Notes: Abstract Females heterozygous for the X-linked urea cycle disorder, ornithine transcarbamylase (OTC) deficiency have a significant risk of developing hyperammonaemia. Diagnosis of this genetic defect in a proband is the essential starting point for family studies. By an immunohistochemical analysis of the liver specimens fixed in 10% formalin, we confirmed heterozygous status for OTC deficiency in two female patients, a 15-year-old girl and a 2-year-old girl, who died of hyperammonaemia. Since most affected males lack cross reactive materials (CRM), an immunochemical analysis should be useful for the diagnosis of most heterozygous females.

Notes: Abstract Experiments were performed to study the effect of air fluidization velocity, particle diameter, tube diameter, and pitch between tubes on heat-transfer coefficient for a bundle of horizontal tubes immersed in an aggregative fluidized bed. Not only horizontal but also vertical distributions of the heat-transfer coefficients within the bundle were also extensively determined. The heat-transfer coefficient and its maximum value were found to be dependent on the particle diameter, the air fluidization velocity, and the gap between the tubes in the bundle. The proposed correlation for the maximum heat-transfer coefficient was in good agreement with the present results.

Notes: Abstract During the last 2 years, eight patients with hepatocellular carcinoma who were suspected of having distant metastases have been studied to determine whether a new biliary agent, 99mTc-Sn-N-pyridoxyl-5-methyltryptophan (99mTc-PMT), is taken up by extrahepatic tumors. In all eight patients, scintigrams showed a clearly increased uptake of 99mTc-PMT radioactivity by the extrahepatic tumors. In contrast, an increased uptake by the tumors of gallium citrate Ga 67 was only detected in four of the seven patients examined. The results obtained in this study suggest that 99mTc-PMT is useful both for characterizing the nature of extrahepatic tumors in patients with hepatoma and for detecting the metastases.

Notes: Abstract Images were obtained both with a biliary agent, 99mTc-Sn-N-pyridoxyl-5-methyltryptophan (99mTc-PMT), and with 67Ga-citrate in 40 cases of hepatocellular carcinoma and in 43 cases of other hepatic diseases and results were compared. Positive results were obtained by delayed 99mTc-PMT imaging in 25 (63%) of 40 cases of hepatoma: the hepatic tumor showed increased uptake in 18 cases (45%) and equilibrated uptake in 7 cases (18%). Positive 67Ga-citrate imaging was found in 28 (70%) of the 40 cases of hepatoma: increased uptake was seen in 24 cases (60%) and equilibrated uptake in 4 cases (10%). Of 15 cases of hepatoma giving negative results in the 99mTc-PMT study, 7 cases (47%) took up 67Ga-citrate, and 6 of these showed increased 67Ga-citrate uptake by the hepatic tumors. A certain relation existed between the degree of histological differentiation of hepatomas and the intensity of 99mTc-PMT uptake by those tumors, while such a histological correlation was denied for 67Ga-citrate uptake by the tumors. Delayed 99mTc-PMT imaging is preferable to 67Ga-citrate imaging for increasing the specificity of diagnosis of hepatoma. 67Ga-citrate should be used in those cases that do not give positive results with 99mTc-PMT.

Notes: Abstract We examined tyrosinase activity in pigmented specimens from three cases of ocular malignant melanoma. Tissue (cholate-trypsin-treated) extract was prepared in cholate-phosphate buffer by homogenization, centrifugation, trypsin digestion, and hydroxylapatite column chromatography. Tyrosinase activity was spectrophotometrically assayed as dopa (L-3,4-dihydroxylphenylalanine) oxidase activity. Tyrosinase activity was detected in the cholate-trypsin-treated extracts. Enzyme activity was inhibited by phenylthiourea but not by 3-iodo-tyrosine. The enzyme was inactivated when extract was preheated or digested with pronase. We believe that our findings confirm the presence of tyrosinase activity in ocular malignant melanoma.

Notes: Abstract Intramolecular and liquid structure of 2,2,2-trifluoroethanol (TFE) have been investigated by x-ray diffraction at 25°C. The structural parameters for the skeleton of the molecules in the liquid phase are similar to those in the gas phase. The conformers of TFE molecules in the liquid phase are discussed. The O...O distance at about 284 pm and additional F...O one at about 302 pm were found to be characteristic for the first neighbor interactions. Various models (dimers and trimers) have been examined for analyzing the first neighbor structure. The liquid structure was explained in terms of small clusters consisting of two to three molecules rather than of a more extended polymeric network.
