    Notes: Capillary Gas Chromatography (CGC) is capable of determining underivatized cyclophosphamide (CPA) using SCOT OV 275 columns. Then CPA is subjected to in situ degradation resulting in formation of a cyclization product which can be determined selectively in biological fluids. In routine bioanalysis however cyclization products of CPA metabolites might interfere, e.g. 4-keto CPA. In the present study possible formation of cyclization products of 4-keto CPA similar to CPA was monitored by Mass Spectrometry. Cyclization of 4-keto CPA in situ was demonstrated to occur, resulting in a product similar to that of CPA. Both cyclization products could be determined selectively and it appeared that in situ cyclization of 4-keto CPA was negligible (〈5%), probably owing to extra stabilization of the CPA metabolite by keto-enol tautomerism as has been demonstrated by NMR.

    Notes: The Golay-Giddings and Poiseuille equations are used to derive equations for the calculation of the maximum plate number and minimum time conditions for given columns at fixed, but selectable, outlet pressures. In addition, expressions are presented for the determination of minimum analysis times for separation problems requiring given plate numbers. In this instance, the optimum column length can be calculated as a function of outlet pressure. A Basic computer program, incorporating the equations for the various optima, together with the H-ū curves, is described. Input variables are either column length or desired plate number, column diameter, film thickness, capacity ratio of the solute, column outlet pressure, seperation temperature, and carrier gas. The carrier gas viscosity is automatically calculated in the case of hydrogen, helium, or nitrogen. For these gases, and if the solute is a n-alkane, the diffusivity of the solute in the mobile phase is calculated. In this case, the carbon number of the solute is needed in the computation.For high molecular weight polydimethylsilicone phases (e.g. SE-30), the program can approximate the diffusivity of n-alkanes in the stationary phase at the given temperature as a function of the carbon number. Of course, manually entered values of viscosity and diffusion coefficients can be included in the calculations.

    Notes: In comparison with conventionally packed HPLC columns, from a theoretical point of view, open capillary liquid chromatography (OTLC) systems offer a number of advantages like high plate numbers and short analysis times. On the other hand, drastic changes have to be made to the instrumentation. In particular, the contribution to band broadening by the chromatographic equipment must be considerably reduced. In the present study an OTLC system was developed and evaluated, which yields satisfactory results for 26 μm i. d. columns. The determination of the contribution of the chromatographic equipment to the total band broadening is discussed.

    Notes: Adult male volunteers were administered orally 10 mg of the espective drug. Urine samples were investigated by micro-column liquid chromatography and capillary gas chromatography combined with on-line mass spectrometry. The former technique was found to be usable for the detection of these drugs. The latter method appeared to be superior in terms of sensitivity and for metabolism studies. The major excretion products of dexamethasone were identified as isomeric 6-hydroxy metabolites. Dexamethasone as such and its 20-hydroxy metabolite were found in minor quantities. Unlike dexamethasone, triamcinolone is excreted largely unchanged from the human body. Two metabolites were identified as 11-keto and 4,5-dihydrotriamcinolone. With the gas chromatography methods developed, abuse of these drugs can be detected up to 24 h after administration of a 5 mg single dose.

    Notes: Previous studies on open tubular column operation at vacuum outlet vs. atmospheric outlet pressures focused on comparisons of given columns, or comparisons of columns with the same inner diameters. It was demonstrated that, for a given separation problem, vacuum outlet operation of columns with a constant i. d. always yields the shortest analysis times (under minimum plate height conditions).In this paper, the comparison of vacuum vs. atmospheric outlet operation is broadened to columns with different dimensions. A general equation for the gain in speed of analysis by vacuum outlet operation of any column, as compared to atmospheric outlet operation of all possible open tubular columns with the same maximum plate number is presented. The resulting equation is further evaluated for thin film columns of different dimensions.It appears that vacuum outlet operation is beneficial only, in terms of speed of analysis, if low maximum plate numbers are required. The gain in speed of analysis is more pronounced for wide-bore than for narrow-bore columns.

    Notes: Identification and detection limits for scanning and non-scanning mass spectrometers are discussed. It is theoretically deduced and experimentally confirmed that these limits are on the low pico-and femtogram levels, respectively, when using conventional secondary electron multiplier-amplifier systems. The sensitivity can be increased at least tenfold by pulse-counting techniques, instead of current amplification, provided the chemical noise is sufficiently low. The potential advantages of a detection system for simultaneous ion detection in a significant mass range, for obtaining complete mass spectra in fast GC/MS analyses, are demonstrated. A double focusing mass spectrometer was constructed, using the well-proven Mattauch-Herzog principles. By application of an “electronic photoplate”, substance identification in the low femtogram range on a millisecond time scale, so far only accessible for single ion monitoring techniques, is feasible.

    Notes: The main principles of solid-phase extraction techniques are reviewed in this paper. Various solid sorbents can be used as a suitable trap for direct accumulation of organic compounds from aqueous solutions. The trapped analytes can be desorbed by elution with suitably chosen liquid phases. These preconcentration procedures can be considered as low performance liquid chromatography and the efficiency of the procedure can thus be related to the retention characteristics of the preconcentration column. The main sorbents used for trace enrichment purposes are also reviewed. Besides, the concise methodology, sample storage, and automation are discussed. The advantages of solid phase extraction as compared to liquid-liquid extraction are given as well as some drawbacks of this method.

    Notes: The selectivity of a capillary gas chromatographic assay for the anti-neoplastic and immunosuppressive agent cyclophosphamide (CPA) towards one of its naturally occurring metabolites, i.e. 4-ketocyclophosphamide (4-keto CPA), has been studied. Mass spectrometry studies showed that a cyclization product of 4-keto-CPA can form using the same chromatographic conditions as those under which CPA was determined. However, the amount produced is relatively small (less than 10%) compared with the percentage produced from CPA. Furthermore, both cyclization products, formed by loss of HCI and intraalkylation of the parent compounds, can be separated well under suitable chromatographic conditions.
