    Notes: Summary Experimental neurofibrillary change was produced in rabbit brains by daily subcutaneous aluminum tartrate injection for 40 days. The production of experimental neurofibrillary changes was confirmed by immunostaining with antibodies against neurofilament triplet proteins and the brain tissue was studied immunohistochemically with antibodies against microtubule-associated protein (MAP) 2 and ubiquitin. The hippocampal neurons of the chronically aluminum-intoxicated rabbit brain showed diminished staining of dendrites by anti-MAP2 antibody. The length of anti-MAP2-positive dendrites in hippocampus was significantly shorter than that of the control brain. In the cortex somata of a subset of pyramidal neurons were intensively stained by anti-MAP2 antibody, while the MAP2 immunoreactivity of distal dendrites was diminished. The immunostaining by anti-ubiquitin antibody revealed the positive staining of the neurons bearing experimental neurofibrillary changes in the lower brain stem nuclei. It is speculated that MAP2 dislocation and ubiquitination are accompanying phenomena of the production of experimental neurofibrillary changes in chronically aluminum-intoxicated rabbit brains.

    Notes: Abstract As part of our effort to isolate and characterise the von Hippel-Lindau (VHL) disease gene, we constructed a physical map of chromosome 3p25-26 by fluorescence in situ hybridisation (FISH) studies on a panel of cytogenetic rearrangements involving this region. Biotinylated cosmid and lambda probes were hybridised to metaphase chromosome spreads and positioned with respect to each cytogenetic breakpoint. These studies unequivocally established the order of five loci linked to the VHL disease gene: cen-(RAF1,312)-D3S732-D3S1250-D3S601-D3S18-pter and determined the position of three other probes within this map. These results ordered RAF1 and D3S732 for the first time, confirmed the localisation of D3S1250 between RAF1 and D3S601 and determined the position of D3S651 with respect to other chromosome 3p25-p26 loci. The establishment of an ordered set of cytogenetic aberrations will enable the rapid assignment of polymorphic and nonpolymorphic cloned sequences within the chromosome region 3p25-p26.

    Notes: Summary Changes in tumour blood flow under an induced hypertensive state were examined in malignant brain tumours to know if the precondition for the effectiveness of induced hypertensive chemotherapy — relative increase in tumour blood flow — are fulfilled. Tumour blood flow was measured under both a resting and an induced hypertensive state in 12 patients with various malignant brain tumours (6 gliomas, 6 metastatic brain tumours) using xenon-enhanced computed tomography. The blood pressure was elevated 40% above the systemic blood pressure of the resting state by the infusion of angiotensin II. Tumour blood flow increased 30% on average above the normal brain tissue blood flow after the induction of an induced hypertensive state (p 〈 0.05). The tumour blood flow increased in 11 cases of malignant tumours, but decreased in one case with massive brain oedema after induced hypertension. The increase in blood flow was higher in hypervascular tumours and less in hypovascular tumours. Therefore, induced hypertensive chemotherapy probably will be more effective in hypervascular malignant brain tumours with small mass effects.

    Notes: Summary Temporary occlusion of the internal carotid artery with a balloon atheter (balloon Matas test) and simultaneous dynamic computerized tomography scanning (DCT) were performed to determine the tolerance of permanent carotid occlusion in eight cerebral aneurysm and two carotid-cavernous fistula (CCF) cases, in whom internal carotid occlusion might be necessary during operation or as a choice of treatment. All patients were evaluated by mean transit time (MTT), especially % transit time (MTT of the occluded side×100/MTT of the control side). In six patients, % transit time (%TT) was less than 155 and no neurological signs appeared after permanent internal carotid occlusion. Neurological deficit appeared when mean arterial blood pressure was 80, and disappeared when mean arterial blood pressure was 100 during the balloon Matas test in a case whose %TT was 200. Neurological deficit appeared several seconds after the balloon Matas test in a case whose %TT was 250. The critical %TT value to cause symptomatic ischaemia was 200 from our results. Therefore, it is necessary to undertake treatment such as bypass surgery for the patients whose %TT is near 200, even if the balloon Matas test was negative.

    Notes: Abstract An improved rabbit ear chamber was used to evaluate calcium hydroxide-containing root canal sealers on their potential in induce dystrophic calcification in connective tissue. Four sealers and two control materials were introduced into the chambers and the effects of these materials on the living vascular tissue were observed continuously under a biomicroscope up to 9 weeks. Conventional histopathological investigation and examinations with a scanning electron microscope and an X-ray microanalyzer were done to supplement the results. Sealapex and Calvital (Ca(OH)2+ iodoform) revealed almost the same tissue reaction as calcium hydroxide-saline paste; they rapidly made a precipitate-barrier of calcium phosphate in the connective tissue, inducing calcification. However, Dentalis KEZ(ZnO-ca(OH)2+ eugenol) caused mild disorders of microcirculation without calcification, as well as Canals(ZnO + eugenol). New A (Ca(OH)2+ fatty acid) had good compatibility with micro-vessels as well as New B(ZnO + fatty acid), however they induced no calcification and disintegrated rapidly in the tissue. These sealers were reclassified according to what they actually bring about in the tissue, not according to what they include.

    Notes: Lysyl-tRNA synthetases are synthesized in Escherichia coli from two distinct genes, lysS and lysU, which are regulated differentially. A strain which is null for lysS, the constitutive gene, was created by gene disruption (lysS1) and exhibited cold-sensitive lethality. Hence, lysS is dispensable at high temperatures. This cold sensitivity was suppressed by a multi-copy plasmid carrying lysU, the inducible gene. These data are interpreted as indicating that lysS is functionally replaceable by lysU for cell growth, and that the cold sensitivity of lysS1 is caused by insufficient expression of lysU at low temperatures. To investigate the mechanism of lysU expression, cold-resistant bypass mutations were isolated from lysS1, and named als (for abandonment of lysS.). Two als mutations which were linked to lysU contain IS2 insertions upstream of the lysU promoter. They caused a 16–19-fold increase in the lysU-mRNA level. Furthermore, deletion mutations created immediately upstream of the lysU promoter restored growth of lysS1. These results suggest that transcription of lysU is negatively controlled by a cis-element located upstream of the promoter.

    Notes: A ridge quantum wire structure has been successfully fabricated on a patterned (001) GaAs substrate by first growing a (111)B facet structure with a very sharp ridge and then depositing a thin GaAs quantum well on its top. Electron microscope study has shown that a GaAs wire with the effective lateral width of 17–18 nm is formed at the ridge top. Photoluminescence and cathodoluminescence measurements indicate that one of the luminescence lines comes from the wire region at the ridge and its blue shift (∼60 meV) agrees with the quantum confined energy calculated for the observed wire structure.

    Notes: Mechanisms of molecular beam epitaxy have been investigated for GaAs and AlAs by growing and analyzing the shapes of facet structures consisting of an (001) top surface and two (111)B side surfaces. It is found that all of the Ga flux on the three facet planes is incorporated into the film, but the growth rates on (111)B and (001) depend strongly on the As flux and are mainly determined by the diffusion of Ga ad-atoms between the two planes. In contrast, the diffusion of Al is found to be almost negligible, irrespective of the As flux. By analyzing the shape of the facet, the diffusion length, λ, of Ga on a (001) surface is estimated to be about 1 μm at 580 °C, while that of Al is about 0.02 μm. On (111)B, λ of Ga is found to be several μms. The reflectivity of diffusing Ga atoms is found to be far less than 1 for the (001)-(111)B boundary, and almost unity at facet boundaries where the (111)B side surfaces are bound by the (11¯0) side walls.
