    Notes: Abstract: A novel, apparently acid-stable cytochrome has been purified from Leptospirillum ferrooxidans in which it probably functions in the ‘downhill’ transfer of electrons from ferrous iron. It appeared to comprise a single polypeptide of apparent Mr 17.9 kDa as determined by SDS-PAGE. Non-denaturing PAGE showed this to be the approximate size of the native protein. The oxidized cytochrome showed a broad absorption maximum at 422 nm and was readily reduced with dithionite or ferrous iron to give a form with absorption maxima at 440 and 579 nm. A mid-point potential of +680 mV at pH 3.5 was determined. Iron and zinc were found at concentrations approaching one atom of each per cytochrome molecule.

    Notes: Paracoccus denitrificans was cultivated in continuous culture in media deficient in manganese, or copper or both metals and the effects on growth and the respiratory chains studied. Manganese deficiency had no effect on growth yield or the levels of cytochrome c oxidase but the amount of c-type cytochromes was decreased to about 35% of control values and a CO-binding b-type cytochrome was also produced. However, the major functioning oxidase was still cytochrome c oxidase. The ESR and optical signals of copper were present or absent if cells were grown in the presence or absence of copper respectively, independent of the concentration of manganese available. This allows the ESR properties of the native and copper-deficient cytochrome c oxidase to be studied as the spectra are now not obscured by signals from manganese.

    Notes: Abstract: A moderately thermophilic mixed culture of bacteria catalysed the oxidative solubilization of arsenopyrite to give Fe(III), S(VI) and As(V). Toxic effects were observed in a few experiments due to teh build-up of As(III). The bacterial oxidation of arsenopyrite involved direct attack of the bacteria on the mineral to give AS(III). Subsequent oxidation of AS(III) to AS(V) occurred reaction with FE(III), but only in the presence of pyrite, which provide a catalytic surface. Arsenopyrite was unable to act as a catalyst. The pyrite- catalysed oxidation of As(III) to AS(V) by FE(III) usually only went to completion in the presence of bacteria, possibly due to their role in the provision of clean catalytic surfaces. Thus, toxic concentrations of As(III) may accumulate in reactors during the bacterial oxidation of arsenopyrite due to the absence of pyrite or a clean pyrite surface or to low concentrations of the effective oxidizing agent, Fe(III).
