    Notes: Maize seedlings (Zea mays L. cv. DK 246) grown for 1–4 days in the presence of abscisic acid (ABA) or triadimefon (a fungicide) demonstrated an enhanced ability to withstand the effects of a 3-h sub-lethal (40°C) or lethal (45°C) heat shock. Both the ABA and triadimefon treatments were applied solely to the roots of seedlings; however, the ability to withstand a heat shock was induced in both the root and the shoot. The level of protection provided by these agents was dependent upon the time that plants were exposed to them; prolonged exposure reduced tolerance to subsequent stress.

    Notes: Summary Cell-free protein synthesis systems were prepared from normally-grown (N-lysate) and heat-shocked (HS-lysate) Neurospora crassa mycelium. Although both lysates translated homologous mRNA, the HS-lysate was more active, yielding a higher incorporation of [35S]-methionine into hot TCA-insoluble material and a vastly superior protein synthesis profile. The optimal temperature for translation by both lysates was 21 °C; the HS-lysate did not translate heat-shock mRNA preferentially at any temperature tested. Fortuitously, heterologous messenger RNAs from diverse eukaryotic and viral sources — Drosophila, dog pancreas, rabbit globin mRNA, brome mosaic virus, tobacco mosaic virus — were translated by the HS-lysate with an efficiency comparable to that of the commercial rabbit reticulocyte system and superior to the wheat germ system. The cap analogues, m7G(5′)ppp(5′)G and m7G(5′)ppp(5′)Gm, inhibited translation significantly.

    Notes: Abstract InNeurospora crassa, there is a single pyruvate kinase (PK) consisting of four identical subunits of ∼60k daltons. Northern and dot blot hybridization studies, using most of the yeast pyruvate kinase gene as a probe, suggest the presence of two distinct mRNA species for pyruvate kinase, separable on the basis of the length of their polyadenylated tails, by oligo(dT)cellulose chromatography. These messages are present in polysomes, immuno-precipitated by anti-PK antibodies, indicating probable translation in vivo. Fractions containing both messages were translatedin vitro in the heterologous systems as well as in a homologousN. crassa lysate, the newly-synthesized PK being detected by immunoadsorption. Protection studies using S1-nuclease suggest no major structural differences in the 5′-untranslated and most of the coding regions of the two messages.

    Notes: Abstract The role of microstructure in modifying the tensile strength of rheocast Al-10 wt % Cu and Al-4.5 wt % Cu is investigated by modelling these products as particulate composites. The tensile strength of rheocast AI-10 wt %Cu alloy is found to depend on experimentally determinable parameter $$\bar D\bar Z$$ which is an undefined function of the size distribution of proeutectic α-particles. These products, however, possess lower strength than the conventional casting at equal porosity level. The tensile strength of the rheocast AI-4.5wt%Cu alloy shows an inverse square root dependence on the proeutectic α-particle size, $$\bar D$$ , and therefore appropriate rheocasting process parameters may be employed to bring about grain refinement in order to produce rheocastings possessing higher strength than the conventional casting at equal porosity level.
