    Notizen: We have examined the changes induced by the monoamine oxidase (MAO; EC 1.4.3.4) inhibitors tranylcypromine, clorgyline, and deprenyl on MAO activity and 5-hydroxytryptamine (serotonin, 5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) content in rat brain and blood (plasma and whole blood). The decreases of MAO-A activity observed in the liver and lungs after different doses of clorgyline or tranylcypromine correlated significantly (r 〉 0.80 in all cases) with the decline of plasma 5-HIAA. This was unaffected by 0.25 and 5 mg kg−1 of deprenyl, indicating that 5-HT was deaminated exclusively in the periphery by MAO-A. It is interesting that very potent and significant correlations (r 〉 0.75) were found between plasma 5-HIAA and MAO-A activity, 5-HIAA and 5-HT content in brain tissue. These results suggest that plasma 5-HIAA can be used confidently as a peripheral indicator of the inhibition of MAO-A in brain. This may represent a favorable alternative to the analysis of 5-HIAA in CSF in psychiatric patients undergoing antidepressant treatment with nonspecific MAO inhibitors or with the new selective MAO-A inhibitors.

    Notizen: Abstract: The effects of a single dose of p-chlorophenylalanine on the mRNA encoding tryptophan-5-hydroxylase (EC 1.14.16.4) in the rat dorsal raphe nucleus were analyzed using in situ hybridization. The levels of tryptophan-5-hydroxylase mRNA were markedly increased in cell bodies located in the ventromedial part of the dorsal raphe 1–2 days after p-chlorophenylalanine (300 mg/kg, i.p.) administration. This was followed by a decrease in the amount of tryptophan-5-hydroxylase mRNA, which returned to basal values by 5 days after treatment. An almost symmetric time course was observed for the midbrain serotonin concentration. Our results on the temporal pattern of changes in tryptophan-5-hydroxylase mRNA levels in the ventromedial part of the dorsal raphe are opposite to those reported for the enzyme activity and serotonin concentration after p-chlorophenylalanine treatment. These changes may result from modifications in enzyme mRNA expression, suggesting that tryptophan-5-hydroxylase gene transcription is involved in feedback mechanisms regulating serotonin synthesis.

    Notizen: Abstract. The study includes mating and spawning data of freshwater crayfish, Austropotamobius pallipes Lereboullet, obtained in four consecutive years (1987–1990). A total number of 586 wild-caught female A. pallipes in the 21·65 mm to 45·60mm carapace length (CL) size were held under laboratory conditions at different dates in September–October. Initial densities at mating time were 21 and 24 crayfish/m2 with a 1:2 male:female ratio. High percentages of mating were obtained (97·4–100%) except in 1989 (57·5%). In all years, most of the matings (90%) were concentrated in a period of 8–12 days with mean water temperatures between 12°C and 13·5°C. Spawning took place a few days after mating (minimum 2 days, maximum 14 days) at mean water temperatures between 8·2°C and 10·8°C. In the first 3 years, almost 100% of mated females spawned. However, in the fourth year (1990), when the mean size of crayfish was smaller, only 72·4% of mated females spawned. The mean number of pleopodal eggs was 64 (maximum 220, minimum 18). Egg diameter ranged between 2·30 and 3·25 mm (mean value 2·78 mm). Pleopodal egg number was positively correlated with carapace length (r= 0·72).

    Notizen: The CD5 membrane molecule, initially identified as an exclusive T-cell marker, also defines a phenotypically and functionally distinct B-lymphocyte population. In normal individuals, CD5+ B cells are committed to secrete‘natural’polyreactive (auto)antibodies, that is antibodies, mainly IgM, endowed with multiple antigen-binding capabilities, including rheumatoid factor (RF) activity. At variance with this, in rheumatoid arthritis (RA) as well as in other autoimmune conditions, monoreactive autoantibodies binding with high affinity and specificity to a given self antigen have been isolated and the cells from which they originate differently related to the CD5+ B-lymphocyte subset.Here, we studied the proportions of CD5+ B cells and the characteristics, in terms of polyreactivity and monoreactivity, of RF produced by B lymphocytes in RA patients with classified disease activity. Our results suggest that patients with a more severe disease activity have higher proportions of CD5+ B cells and higher frequencies of B lymphocytes committed to secrete RF, with the characteristics of polyreactive antibodies. On the other hand, we did not find a significant difference between the proportions of peripheral B cells producing monoreactive RF in patients with high- versus patients with low-activity RA. However, in two highly active RA patients, we found that synovial fluid, compared with peripheral blood, was significantly enriched for (IgG and IgA) monoreactive RF-producing B cells.

    Notizen: Abstract In a four year study data on the presence of red squirrel were collected in an agricultural landscape by counting dreys in 49 woods ranging from 0.5 to 14 ha, and differing in quality of habitat and isolation. Logit regression analysis showed that the area per woodlot covered with conifers is a good predictor of squirrel presence for each year and during the whole period, but the significance of the regression decreases with time. During the study the number of woods occupied by red squirrel increased, and smaller woods and those without conifers also became inhabited. This trend is in accordance with the positive effect of time in regression analyses on the presence of the species and on the colonization of woods, and it suggests an increase of squirrel numbers in the area. Addition of several isolation variables in the regression analyses showed significant effects in different years, and the effect of isolation was independent of time. In the first two years the area of habitat around a woodlot, the distance to the nearest woodlot larger than 30 ha, and the density of possible movement corridors have significant effects on the presence of red squirrel. In the last two years, with presumably a high number of squirrels, the (short) distance to the nearest woodlot and also the area of habitat around woods have significant effects. It is concluded that the spatial dynamics of the population can be understood as the outcome of individual spatial behaviour, rather than as the result of metapopulation processes.

    Notizen: Summary We have examined the local and systemic effects of clorgyline, tranylcypromine and deprenyl on extracellular serotonin (5-HT) and 5-hydroxyindoleacetic acid in the raphe nuclei and in frontal cortex of awake, freely-moving rats using microdialysis. When administered through the dialysis probe, monoamine oxidase (monoamine: oxygen oxidoreductase (deaminating), E.C. 1.4.3.4., MAO) inhibitors increased 5-HT output in a dose-dependent manner in both brain areas. The effects were more pronounced in the raphe nuclei for the three MAO inhibitors at all doses assayed. When the monoamine oxidase inhibitors were given i.p., dialysate 5-HT increased dramatically, after tranylcypromine (15 mg/kg), in raphe nuclei and frontal cortex (area under the curve (AUC) to 4 h post-treatment: 63-fold and 11-fold, respectively) whereas the effects of clorgyline (10 mg/kg) were much less pronounced (+ 47% increase in the AUC for raphe nuclei, P 〈 0.09; + 18% increase in the AUC for frontal cortex, n.s.). Deprenyl (2.5 mg/kg, i.p.) induced a moderate (+ 22%) increase of dialysate 5-HT from the raphe nuclei but did not cause a change in dialysate 5-HT from the frontal cortex (+ 4%). However, clorgyline, or deprenyl, dramatically increased dialysate 5-HT in animals which had been pre-treated with the above dose of deprenyl, or clorgyline, respectively, showing that the blockade of both forms of MAO results in much larger increases of extracellular 5-HT than does the blockade of either form alone. These results indicate that: (a) deamination by MAO participates actively in the control of the extracellular concentration of 5-HT in those areas of the brain that are rich in serotoninergic nerve terminals as well as in cell bodies, (b) in vivo, brain 5-HT is deaminated preferentially by MAO-A but its full inhibition does not result in an increased release of 5-HT, in spite of a large accumulation of 5-HT in the brain tissue, (c) MAO-B deaminates 5-HT when the A-form is inhibited (in this situation, MAO-B participates actively in the control of a releasable pool of 5-HT), (d) the raphe nuclei appears to be a preferential site of action of MAO inhibitors, administered either locally or systemically. These results may help to understand the model of action of MAO inhibitors as antidepressant drugs.
