Notes: Summary To elucidate the localization of the subunit C of aldolase (aldolase C) in peripheral neuroendocrine cells, we made an immunohistochemical study with monospecific antibodies against human aldolase C. Aldolase C was found to be localized in various types of neuroendocrine cells; in the pituitary gland, thyroid, pancreas, adrenal gland, bronchus, and gastrointestinal tract.

Notes: Summary The effect of dibutyryladenosine 3′,5′-cyclic monophosphate (dibutyryl cAMP) on the expression of glutathione S-transferase placental type (GST-P) was examined in rat glioma cell line using an immunohistochemical technique. Cultured T9 glioma cells were negative for GST-P activity under normal conditions. However, treatment with 1 mM dibutyryl cAMP produced GST-P expression in about 50% of the cells, as well as some morphological changes. The expression of GST-P was increased with addition of dibutyryl cAMP together with 1 μg/ml allyl isothiocyanate (AITC) or 0.1 μg/ml benzyl isothiocyanate (BITC). With these combinated treatments, almost all cultured cells showed a strong positive reaction for GST-P, although AITC or BITC alone elicited GST-P in only 5% of the cultured cells. The results of the present study indicate that dibutyryl cAMP causes functional as well as morphological differentiation of T9 glioma cells.

Notes: Summary We have demonstrated the histochemical fiber types of human skeletal muscle in paraffin sections by the immunohistochemical method, together with compatible observations previously made in frozen sections that carbonic anhydrase III is mainly localized in type 1 fibers (Shima et al. 1983), and muscle-specific enolase in type 2 fibers (Ibi et al. 1983). This method is useful to analyze the fiber types when frozen muscle samples at biopsy or autopsy cannot be obtained.

Notes: Immunohistochemical distribution of human epidermal growth factor (hEGF) was described in 17 cases of mixed tumour of the skin with monoclonal antibody. In normal sweat glands, epithelial cells in the secretory portion and in the transitional area between secretory portion and duct showed prominent staining for hEGF. In the salivary pleomorphic adenoma type of mixed tumour of the skin, luminal tumour cells of tubular and duct-like structures gave a very characteristic hEGF staining reaction. The tumour cells showing strong staining for hEGF were scattered throughout the solid foci in this type of mixed tumour. Tubular epithelial cells in the clear cell adenoma type also displayed a positive hEGF reaction. And apocrine mixed tumours strong staining for hEGF occurred on the apical side of tubular and ductal tumour cells. In view of the immunohistochemical staining patterns for hEGF, the histologic origin of mixed tumours of the skin is suggested to be cells in the secretory portion and (hose in the transitional portion between secretory portion and duct of the sweat gland.

Notes: We observed the presence of the novel pituitary protein „7B2” and its release in the bovine adrenal medulla. The 7B2 concentration (mean ± SEM) in extracts of the bovine adrenal medulla was 952 ± 155 pg/mg tissue (n = 6). 7B2 was distributed in the chromaffin granule fraction prepared from the bovine adrenal medulla and was released by high K+ and/or nicotine from cultured cells of the bovine adrenal medulla. Co-release of 7B2 with catecholamine induced by nicotine from the cultured bovine chromaffin cells was also observed. In an analysis of the bovine adrenal medulla chromaffin granule fraction on gel permeation chromatography, there was a major peak with an apparent molecular weight of 45,000, whereas a major peak with an apparent molecular weight of 20,000 was found in that on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. On reverse-phase HPLC, a major peak with a retention time of 35 min was observed in the bovine chromaffin granule fraction and in the bovine anterior pituitary extract. These findings indicate that 7B2 is a secretory protein in the bovine adrenal medulla. The possibility that 7B2 might be released with catecholamine, possibly in response to stress, warrants investigation.

Notes: Intense relativistic electron beams fired into air at varying pressures display a wide range of microwave signatures. These experiments held beam current, energy, and pulse length constant while varying gas pressure. Our observing window is 10 to 40 GHz. At low pressures (〈10 mTorr) exponential spectra result, consistent with beam reflexing or virtual cathode oscillations. Above 20 mTorr the spectrum flattens and suggests collective emission at the beam-generated plasma frequencies. Power falls linearly with pressure above 20 mTorr, until electron-Neutral collisions damp the emission at a few Torr. However, weak 10 GHz emission appears at full atmospheric pressure.

Notes: Electron cyclotron emission from nonthermal electron distributions has been measured with a vertical view and control of multiple reflections. The observed intensities at the first few harmonics provide information that can be used to deduce the electron distribution function. Although harmonic overlap restricts the detail that can be obtained when the distribution is very energetic, a fitting procedure enables useful results to be obtained. The distribution functions during Ohmic runaway and lower-hybrid current drive and heating are presented. The current-drive distributions are not inconsistent with theoretical Fokker–Planck calculations, giving perpendicular and parallel "temperatures'' around 60 keV and 200 keV, respectively. The runaway cases have quite similar distributions.

Notes: Electron cyclotron emission measured vertically through the center of a tokamak plasma yields detailed information about the electron velocity distribution. A diagnostic developed for this purpose on Alcator C tokamak uses specialized focusing optics to obtain a well-collimated viewing chord, a compact viewing dump made of Pyrex or Macor to reduce the effects of wall reflection and depolarization, and a rapid-scan polarizing Michelson interferometer InSb detector system for the spectrum measurement; all constrained by the limited access and the compact size of Alcator C. Results of diffraction analysis are used to evaluate the theoretical performance of the optical system.

Notes: Compact, vacuum-compatible submillimeter viewing dumps, whose design parameters were based on the results of simple ray calculations, have been fabricated from Pyrex, Macor, and alumina. The measured performance of the dumps agrees qualitatively with calculated results. The absorption is somewhat lower than expected, but still acceptable (≥99% absorption for Pyrex and Macor). In an electron cyclotron plasma emission experiment on Alcator C tokamak, comparison of the emission spectra with and without the dump shows that the dump in this configuration is effective in suppressing the unwanted radiation by approximately 90%. The additional reflection is believed to be due to a portion of the antenna pattern missing the dump.

Notes: Nonthermal electron distribution functions, arising during lower hybrid heating and current drive, have been measured on Alcator C tokamak using vertical electron cyclotron emission and also bremsstrahlung x-ray emission. The methods used for deconvolving the distribution from the radiation measurements are summarized and examples are given of results from different types of plasma, illustrating the present capabilities of the techniques. The cyclotron emission results are consistent with the x-ray analysis but provide additional information, particularly at lower energies (30–100 keV). The x rays are less ambiguous at high energy and give good forward/backward asymmetry diagnosis. The quality of information appears to be already adequate for a quantitative comparison with theory, but further experimental improvements seem possible.
