analysis, in Turkey.

Notes: A new algorithm has been developed for a single phase relay. This can be used for the protection of both single phase and three phase transformers. This paper presents a new scheme using the real time integration technique to determine the differential current caused by either magnetizing inrush current or an internal fault. The proposed relay has been simulated for continuous monitoring of the behavior of the differential current wave form, through the peak value of each current cycle and digital integration of that cycle. The trip of the relay depends on any changes in the area under the differential current wave form. The magnetizing inrush current waveform has less area under its curve than the fault current wave form which contains a sinusoidal wave and a transient dc component. This phenomenon has been used to block the operation of the relay on "not true'' fault current seen as differential current.

Notes: The dc linear motor described here is a moving coil type using four permanent magnets (SmCo5) in the stationary unit. Four frictionless U-shaped springs are used as a new type of supporting mechanism for the mover. This linear motor can be controlled by either proportional controller or proportional, integral, and derivative controller. However, the springs have a natural frequency and it vibrates the motor. For this reason, an additional circuit is used in the feedback loop which eliminates the vibration of the springs and compensates for the delay of phase of the filter. This filter avoids the resonance of the springs and therefore the controller cannot only be used in this motor but also for the control of machines with some kind of vibrating mechanism. This dc linear motor has a new type Linear Variable Differential Transformer as a position detector.

Notes: Summary The LEU2 gene of a his3 strain was inactivated by inserting the HIS3 gene between two overlapping inactive leu2 gene fragments, and mitotic stability of the resulting leu2: HISS: : leu2 sequence was measured under leucine repression and derepression. Both inactive leu2 regions were transcribed under derepressing conditions (growth in low leucine), and the LEU2 gene was completely restored by illegitimate recombination between the overlapping tandem repeats, leading to the loss of the intervening HIS3 gene. In contrast, only the downstream leu2 fragment was transcribed upon leucine repression, and the HIS3 insert in the leu2 region remained intact. The reciprocal experiment (inactivation of the HIS3 gene by inserting the marker gene LEU2) revealed a moderate rate of HIS3 restoration and LEU2 excision, reflecting transcriptional activity of the HIS3 region intermediate between that of LEU2 transcription in the repressed and derepressed state.
