    Notizen: 2 patients with leg ulcers got worst after the application of a compound mixture of sugar and povidone-iodine (sugar/PI compound), Because they had been suffering from stasis dermatitis, symptoms of contact dermatitis were ambiguous. Patch tests showed positive reactions to 10% povidone-iodine in water and 5% potassium iodide in water, with no response to sugar. They were also tested with sugar/PI compound, containing 3% povidone-iodine, resulting in another positive reaction. They improved after the application of sugar/PI compound was discontinued. Contact dermatitis from topical agents should be considered as more probable than angry back syndrome in cases of leg ulcer.

    Notizen: Abstract The effect of the supernatants obtained from the liquid culture medium ofSporothrix schenkii andCandida albicans on the generation of superoxide anion (O 2 − and hydroxyl radicals OH., the elements of reactive oxygen species (ROS), and chemilimunescence (CL), a measure of several ROS, by polymorphonuclear leukocytes (PMNs) was examined. In our study, it was shown that the supernatant ofS. schenkii increased all types of ROS generation examined and CL, while that ofC. albicans increased OH. generation and CL. The effect of the supernatants ofS. schenkii on OH. generation and CL and that ofC. albicans on CL were most remarkable when the supernatant obtained 8 weeks after the inoculation was used. The supernatant ofS. schenkii was shown to be a much more potent stimulant than the supernatant ofC. albicans. This ROS-stimulating effect of the supernatant ofS. schenkii was heat stable but not dialyzable. These findings suggest the possible role of ROS produced by infiltrated PMNs in the inflammatory skin lesions induced byS. schenkii.

    Notizen: Abstract The scavenging effects of reactive oxygen species (ROS) by two natural health products, antioxidant analogs (AOA), and green magma (GM), and 16 medical Chinese herbs were investigated in two in vitro ROS-generating systems, activated neutrophils and xanthine-xanthine oxidase. Native, unheated AOA and GM products significantly reduced ROS levels, while unheated Chinese herbs had a negligible effect on ROS levels. In contrast, heat-extracted Chinese herbs and AOA markedly, and GM mildly, suppressed the levels of ROS in both systems. The ROS scavenging activity of these native, unheated products was unaffected by dialysis, but that of heated products was markedly diminished by dialysis. Further, the incubation of these products with gastric juice obtained by a gastric tube from healthy volunteers revealed results comparable to those induced by heat treatment with or without dialysis. Although the antioxidant activity of these natural products appears to be partly due to enzymes such as superoxide dismutase (SOD), the predominant factor seems to be low-molecular-weight ROS scavengers that are liberated or activated by gastric juice digestion as observed after heat treatment.

    Notizen: Abstract The effect of several glucocorticosteroids on the generation of reactive oxygen species (ROS) was examined. The ROS assessed were O 2 − , H2O2, OH·, and chemiluminescence (CL) (determined in the presence or absence of luminol), generated by both opsonized zymosan-stimulated neutrophils or monocytes and by the xanthine-xanthine oxidase system. Except for luminol-independent CL, only high concentrations (10−4 M) of steroids could decrease each ROS. In contrast, luminol-independent CL generation in the phagocyte system was increased in a dose-dependent manner by the addition of dexamethasone, but not by any other steroid. Further, in lymphocyte cultures stimulated with Con A for four days, luminol-independent CL generation was demonstrated and enhanced by the addition of dexamethasone, although CL generation was not detected in the absence of dexamethasone. These findings provide evidence that CL does not always represent light specific to ROS, and they suggest the possibility that dexamethasone induces emission of light at sites of inflammation.

    Notizen: Abstract To examine the possible correlation between tissue injury and neutrophil produced active oxygen (AO) species in patients with systemic lupus erythematosus (SLE), we studied the capacity of the serum from six patients with untreated, active SLE to generate AO and release lysosomal enzymes by normal neutrophils. Cultured endothelial cells from human umbilical cord vein were incubated with serum-stimulated neutrophils to assess AO-induced tissue injury. Serum from patients with bacterial infections and healthy individuals served as controls. AO production was highest in the neutrophils stimulated with SLE patient-derived serum, while lysosomal enzyme release was only slightly increased. SLE neutrophils with or without stimulation and SLE serum-stimulated normal neutrophils produced significantly high levels of cytotoxicity upon coincubation with51Cr-labeled human endothelial cells. These excessive cytotoxicities were reversed by the presence of superoxide dismutase and catalase, indicating the specificity of the AO effect on endothelial cell damage. These findings suggest that tissue damage in SLE may be partially due to excessive production of AO and that both neutrophils themselves and a serum factor which activates neutrophils are involved in the mechanism for vascular injury.
