    Notes: We have constructed an extremely low noise constant current power supply. The design was optimized for low noise at 0.5 Hz. For a 134 mA supply, noise at this frequency is 2.5 nA rms/ (square root of)Hz. At 1 Hz, it is 1.0 nA rms/(square root of)Hz. The supply is roughly an order of magnitude quieter than the quietest commercial unit we know of.

    Notes: The paper outlines how consent standards are normally calculated in Scotland for both river systems and tidal waters. A background is given to the reason why a more statistical approach has been adopted in England and Wales where the use of combining distribution methods is more widespread. Comparison of the two approaches is made, particularly with regard to their potential impact on resources required for treatment plants, and also the problems of taking enforcement action. The setting of standards under the EC urban waste water treatment Directive is considered where, in addition to the implications of primary, secondary and more stringent treatment, consideration is given to storm-sewage overflows and industrial discharges.

    Notes: Abstract The gene for the β-(1,3) exoglucanase of Candida albicans was used as a probe to detect transcripts of related genes in C. albicans and in several other Candida species. A single homologous transcript was detected in all of the species tested. Expression of the exoglucanase gene in C. albicans was found to be coincident with the onset of growth and the levels of the transcript were proportional to the growth rate. Comparable levels of the transcript were produced during yeast and hyphal forms of growth.

    Notes: Abstract This paper deals with the question of optimal debugging times for large-scale computer programs, given certain assumptions concerning the distribution of error within the program, the cost that is incurred whenever a fault occurs, and the cost of debugging. Modeling takes place within the framework of independent Poisson processes describing faults caused by errors. The optimization aim is to minimize the long-run total expected discounted cost. Some general results concerning the optimal polity are presented.

    Notes: Summary Diabetic retinopathy is the leading cause of blindness in adults aged 30 to 65 years. However, 20% of the diabetic population does not develop significant retinopathy. To examine the influence of immune-related genetic factors on the development of diabetic retinopathy, we studied immunoglobulin allotypes in 102 subjects aged 8 to 20 years, who had had Type 1 (insulin-dependent) diabetes mellitus for 4.5 to 11 years (mean 7.3 years). HLA had been previously typed on 59 of these subjects. Retinopathy was assessed by expert review of retinal photographs. Among the 44 patients who had evidence of retinopathy, 33(75%) were G2m(23 +), while among the 58 patients without retinopathy but with similar duration of disease, only 28(48%) were G2m(23 +) (p = 0.006). The HLA-DR types of patients with and without retinopathy were not significantly different. We conclude that there is significant evidence of an association between G2m(23) at the locus encoding IgG2 subclass heavy chains and susceptibility to the development of diabetic retinopathy early in the clinical course of the disease. Our findings provide important independent confirmation of a previous report of association between Gm allotypes and predisposition to diabetic retinopathy. We are unable to determine if the Gm effect on development of retinopathy is due to the G2m(23) allotype itself, or due to genes that are closely linked to, and in linkage disequilibrium with, the locus encoding the G2m(23) allotype.

    Notes: Summary The biochemical and behavioural effects of isamoltane, a \-adrenoceptor and 5-HT1B receptor antagonist that has higher affinity for 5-HT1B receptors than for 5-HTIA receptors, on 5-HT neurotransmission in the rat brain were examined. In binding experiments isamoltane was found to be about five times more potent as a ligand for the 5-HT1B receptor than for the 5-HT1A receptor (Ki values 21 and 112 nmol/l, respectively). Isamoltane increased the K+-evoked overflow of 3H from 3H-5-HT loaded slices of rat occipital cortex at 0.1 μmol/l, consistent with inhibition of the terminal 5HT autoreceptor. In vivo, isamoltane significantly increased the concentration of 5-hydroxyindoleacetic acid in hypothalamus and hippocampus indicating an increased 5-HT turnover with a maximal effect at 3 mg/kg s.c. A higher dose produced a less pronounced effect. This effect did not seem to be due to the β-adrenoceptor blocking action of isamoltane since the β-adrenoceptor antagonists, (−)-alprenolol, betaxolol or ICI 118,551 had no significant effects on 5-HT turnover at 5 mg/kg s.c. Isamoltane at 3 mg/kg s.c. induced the wet-dog shake response which was blocked by the tryptophan hydroxylase inhibitor p-chlorophenylalanine. In contrast, the same response induced by the 5-HT2 receptor agonist quipazine was not blocked by pretreatment with p-chlorophenylalanine. The wet-dog shakes evoked by isamoltane and quipazine were blocked by ritanserin, which indicates that 5-HT2 receptors are involved in their expression. These observations indicate that isamoltane, by inhibiting the terminal 5-HT autoreceptors, increased the synaptic concentration of 5-HT to a level that induced a behavioural response.

    Notes: Abstract NCQ 298 ((S)-3-iodo-N-[(1-ethyl-2-pyrrolidinyl)methyl]-5,6-dimethoxysalicylamide) has an iodine substituent. We have labelled NCQ 298 with123I and125I, and used the radioligands as tracers in receptor studies in vitro, in vivo in autoradiography and in SPECT studies on Cynomolgus monkeys. [125I]NCQ 298 bound in vitro to a single binding site with a KD=19 pM. NCQ 298 has thus a 10-fold higher affinity for the dopamine D2 receptors than the corresponding des-5-methoxy compound FLA 961 (IBZM), previously used in SPECT studies. The binding of [125I]NCQ 298 was entirely reversible (T1/2=17.5 min at 37° C). Autoradiographical studies in vitro on rat and monkey brain tissue sections showed a distinct binding in caudate-putamen, nucleus accumbens, substantia nigra, and in layer 5 of the cerebral cortex. In vivo binding studies in mice showed a ratio of 10 between [125I]NCQ 298 binding in striatum and cerebellum. Binding was displaced by the selective dopamine D2 receptor antagonist raclopride. In SPECT studies with [123I]NCQ 298 in two Cynomolgus monkeys, radioactivity accumulated in the basal ganglia. The measured striatum to cerebellum ratio was about 15 after 3 h. A monkey brain phantom was constructed for the determination of conversion factors from pixel events to actual radioactivity. The resulting, corrected striatum to cerebellum ratio obtained was 30. After administration of 12 mg raclopride to one of the monkeys there was a substantial decrease in striatal radioactivity. [125I]NCQ 298 is a suitable ligand for the labelling of dopamine D2 receptors in vitro and in vivo. The specific properties of [123I]NCQ 298 suggest that this compound is a useful ligand for quantitative SPECT studies of dopamine D2 receptors in man.
