    Notes: It is demonstrated that a longitudinal magnetic field applied to a low-pressure, low-voltage, hot cathode glow discharge will substantially increase the intensity of metastable atoms effusing from a central anode slit. For an argon discharge there is a fourfold increase in the effusing Ar(3P2,0) atom intensity and for neon there is a 14-fold increase in Ne(3P2,0) intensity.

    Notes: Metastable atoms emitted from the anode of a hot-cathode low-voltage Ar gas discharge produce two peaks in the time-of-flight distribution. The slow peak typifies atoms with a thermal velocity distribution in the discharge. The fast peak can be approximated by a Gaussian function, and its dependence on the discharge current, voltage, and pressure is presented. The origin of the fast peak is investigated experimentally and shown to represent fast Ar(3P2,0 ) from the discharge.

Since plasma renin activity is increased in cyclosporin A (CsA)-induced hypertension in the rat, the role of the vascular renin-angiotensin system (RAS) in CsA-induced hypertension was investigated in rat mesenteric resistance vessels.2. Female Wistar rats received CsA (10 mg/kg per day, s.c.) or vehicle for 30 days. CsA treatment increased tail-cuff systolic blood pressure (CsA treated 135 ± 3 mmHg vs control 125 ± 1 mmHg, P〈0.0001).3. Mesenteric resistance arteries (200–300 μm) were isolated and mounted in a microvessel myograph. Concentration-response curves to tetradecapeptide renin substrate (10-11-10−6 mol/L), angiotensin I (10-l1-10−6 mol/L) and angiotensin II (10-12-10−6 mol/L) showed no differences between CsA-treated and control groups.4. Mesenteric vascular angiotensin-converting enzyme (ACE) characteristics were determined by radioligand binding. There were no differences in the content or affinity of ACE between CsA-treated and control rats.5. These results suggest that the mesenteric vascular RAS does not play a major role in CsA-induced hypertension in the rat.

    Notes: 1. The aim of these studies was to determine the role of endothelin in cyclosporin A ([CsA], 10 mg/kg per day, s.c., for 30 days) hypertension in the rat.2. There were no significant differences in plasma endothelin concentrations, in vitro mesenteric vascular membrane [125I]-endothelin-l (ET-1) binding characteristics or myographic vascular responses to endothelin-1 between the CsA and control groups (systolic blood pressure 135 ± 1 vs 127 ± 1 mmHg, respectively, P〈0.001).3. Twenty-four hours after selective endothelin type A receptor blockade in vivo with BQ123, blood pressure in CsA hypertensive rats was lowered to levels of normotensive controls.4. These results suggest that changes in endothelin synthesis rather than changes in vascular sensitivity and/or ET-1 receptor characteristics may contribute to CsA-induced hypertension.

    Notes: Abstract Kinetensin is a nonapeptide, originally isolated from pepsin-treated plasma, that shares some sequence homology with the C-teminal end of neurotensin. The present study was designed to determine, by infusing kinetensin to conscious sheep, the pharmacokinetics and a neurotensin-like biological activity (pancreatic polypeptide response) of kinetensin. Kinetensin was rapidly metabolized, approximately 200-fold more rapidly than neurotensin [1–13]. The majority of the metabolism occurred in the circulation as demonstrated bothin vivo andin vitro. The lung and gut cleared kinetensin also. Inhibition of converting enzyme, present in highest concentration in the lung, abolished lung clearance but was without effect on kinetensin metabolism by the gut or in the general circulation. Arterial infusion of kinetensin which achieved high blood kinetensin levels at the pancreas did not increase plasma pancreatic polypeptide. We conclude that the extremely rapid degradation of exogenous kinetensin, together with the lack of biological activity, makes it unlikely that kinetensin plays a role as a circulating regulatory peptide. Nevertheless, since the putative kinetensin substrate circulates at μM concentrations, it is feasible that kinetensin is generated and metabolized at the target organ.

    Notes: Summary After performing selectively 25 laparoscopic cholecystectomies (LC) to determine the place of LC in the management of complicated gallstones, all patients presenting with gallstones were evaluated by the authors for LC. Eighty-six consecutive patients were evaluated and 84 were studied. Follow-up in every case exceeded 6 months. In three of 10 patients with acute cholecystitis, LC was not possible; each had a history longer than 48 h and all had gangrene of the gallbladder. In four patients with empyema, LC was successful, but operative cholangiography failed. Operative cholangiography was successful in 76 of the remaining 77. Of eight patients suspected of having stones in the CBD, cholangiography excluded stones in six and confirmed them in two. Cholangiography identified three other patients with totally unsuspected CBD stones. Of the five patients with CBD stones, four had them flushed to the duodenum at LC following transcystic balloon dilatation of the papilla and one had a post-op. ERCP. Of four patients with acute pancreatitis, three had LC in the same admission. LC was possible in all three patients with morbid obesity. We conclude that with experience, LC is possible for complicated gallstones. In acute cholecystitis, the probability of success is higher with earlier operative intervention. Operative cholangiography is essential. It not only identifies unsuspected CBD stones but also allows LC without ERCP in those with suspected CBD stones and with modification it allows treatment of those stones.
