    Notes: Abstract A recombinant CHO cell line producing human prorenin was cultivated on microcarriers in serum-free medium. Best growth was obtained when the cells were cultivated on a collagen coated microcarrier (Cytodex 3) using a serum-free medium (SF-02) supplemented with fibronectin. It was possible to reduce the necessary concentration of fibronectin in the feed medium from 10 μg/cm3 to 2 μg/cm3 during perfusion cultures in a spinner reactor equipped with an UF-membrane. Also in this system, the prorenin concentration increased up to 8 times higher compared to that in a conventional repeated-batch culture. The cells grew in multilayers on the microcarriers during the perfusion culture. The specific prorenin productivity was not significantly affected by the cell growth rate, and the secretion of prorenin continued even after the cells had ceased to grow.

    Notes: Total discectomy was performed experimentally in the rabbit temporomandibular joint (TMJ). In order to evaluate the injury and repair of the articular cartilage of the mandibular condyle after discectomy, both histologic and immunohistochemical observations were made. Immunohistochemical observation by using bromodeoxyuridine (BrdU) and its monoclonal antibody in vivo labelling method was used.The present study demonstrated that the cartilage on the articular surface of the condyle disappeared at 1 week post-operatively. This defect in the injured articular cartilage was not repaired by the cartilage, but was covered by a fibrous connective tissue at 6 weeks post-operatively, where the condylar recontouring by the adjacent articular cartilage, and flattening and hyperplasia of the condyle were observed. Thus, the cartilaginous repair following discectomy was characterized by the proliferation of the adjacent cartilage to the injured site and osteoblastic formation in the bone marrow at the exposed subchondral bone.

    Notes: The ion beams are produced by a cylindrical thermal contact-ionization plasma source with an electrostatic lens system. The source plasma is produced at the inside surface of a 5-mm-diam 20-mm-long Re foil cylinder protected with a W cylinder. The surface is heated up to 3000 K by electron bombardment. The material to be ionized is fed to the surface through a guide pipe from a reservoir. Generation of all alkaline metal ions have been tested and a beam current of hundreds of μA is obtained. The efficiency for Li+ production is 30%, while that for Cs+ is nearly 100%. Alkaline earth ion beams, except Mg, are generated. A current of Ba+ up to 200 μA is obtained, while a Tl+ beam of over 200 μA is achieved. Halogen negative ion beams, except F−, of the order of 10 μA are also obtained when alkali halides are used. Attempts to extract Li− from the source are also being made.

    Notes: Both renin and cathepsin B were co-localized in identical granules of adrenocortical cells. At day 16 of gestation, many renin-containing granules were observed and gold particles showed homogeneous intragranular distribution; whereas, those for cathepsin B was distributed heterogene-ously. At day 18 of gestation, renin immunoreactivity was decreased or undetectable, whereas cathepsin B was still demonstrated at the same level as on day 16 of gestation.

    Notes: Abstract We evaluated the expression of inflammatory cytokines in renal tissues obtained from 45 patients with several types of glomerulonephritis. Immunofluorescence studies with specific antibodies to interleukin (IL)-1α, IL-1β, IL-6, tumour necrosis factor (TNF)-α, and TNF-β showed intense cytoplasmic staining in the glomeruli and interstitium. Cells positive for these cytokines were found frequently in tissue from patients with lupus nephritis (WHO Class IV) and membranoproliferative glomerulonephritis, and, to a lesser extent, in tissue from patients with mesangial proliferative glomerulonephritis, Henoch-Schönlein purpura nephritis, and minimal change nephrotic syndrome. Most of these cells were dual-stained with a monoclonal antibody to monocytes-macrophages. In situ hybridization for cytokine mRNA, combined with immunoperoxidase staining for monocytes-macrophages, detected IL-1α, IL-6, and TNF-α mRNA in monocytes-macrophages infiltrating the glomeruli and interstitium. Occasionally, there was weak or moderate immunostaining for IL-1α, IL-6, and TNF-α in the glomerular mesangial and epithelial cells, but in situ hybridization signals were rarely found in these loci. These findings suggest that infiltrating monocytes-macrophages, rather than resident glomerular cells, are the major source of inflammatory cytokines in human glomerulonephritis.

    Notes: Abstract The effect of illumination on transport of sulfonated bisazo direct dyes, CI Direct Yellow 12, and CI Direct Yellow 4, into a cellulose membrane has been studied at various temperatures. Transport of CI Direct Yellow 12, which exhibits photoinduced reversible trans-cis isomerism in aqueous solution into a cellulose membrane, was influenced by illumination. It is likely that the transport was influenced by transisomerization of the photoisomeric dye and the diffusivity was controlled by surface diffusion rather than by pore diffusion under both light and dark conditions.

    Notes: Abstract Pyridine insoluble fractions in the mesophase pitches derived from a decant oil (PMP-PI) and naphthalene (NMP-PI) were found to be rendered almost completely soluble in pyridine by the Diels-Alder reaction with maleic anhydride, maintaining their optical anisotropy and molecular association. Their solubility in pyridine reached up to 95 wt % by the reaction at 175–200 °C. Solubilized fractions were analysed to obtain their average structures, suggesting that PMP-PI consists of aromatic nuclei of peri-condensed rings connected through a small number of methylene and aryl-aryl linkages, having a molecular weight (MW) of ∼ 1800, and that NMP-PI consists of oligomeric naphthalene with a large number of naphthenic groups (MW ≃ 1000). Such structures are basically much the same as those of their soluble fractions, although the molecules in Pls were much larger. The reaction sites in such structures for the Diels-Alder reaction are discussed.

    Notes: Summary The phenotype of bovineβ-mannosidosis (β-mannosidase deficiency), recently identified in Salers cattle, is similar to the caprine form of the disease (Abbittet al., 1991). This investigation was designed to characterize accumulated kidney oligosaccharides in bovineβ-mannosidosis. Oligosaccharides were extracted from the kidney of an affected Salers calf and purified by chromatographic techniques. The amount of accumulating oligosaccharides in 1 g of wet tissue was about 21µmol. Structures of derivatized oligosaccharides were characterized by high-performance liquid chromatography, mass spectrometry, methylation analysis and sequential exoglycosidase digestions. The major accumulating oligosaccharides were Manβ1-4GlcNAc and Manβ1-4GlcNAcβ1-4GlcNAc. Oligosaccharides accumulating in minor amounts were Manβ1-4GlcNAcβ1-4Manβ1-4GlcNAc, Manα1-6Manβ1-4GlcNAcβ1-4GlcNAc and Manβ1-4GlcNAcβ1-4Manβ1-4GlcNAcβ1-4GlcNAc. As in caprineβ-mannosidosis, oligosaccharides with terminalβ-mannose residues and cleaved as well as uncleaved chitobiose linkages were identified in bovineβ-mannosidosis kidney. The accumulating oligosaccharides in tissue were thus identical in bovine and caprineβ-mannosidosis; however, the source of the novel oligosaccharides remains to be determined.
