    Notes: Summary We have investigated whether the previously demonstrated stimulatory actions of growth hormone on DNA synthesis and (pro)insulin biosynthesis and release of isolated adult rat islets of Langerhans are mediated by an autocrine release of somatomedin-C/insulin-like growth factor I (SM-C/IGF I). In medium containing 1% fetal calf serum, the presence of 16.7 mmol/l glucose, or 2.7 mmol/l glucose supplemented with a concentrate of essential amino acids, caused a significant increase in 3H-thymidine incorporation and insulin release compared to 2.7 mmol/l glucose alone but no increase in SM-C/IGFI release. Further supplementation with 1 μg/ml growth hormone increased 3H-thymidine incorporation and SM-C/IGF I release within all groups, and insulin release in the 16.7 mmol/l glucose and 2.7 mmol/l plus amino acid groups. The ability of growth hormone to increase 3H-thymidine incorporation in the presence of 16.7 mmol/l glucose, but not its action on insulin release, was partly inhibited by a monoclonal antibody against SM-C/IGF I (control cultures 100%; growth hormone alone 261±27%, mean±SEM; growth hormone+anti-SM-C/IGFI 179±21%; p〈0.05, n=18). Growth hormone, but not 100 ng/ml SM-C/IGF I, increased insulin biosynthesis assessed as immunoprecipitable 3H-labelled insulin by 45%, but this was accompanied by a similar increase in overall protein synthesis. Similarly growth hormone, but not SM-C/IGF I caused a 75% increase in glucose oxidation by islets. Both growth hormone and SM-C/IGF I failed to increase the cellular uptake of α-aminoisobutyric acid or 3-O-methyl glucose over a 90 min period. The results suggest that while the stimulatory effect of growth hormone on islet cell insulin biosynthesis and release, glucose oxidation and general protein synthesis is probably direct, its action on B-cell replication is partly mediated by a paracrine release of SM-C/IGF I. This may provide a mechanism for increasing B-cell mass and consequently total insulin output during times of increased metabolic demands on insulin secretion.

    Notes: Abstract The evidence reviewed here shows that the endocrinology of fetal growth is very different from that operating postnatally. Pituitary hormones play little part in stimulating growth of the lean body mass or skeleton although growth hormone (GH) may be involved, in some as yet ill defined way in the ontogeny of the fetal pancreatic islet and insulin secretion. Insulin is important because it stimulates fetal cellular anabolism but acts in a permissive manner: with too little insulin growth is inhibited, with too much growth proceeds at a genetically predetermined rate. Placental lactogen (PL), or other peptides within the GH/PL family, may act as a true growth-promoting hormone in the fetus; it stimulates both cellular metabolism and mitosis. The part played by endocrine control mechanisms in the fetus is set in context by an appreciation of the importance of locally acting tissue growth factors, and in particular the somatomedins. Their part in fetal growth control is intimately bound up with the plane of nutrition experienced by the fetus. It is concluded that the simplest analysis that makes biological sense involves a consideration of hormones, tissue growth factors and nutrition, not hierarchically but as mutually interacting variables.

    Notes: Summary Due to its capability of producing a well localized intense heat field of predictable dimensions, the electromagnetic field focusing probe was evaluated experimentally as a surgical tool in aneurysm thrombosis. Aneurysm models were created by anastomosing a segment of vein to the abdominal aorta. Seventy-five such aneurysms were created in seventy-five animals. The aneurysms were then placed under the solenoidal radiofrequency coil and the tip of the field focusing probe was inserted into them. Intense heat was delivered with well defined, effective and quick thrombosis and shrinkage of the aneurysm. Nineteen of the aneurysms were followed for up to one week. Histological studies were conducted showing complete thrombosis of the aneurysm with good preservation of the aorta. With further refinement of this technique, it is hoped that stereotaxic aneurysm thrombosis might be possible.

    Notes: Summary Vaporization of atheromas in 10 human aorta segments immersed under flowing blood in vitro were done using the EFF probe. No increase in packed cells of the flowing blood following atheroma vaporization were noted suggesting absence of post vaporization debris. No damage to the aorta underlying the vaporized atheroma was noted on light microscopy suggesting well localized vaporization process. This study suggests that EFF probe might be a safe tool for intravascular endarterectomy.

    Notes: Summary Autologous adrenal cortex was completely dissected off the medula using microdissection. The adrenal medulla tissue was transplanted to the lateral ventricle in twenty-one adult rats. Group A consisting of ten animals received 200 units of HCG per 100 gram body weight daily for one week and then on every alternate day for seven weeks. The remaining rats (group B) served as control. After eight weeks the brains were removed and a volumetric study was done using histological sections. High pressure liquid chromatography (HPLC) with electrochemical detection for catecholamine assay of the graft was also undertaken. The graft survival in group A was significantly higher (p〈0.005) than group B. The level of norepinephrine was significantly higher in group A while dopamine was higher in group B. The higher volume of graft tissue in group A suggests that HCG may help in increased survival and growth of the transplanted tissue. Higher levels of norepinephrine in group A would suggest a tendency for such grafts to be active to a greater extent with the treatment of HCG.

    Notes: Summary The electromagnetic field focusing (EFF) probe is capable of producing well circumscribed, intense heat at the point of contact with the tissue. Experimental studies were carried out to assess this probe as a neurosurgical tool using 38 rats and 4 mice with mammary carcinoma. The study on the rats included study of the cutting, coagulating and vaporizing effect on brain tissue including study of blood brain barrier disruption and heat dissipation. The study on the mice included the study of vaporizing property of the probe on solid tumours. The probe proved to be an excellent tool for cutting, coagulating and vaporizing purposes with very minimal disruption of blood brain barrier and demonstrated well circumscribed heating pattern. The study indicates that this tool combines the beneficial effect of both the YAG and CO2 laser.

    Notes: Several diagnostics measure the particle sources and losses in the Tandem Mirror Experiment-Upgrade (TMX-U) plasma. An absolutely calibrated high-speed (0.5 ms per frame) filtered (6561 A(ring)) video camera measures the total ionization source as a function of radius. An axial view of the plasma automatically integrates the axial variations within the depth of field of the system. Another camera, viewing the plasma radially, measures the axial source variations near the deuterium fueling source. Axial ion losses are measured by an array of Faraday cups that are equipped with grids for repelling electrons and are mounted at each end of the experiment. Unequal ion and electron (nonambipolar) radial losses are inferred from net current measurements on an array of grounded plates at each end. Any differences between the measured particle losses and sources may be attributed to ambipolar radial losses and/or azimuthal asymmetries in the particle-loss profiles. Methods of system calibration, along with details of computer data acquisition and processing of this relatively large set of data, are also presented.

    Notes: This study examined the effects of academic stress upon components of Type A behavior and cardiovascular reactivity. In a longitudinal design, heart rate and blood pressure responses to the Type A Structured Interview (SI) and additional laboratory challenges were measured in medical students at three points during an academic semester. Sessions 1 and 3 were scheduled during vacation periods; Session 2 took place during an intensive examination week. Results indicated that three “stylistic” components of Pattern A derived from the SI—loud and explosive speech, short response latency, and potential for hostility—increased significantly during the exam period. Similar increases achieved marginal significance for another stylistic component, rapid and accelerated speech. By contrast, global Type A assessments did not increase with academic demands. Cardiovascular reactivity was significantly but only moderately stable across the three sessions, showing less consistency than has been reported in previous research. In addition, there was little evidence in this study that cardiovascular reactivity was potentiated by academic stress or by Type A behavior. These results demonstrate the influence of naturalistic environmental factors on overt behavioral components that have recently been suggested as coronary risk factors. Therefore, it is advisable for future experimental and epidemiological research to examine the social and environmental context in which these behaviors occur.

    Notes: The power flow to the DIII-D divertor targets is routinely measured using infrared (IR) thermography. An IR television camera sensitive to radiation in the 8–12-μm range views the divertor region using a set of germanium optics. Digital signal processing is used to extract the desired surface temperature profiles from the analog data ((approximately-equal-to)12 Mbytes) stored on videotape. Inversion of these data using a simple matrix formulation of the inverse heat conduction problem then yields the incident heat flux as a function of space and time. Results from a DIII-D discharge are included.
