    Notes: Summary Rats were injected either with synthetic125I-Arg 101-Tyr 126 atrial natriuretic factor (ANF) or with125I-ANF together with an excess of cold Arg 101-Tyr 126 ANF. Binding sites in various tissues were accepted depending on two criteria: displacement of radioactivity by cold ANF and absence of localization of silver grains on putative target cells in the presence of cold ANF. Binding sites were localized on zona glomerulosa cells and on adrenergic and noradrenergic cells of adrenal medulla, on hepatocytes, on the base of mature epithelial cells of villi in the small intestine, on smooth muscle cells of the muscularis layer of the colon and on the base of epithelial cells of the ciliary bodies. In addition, binding sites were localized in the vasculature of kidney, adrenal cortex, lung and liver. Binding sites were particularly numerous on renal glomerular endothelial cells. These results indicate that ANF may have important hemodynamic effects in kidney, lung, liver and adrenal cortex, may regulate water and ion transport in small intestine and ciliary bodies and may have metabolic effects in the liver. The presence of binding sites on the zona glomerulosa is in agreement with the important inhibitory effect of the peptide on aldosterone secretion.

    Notes: Summary A highly conserved polypeptide termed “7B2”, isolated from human and porcine pituitaries, has been reported by immunoreactivity to be distributed in various organs. However, the highest concentration has been found in the pituitary as demonstrated by a specific radioimmunoassay. In order to determine the type of cells within the pituitary that contain 7B2 and to analyse its intracellular localization, specific immunocytochemistry techniques (unlabeled antibody, peroxidase-antiperoxidase) were used both for light and electron microscopy. Immunocytochemistry of both expiants and monolayer-cell cultures of the adenohypophysis was studied. Immunoreactivity to 7B2 has been found in 21.9% of the total number of cells. After simultaneous staining of serial sections with appropriate antibodies, 7B2 was found to be colocalized with β-LH/β-FSH in gonadotrophs and with β-TSH in thyrotrophs. In situ immunocytochemistry at the electron-microscopic level showed that immunoreactive 7B2 is compartmentalized within secretory granules. The small (130 to 250 nm) but not the large granules (400 to 700 nm) were labeled in gonadotroph-like cells and small granules (90 to 150 nm) were also labeled in thyrothrophlike cells. Study of the gonadotrophs in cell culture after Zamboni's fixation revealed weak to moderate immunoreaction in rough endoplasmic reticulum. The current findings as well as previous data indicate that 7B2 is synthesized, stored and possibly released from the adenohypophysis similarly to many other secretory products.

    Notes: Summary Immunoreactivity to the polypeptide designated “7B2” recently isolated from human and porcine pituitary glands, appears to be consistently confined to neuroendocrine and endocrine cells in various tissues. In rat gut, immunoreactive 7B2 was found in endocrine-paracrine cells. Highly labeled cells were found in the antrum of the stomach and, cells with lower concentrations, in the fundus, duodenum, jejunum and ileum. Except for a few cells which were simultaneously positive for 5-hydroxytryptamine, and a few others showing Grimelius's reaction, “7B2” cells do not exhibit argentaffin and/or argyrophil character. The 7B2 polypeptide seems to be distributed amongst several different types of endocrine cells in the gut.

    Notes: Summary A complex network of atrial natriuretic factor-producing cells has been delineated by biochemical and morphological techniques in the rat ventricular myocardium. The chordae tendineae spuriae (CTS; false tendons) contain ANF mRNA and the ANF propeptide (Asn 1-Tyr 126) as assessed by Northern blot analysis, high-pressure liquid chromatography and immunohisto- and -cytochemistry, using three different affinity-purified antibodies: monoclonal and polyclonal antibodies against C-terminal ANF (Arg 101-Tyr 126) and polyclonal antibodies against N-terminal ANF (Asp 11-Ala 37). Two types of cells harboring ANF-containing secretory granules constitute the CTS: the majority (Purkinje type I) have ultrastructural similarities with both atrial and classical Purkinje fibers. Purkinje type-II fibers resemble working ventricular cardiocytes. Both cell types harbor a large paranuclear Golgi complex. The subendocardial Purkinje network is also made up of these two cell types. In this location, Purkinje type-I fibers form cable-like structures while Purkinje type-II fibers are either located beneath the former or abut directly on the endocardium. The latter are not separated from adjacent working ventricular cardiocytes by connective tissue septa. Coronary arteries and arterioles, as in birds, are surrounded by a cushion of Purkinje type-II fibers which blend with the surrounding myocardium. These results indicate that, in the rat, the entire intraventricular conduction system is constituted of endocrine cells producing ANF.
