    Notes: The correlation function presented by Debye et al. for characterizing the random two-phase structure in solids is further extended by considering an effect of partial ordering to describe the statistical features in phase separation structures of polymer blends. The new correlation function is in the form γ(r)=exp(−r/a0 )cos(q0 r) with two parameters. The scattering intensity distribution calculated from this function can predict a maximum whose sharpness and position are characterized by a0 and q0 . Good agreement was achieved between the calculated intensity and measured small-angle light scattering curve from phase-separated poly(methyl methacrylate)/poly(vinyl acetate) blends.

    Notes: The effect of regeneration of Lolium perenne and Festuca rubra from embryogenic suspension cells and protoplasts on fertility and growth was evaluated. Embryogenic suspension cultures were either routinely subcultured or cryopreserved and re-established. Phenology, morphology and fertility of regenerated plants were studied for two growing seasons in a replicated field experiment. Most regenerated L. perenne and F. rubra plants showed a delay in inflorescence emergence, a reduced seed yield and differences in morphological traits when compared with seed-grown plants. For L. perenne, performance of plants regenerated from cryopreserved suspension cultures and protoplasts was similar to that of respective plants regenerated from routinely maintained suspension cultures. However, differences in performance were observed for respective regenerants in F. rubra. The phenotypic deviation observed was partly reflected in the randomly amplified polymorphic DNA (RAPD) analysis performed. However, regenerants of both species showing similar, or even superior performance to the seed-grown plants were also found. Embryogenic suspension cells and corresponding protoplasts of L. perenne and F. rubra have the potential for producing fertile, well-performing plants which can be integrated in breeding programs.

Parameters for the bombardment of embryogenic suspension cultures with the particle inflow gun were partially optimized using transient expression assays of a chimericβ-glucuronidase (gusA) gene driven by the maizeUbi1 promoter. Stably transformed clones were recovered with a selection scheme using hygromycin in liquid medium followed by a plate selection. Plants were regenerated from 33% of the hygromycin-resistant calli. The transgenic nature of the regenerated plants was demonstrated by Southern hybridization analysis. Expression of the transgene in transformed adult Italian ryegrass plants was confirmed by northern analysis and a hygromycin phosphotransferase enzyme assay.

    Notes: Abstract  Two sets of plants (Lb and Lc), regenerated from different single-genotype-derived embryogenic suspension cultures of Lolium perenne cv Citadel, were evaluated for agronomic traits in a modified polycross design in the field. Seed from the primary regenerated plants was harvested to evaluate morphological and phenological traits of corresponding progenies in a replicated field experiment. When compared to seed-grown plants of the same cultivar, primary regenerants of the Lb set showed a significant delay in ear emergence and a more-erect growth habit, while primary regenerants from the Lc set showed a significantly higher seed yield. However, progenies of regenerated plants did not differ from those of seed-grown plants. Embryogenic suspension cells of L. perenne have the potential for producing fertile, well-performing, material which can be integrated into breeding programs.

    Notes: Summary We report the construction of an RFLP genetic map of rice (Oryza sativa) chromosomes. The map is comprised of 135 loci corresponding to clones selected from a PstI genomic library. This molecular map covers 1,389 cM of the rice genome and exceeds the current classical maps by more than 20%. The map was generated from F2 segregation data (50 individuals) from a cross between an indica and javanica rice cultivar. Primary trisomics were used to assign linkage groups to each of the 12 rice chromosomes. Seventy-eight percent of the clones assayed revealed RFLPs between the two parental cultivars, indicating that rice contains a significant amount of RFLP variation. Strong correlations between size of hybridizing restriction fragments and level of polymorphism indicate that a significant proportion of the RFLPs in rice are generated by insertions/delections. This conclusion is supported by the occurrence of null alleles for some clones (presumably created by insertion or deletion events). One clone, RG229, hybridized to sequences in both the indica and javanica genomes, which have apparently transposed since the divergence of the two cultivars from their last common ancestor, providing evidence for sequence movement in rice. As a by product of this mapping project, we have discovered that rice DNA is less C-methylated than tomato or maize DNA. Our results also suggest the notion that a large fraction of the rice genome (approximately 50%) is single copy.

    Notes: In this work we present the results from studies on novel poly(arylene ether ketone)s, including gas permeability, wide-angle x-ray diffraction (WAXD), and dynamic mechanical analysis (DMA). Poly(arylene ether ketone)s containing 2,2′- and 3,3′-dibenzoylbiphenyl (DBBP) moieties were characterized to study the effect of biphenyl substitution on gas transport properties. Gas permeabilities of naphthalene-containing poly(arylene ether ketone)s were also measured. Higher permeabilities were observed for polymers prepared with 6F-BPA, compared to 9,9-bis(4-hydroxyphenyl)fluorene (HPF). The naphthalene-containing polymers exhibited higher permeabilities than the DBBP polymers, except for a polymer having the 2,2′-DBBP and tetramethylbiphenyl moieties. Based on our work, and results reported in the literature, the 3,3′-DBBP polymers showed the lowest permeabilities for DBBP-containing poly-(arylene ether ketone)s. The low permeabilities are due to more efficiently packed chains brought on by greater flexibility of the backbone, compared to the other polymers studied. DMA studies confirmed the higher barriers to rotation which are believed to be responsible for 2,2′-DBBP polymers having similar selectivities compared to 3,3′-DBBP polymers. © 1998 John Wiley & Sons, Inc.
