    Notes: Summary A comparative pharmacokinetic study was performed with two types of suppository each containing 20 mg piroxicam. The bioavailability of piroxicam was studied in 24 healthy volunteers in a double blind, randomised, cross over study. No significant difference was found in the pharmacokinetic parameters [AUC(0–72 h), AUC, Cmax, t1/2β calculated from plasma concentration time curves, indicating that the two preparations were bioequivalent.

    Notes: Summary Seven fragments of the spike (S) gene cDNA of transmissible gastroenteritis virus (TGEV), as well as the full length cDNA, were cloned and expressed in baculovirus vectors. Piglets were immunized with cells infected with the recombinant viruses. Each of the recombinants induced TGEV-specific antibodies detected in a fixed cell enzyme immunoassay. The amino terminal half of the S protein, containing all four major antigenic sites (A, B, C and D), and encoded by a 2.2 kb fragment of the S gene, induced virus neutralizing (VN) antibody titers comparable with those induced by the complete S protein. Recombinant proteins lacking the A antigenic site, or with a deletion including the putative receptor binding sites and the D antigenic site, were not capable of inducing levels of VN antibodies similar to those induced by the whole S protein.

    Notes: Abstract Mixing models for bioreactors on the basis of the tanks-in-series concept are presented and a suitable parameter-estimation method is introduced. The Monte-Carlo-optimization procedure with the inhomogeneity-curve included in the objective function is used. Results of the parameter optimization procedure are given for stirred-tank-bioreactors equipped with one and three Rushton turbines under aerated conditions. The model designed for the stirredtank with three Rushton turbines is capable to describe the mixing properties, while in case of the stirred-tank with one Rushton turbine the simulated radial circulation time does not correlate with the measured one.

Metronidazole did not influence the high activity of daptomycin against strains ofStaphylococcus aureus, Staphylococcus epidermidis andEnterococcus faecalis in the absence or presence of the co-culturedBacteroides strains. In contrast, theEnterococcus faecalis isolates protected the co-culturedBacteroides strains against the killing effect of metronidazole, even at a concentration four- or eightfold the MIC of metronidazole. Killing curve experiments confirmed this protective effect of different isolates ofEnterococcus faecalis.

    Notes: Abstract In Canada, environmental regulations for protection of the biota from the adverse effects of effluents from petroleum refineries have tended to focus on acute toxicity. There is concern those effluents may have other subtle, but still deleterious, long-term effects on aquatic ecosystems. We have used a battery of toxicity tests to assess the acute toxicity, genotoxicity, and chronic toxicity of effluent samples from two Ontario refineries. The test organisms included representatives of the bacterial, algal, plant, cladoceran, and fish communities. The results of our preliminary study indicate that the effluent samples had little acute toxicity to the test organisms. There were indications of some sublethal toxicity to Ceriodaphnia dubia, Panagrellus redivivus, and Pimephales promelas. One of the effluents inhibited the growth of Selanastrum capricornutum (IC50 of 59.9%) and Lemna gibba (IC25 of 73.3%) and also caused a 15 percent reduction in the germination of Lactuca sativa seeds. The SOS-Chromotest, a commercially available test that measures the activity of a bacterial DNA repair system, detected genotoxic effects in a single effluent that had been concentrated ten fold. There was no apparent relationship between several chemical parameters and the observed sublethal effects. Further research is needed to establish whether or not the observed toxic effects are typical of effluents from Ontario refineries.

    Notes: Abstract Hydrolysis of olive oil into fatty acids by lipase in a flat membrane module is reported. Lipase was immobilized by adsorption onto a hydrophilic cellulose acetate membrane. Hydrolysis was carried out by circulating pure olive on the enzyme side of the membrane and water phase on the other side. Conversion reached 85 % after 50 hours reaction time.

    Notes: A new scale-up concept based upon mixing models for bioreactors equipped with Rushton turbines using the tanks-in-series concept is presented. The physical mixing model includes four adjustable parameters, i.e., radial and axial circulation time, number of ideally mixed elements in one cascade, and the volume of the ideally mixed turbine region. The values of the model parameters were adjusted with the application of a modified Monte-Carlo optimization method, which fitted the simulated response function to the experimental curve. The number of cascade elements turned out to be constant (N = 4). The model parameter radial circulation time is in good agreement with the one obtained by the pumping capacity. In case of remaining parameters a first or second order formal equation was developed, including four operational parameters (stirring and aeration intensity, scale, viscosity). This concept can be extended to several other types of bioreactors as well, and it seems to be a suitable tool to compare the bioprocess performance of different types of bioreactors. © 1994 John Wiley & Sons, Inc.
