    Notes: In a rotating He II-filled cylinder with an aspect ratio of 4.3 the transient evolution of the vorticity fields of the normal fluid and the superfluid component during spin-up are investigated. The acoustical method of measurement used utilizes the change of propagation time of first- and second-sound signals irradiated through a slit in the middle of the cylinder. The results of the first-sound measurements within the range 1.3 ≦ T ≦ 2.1 K essentially do not depend on temperature and, for the final state, indicate a lack of about 30% of vorticity compared to solid-body rotation. On the other hand, the results of the second-sound measurements are temperature-dependent and show that the superfluid component reaches solid-body rotation, while the normal fluid fraction seems to slip at the wall. Accordingly, the velocity in the inner part of the flow field adopts a value lower than one would expect from the present velocity of the wall, thus confirming the lack of vorticity observed.

    Notes: Summary Cell lines established from advanced mammary and ovarian carcinomas were assayed for the inhibition of in vitro proliferation by various antineoplastic drugs. The assays were performed with multiple experimental cultures derived from stock cultures of the tumor cell lines in early passages of the cultivation. As determined by comparison of the 50% inhibition of in vitro growth, differential sensitivity of the individual cell lines was observed. Based on the 2-h plasma level of the drugs as discriminatory threshold between resistance and sensitivity, the in vitro effectiveness of each, drug on the individual cell lines was compared with the clinical results of chemotherapy applied to the corresponding patients. In total, positive in vitro/in vivo correlations were observed in 39 of 42 cases. The 17 cell lines evaluated retrospectively were resistant to those drugs which had been tried unsuccessfully during chemotherapy. Among the 25 cases tried prospectively 11 cases showed sensitivity in vitro and in vivo, and furthermore 11 prospective cases were resistant in vitro and in vivo.

    Notes: Abstract In Drosophila as in many organisms β tubulins are encoded by a gene family. We have determined the complete nucleotide sequences coding for the β1 and β2 tubulins of Drosophila melanogaster and the β2 tubulin of D. hydei, and found these insect β tubulins to be highly conserved and like β tubulins of other organisms. This is discussed with reference to the possible functional domains of these proteins. — The β1 tubulin gene of Drosophila is constitutively expressed, whereas the β2 tubulin is expressed specifically in the testes. In D. melanogaster the amino acid sequences of these proteins are 95% homologous, differing at only 25 positions. In the testes the β2 tubulin participates in different microtubules as shown by genetic analysis (Kemphues et al. 1982). Interestingly, all of the amino acids characteristic of the testis-specific β2 tubulin are also present in the corresponding gene of D. hydei. Of special interest is the high degree of conservation of the carboxy-terminal domain in these functionally equivalent β tubulins.

    Notes: Summary Procedures for the in vitro determination of the drug-induced inhibition of mammary and ovarian carcinoma cell growth were established. In monolayer cultures derived from advanced tumors, separation of epithelial carcinoma cells from concomitant cells of fibroblast-like or mesothelial appearance was achieved by differential trypsinization. The carcinoma cell character of the stock cultures was verified by chromosome analyses showing a high degree of aneuploidy for the epitheloid cell lines and euploidy for cells of apparently mesenchymal origin. When cultured carcinoma cells were injected in nu/nu mice, the tissue and cell cultures obtained from the heterotransplantation tumors closely resembled the original tumors and cell cultures in morphology, karyotype, and expression of tumor markers. The action of carcinostatic drugs in the logarithmic phase of the carcinoma cell proliferation was tested by kinetics experiments in multiple experimental cultures. In cell proliferation assays based on cell counts the 50% inhibition dose (ID50) of the drug effects was determined from the dose-response curves. Comparison of the ID50s revealed highly differential effectiveness of the drugs examined. The inhibitory effects were reproducible, rendering the procedures used suitable for testing the chemosensitivity of newly explanted gynecological carcinoma cells by proliferation assays.

    Notes: Zusammenfassung Lediglich bei 3/37 Pat. mit Ovarialtumoren fand sich ein pathologisch erhöhter Serum-CEA-Spiegel. In der Peritoneal- bzw. Zystenflüssigkeit konnten dagegen in 14/26 Fällen erhöhte Werte sowohl bei malignen als auch bei benignen Tumoren festgestellt werden. Eine negative serologische CEA-Bestimmung bedeutet daher nicht, daß kein CEA produziert wird. Die Bestimmung des CEA-Gradienten im Serum und in der Peritonealflüssigkeit dürfte eine größere Relevanz zur Verlaufskontrolle bei Ovarialkarzinomen haben.

    Notes: Zusammenfassung Während der Dauertherapie mit Suprefact konnte ein stetiger Abfall von LH, Östradiol und Progesteron im Serum mit einem unteren Grenzwert von LH ab dem 5. Tag und der ovariellen Hormone ab der 3. Woche nach Therapiebeginn festgestellt werden. Die induzierte hypogonadotrope Ovarialinsuffizienz ist reversibel. Die Therapie mit LH-RH-Analoga ist somit eine gute Alternative bei prämenopausalen Patientinnen mit rezeptorpositivem Mammakarzinom zur irreversiblen Ovarektomie, da bekanntlich 50–60% der Patientinnen auf eine endokrine Therapiemaßnahme nicht ansprechen.

In dem von uns untersuchten Kollektiv konnten wir keine Korrelation zwischen der CEA-Konzentration im Tumorcytosol und dem Gehalt an Östradiol- und Progesteronrezeptoren nachweisen. ob sich daraus der Schluß ziehen läßt, daß die CEA-Expression von der Intaktheit der Proteinbiosynthesekette unabhängig ist, muß anhand weiterer Untersuchungen überprüft werden. 2. Pathologische Serum-CEA-Spiegel zeigen sich auch in fortgeschrittenen Erkrankungsstadien nur bei Fällen mit erhöhter CEA-Konzentration im Tumorcytosol. Demnach scheint die Validität von CEA-Serumbestimmungen in der Verlaufskontrolle von Tumoren mit geringer CEA-Expression sehr zweifelhaft. 3. In über 40% der Fälle mit erhöhten CEA-Werten im Tumorcytosol fanden sich niedrige CEA-Serumwerte. Neben geringem Tumorvolumen könnten hier unterschiedliche Diffusionsgradienten, Membraneffekte oder verschiedenartige Vaskularisierungsmuster eine Rolle spielen.
