    Notes: The probabilistic theory of the three-phase structure invariants for a pair of isomorphous structures [Hauptman (1982). Acta Cryst. A38, 289-294] is reexamined. The analysis leads to distributions capable of estimating cosine invariants in the full range of -1 to +1. In particular, it is shown that heavy-atom substructure information can be incorporated easily into the distributions. The initial applications, using calculated diffraction data from the protein cytochrome c550, MR ∼ 14 500, and its PtCl2-4 derivative show that a remarkable increase in accuracy results from the use of the revised distributions, particularly after the incorporation of heavy-atom substructure information. Finally, it is shown that in the individual phase determinations the redundant cosine invariants play a role identical to that of the multiple isomorphous derivatives and thus provide the basis for the solution of the phase problem in the single isomorphous replacement case.

    Notes: The theoretical basis for the integration of direct methods into the single-wavelength anomalous dispersion technique is reexamined. The analysis shows that the approximations responsible for the ability to obtain unique estimates of the two- and three-phase structure invariants [Hauptman (1982). Acta Cryst. A38, 632-641; Giacovazzo (1983). Acta Cryst. A39, 585-592] or twofold estimates of the three-phase structure invariants [Kroon, Spek & Krabbendam (1977). Acta Cryst. A33, 382-385] are also responsible for the substantial errors observed in the applications. It is shown that, in the general case, the method of joint probability distributions leads to twofold estimates of the two-phase invariants and eightfold estimates of the three-phase invariants. Finally, it is shown that more accurately determined three-phase invariant estimates can be obtained by the use of anomalous scatterer substructure information, when available, and the use of a strategy that recognizes cases in which the eight estimates are clustered around one or two values. These cases are then distinguished from those where the eight estimates are widely scattered by a weighting function.

    Notes: We show theoretically and experimentally that enhancements of up to 40 dB in the efficiency of optical transmission of microwave signals can be obtained by using a single quantum well laser transmitter whose contact is segmented into two sections, and modulation is applied to one of the sections. The improvement is dependent fundamentally on the relaxation oscillation frequency of the device, with the improvement factor dropping to around 15 dB at relaxation oscillation frequencies approaching 10 GHz.

    Notes: The preparation of high Tc superconducting thin films of Y1Ba2Cu3O7−δ on (100) single crystals of MgO, ZrO2 with 9% Y2O3 (yttria stabilized zirconia, or YSZ), and SrTiO3 using a simple spray deposition technique is described. Typical film growth procedure involves (a) the spraying of a stoichiometric solution of the nitrate precursors on the heated substrate (180 °C), (b) prebaking in air of the sprayed film (20 min at 500 °C), and (c) oven annealing of the film under flowing O2 (900–950 °C followed by slow cooling to 200 °C in about 3 h). X-ray diffraction analysis of the films after each of the growing steps mentioned above shows primarily the presence of crystalline phases of the nitrates, the oxides, and the orthorhombic superconducting phase, respectively. Resistivity versus temperature measurements show that the onset and completion of the superconductive transition occur at 92 and 87 K, respectively, in films on YSZ substrate; at 95 and 80 K, respectively, in films on SrTiO3 substrate; and at 82 and 77 K, respectively, in films on MgO substrate.

    Notes: Summary Mill Door/79 virus, an orbivirus of the Kemerovo serogroup, Great Island Complex, was shown to induce in infected cells 10 segments of double-stranded RNA with a total molecular weight of 11.46×106 daltons. Using methyl mercuric hydroxide as a denaturing agent the double-stranded RNA was translated in a cell-free system producing 11 polypeptides, 10 of which co-migrated with those produced in Mill Door/79 virus infected Vero cells. The segments were separated and individually translated in a cell-free system allowing their coding assignments to be made. These assignments were confirmed by partial proteolysis.

    Notes: Abstract We report a case of nonfatal complete atrio-ventricular (A-V) block after injection of 125 mg of furosemide by a central vena cava catheter. Accidents with this diuretic are very rare but this observation shows that it could induce A-V conduction disturbances. When large quantities of this diuretic must be used, we would recommend a slow injection rate to avoid such accidents.

    Notes: Summary The genome of FT363 virus, a member of the Kemerovo serogroup of orbiviruses, was shown to consist of 10 segments of dsRNA. Examination of the intracellular proteins expressed by this virus in Vero cells showed the presence of 9 major and several minor products. Similar results were obtained by translation of the virus dsRNA in a rabbit reticulocyte lysate. Comparison ofin vivo andin vitro products by immunoprecipitation using polyclonal FT363 ascitic fluid showed a difference in the numbers of polypeptides precipitated. The significance of these results is discussed.

    Notes: Summary Coriolus versicolor, a white-rot Basidiomycete, secretes cellulolytic and ligninolytic enzymes as well as polyphenol oxidase (PPO). Whereas the former degrade wood polymers, the latter can convert diphenols to diquinones and oligomerize syringic acid, a lignin derivative. Certain phenolic compounds can serve as disease-resistance factors controlling the proliferation of wood-decay fungi within host tissues. BecauseC. vesicolor can be ‘batch-cultured’, overproduction and enhanced secretion of enzymes of biological and commercial interests are feasible. Reported here are the results of attempts to define the timed appearances of intracellular and extracellular PPO, to assess substrate specificity as well as distinguish synthesis versus activation of intracellular PPO and to partially purify extracellular PPO. These efforts were to provide data enabling cell-free synthesis of PPO, cloning of the gene(s) for the oxidase and the establishment of its subcellular route of secretion. Whereas two protein peaks (6 and 12 days in a 16 day time-course) were observed for dialyzed mycelial homogenates, the homogenates' PPO specific activity rose between 4 and 12 days and then declined. Total extracellular protein content climbed from 6 to 15 days for dialyzed growth medium and the medium's PPO specific activity rose at 4 days post-inoculation and except at 9 days increased linearly to 15 days. When aliquots of dialyzed 12 and 15 day media were added to PPO assay mixtures containing catechol and either syringic or gallic acids, statistically significant differences in PPO specific activity between phenolic substrates were noted. Supplementation of cultures with 1.91 μg cycloheximide ml growth medium−1 (control, growth medium only) together with 0.5 μCi [14C]-leucine revealed that cycloheximide inhibited PPO activity and suppressed [14C]-leucine incorporation into TCA-insoluble cytoplasmic protein. As for PPO partial purification, growth medium dialysis followed by 0–30% (NH4)2SO4 fractionation and subsequent 12 000×g dialyzate centrifugation yielded a 3.27-fold enhancement in PPO specific activity within the 12 000×g supernatant. Chromatography of the latter upon DEAE-Sephadex indicated that PPO exchanged with the DEAE counterion as it could be eluted with high ionic strength salt. These results suggest that: the occurrences of intracellular and extracellular PPO are time-dependent, intracellular PPO is de novo synthesized, the preferred substrate for extracellular PPO appears to be catechol and extracellular PPO can be partially purified by a combination of dialysis and ammonium sulfate fractionation as well as possibly DEAE chromatography and/or Sephadex G-150 gel filtration.
