Notes: Summary Age-related changes of pyramidal cell basal dendrites in layers III and V of human motor cortex (area 4) were analyzed quantitatively in Golgiimpregnated sections by Sholl's method of concentric cireles (Sholl 1953). The present data suggested that basal dendrites of the pyramidal cells were decreased in number with advancing age, and that the decrease was more prominent in basal dendrites of layer V pyramidal cells than in those of layer III pyramidal cells.

Notes: Summary The macular mottled mouse is a murine model of the kinky hair syndrome, characterized by a deficiency in copper transport. Cytochromec oxidase (CCO), a respiratory enzyme, is located in the inner mitochondrial membrane and consists of seven subunits, along with copper and iron. Biochemical and histochemical findings indicated that CCO activity was decreased in the cerebellum of the macular mottled mice but not in that of the controls. Immunocytochemical analysis, using anti-CCO and anti-complex III rabbit sera, revealed that CCO in the macular mottled mice was stained more weakly than that in the controls. Immuno-electron microscopic examination of CCO and complex III, using a method of gold labeling, was also performed. In the control mice, a high concentration of gold particles present over CCO and complex III could be seen in the inner mitochondrial membrane. The number of CCO-labeled gold particles was remarkably less, however, in the macular mottled mice, while no significant difference was found in the labeling of complex III between the two groups. It may concluded that the very low CCO enzyme content in the macular mottled mouse results not only from a copper transport disorder but also from a CCO protein synthesis disorder which impairs the localization of CCO protein in the cerebellum.

Notes: Abstract We previously found an antigen-specific factor capable of augmenting delayed-type hypersensitivity (DTH) in the culture supernatant of the mixture of immune spleen cells and erythrocyte antigen, or in the serum of mice immunized with heterologous erythrocytes and exhibiting delayed-type footpad reaction. To elucidate whether this kind of factor (DTH-augmentation factor; DAF) participates in the establishment of DTH to various kinds of antigen besides erythrocyte antigen, we chose a bacterial antigen, Listeria monocytogenes, which is a facultative intracellular bacterium. In the present study, we demonstrated that the immune serum from mice immunized with viable Listeria augmented the delayed-type footpad reaction to Listeria. Furthermore, acquired resistance against Listeria was also augmented by the transfer of such immune serum. Such augmentation of acquired resistance was observed in sites infected locally and in the spleen of mice infected systemically. This effect was also seen in sera from mice immunized with heat-killed Listeria emulsified with complete Freund's adjuvant.

Notes: Summary Neurons projecting to the retina were identified electrophysiologically by antidromic responses in the preoptic retinopetal nucleus (PRN), which is the major origin of the centrifugal fibers to the retina in the filefish. Electrical stimulation of the optic tectum demonstrated that such neurons received excitatory inputs directly from the optic tectum.

Notes: Summary The pharmacokinetics of a new, potent H2-receptor antagonist, famotidine, 20 mg i.v. was studied in 7 subjects with normal renal function and in 24 patients with varying degrees of renal impairment. The volume of distribution at steady state was 1.14 l/kg in normal subjects and was not altered in renal failure. The half-life of elimination was 2.59 h in normal subjects and was unchanged in mild renal failure (creatinine clearance, CLCR 90–60 ml/min/1.48 m2) but was increased to 4.72 h in moderate renal failure (CLCR 60–30 ml/min/1.48 m2), and to 12.07 h in severe renal failure (CLCR below 30 ml/min/1.48 m2). The cumulative urinary excretion and renal clearance of famotidine were correspondingly reduced in patients with impaired kidney function. In normal subjects and in patients with mild to moderate renal failure, about 70% of famotidine was excreted through the kidney, mainly by tubular secretion. In patients with a CLCR above 60 ml/min/1.48 m2 the normal daily dose of famotidine can be employed, but in those with a CLCR between 60 and 30 ml/min/1.48 m2 the dose should be reduced by half, and in patients with a CLCR below 30 ml/min/1.48 m2 a reduction by three quarters of the normal dose is recommended.

Notes: Summary The immediate haemodynamic effects of the calcium antagonist nilvadipine have been studied in ten patients with established mild essential hypertension. Nilvadipine 4 mg p.o. reduced both the systolic and diastolic blood pressures within 60 min, associated with a fall in total peripheral resistance and an increase in heart rate and cardiac index. The peak of blood pressure and total peripheral resistance reached during a cold pressor test were reduced by nilvadipine, but it did not affect the haemodynamic responsiveness to cold stimulation. Plasma renin activity was unaltered and the plasma noradrenaline concentration was increased only slightly. Thus, nilvadipine lowered blood pressure at rest and during cold stimulation as a result of arteriolar dilatation. The hypotensive effect at rest was associated with a reflex increase in heart rate and cardiac index.

Notes: Summary We have studied pharmacokinetics of a new H2-receptor antagonist, TZU-0460, in patients with varying degrees of renal impairment. The apparent volume of distribution at steady-state was 1.70 l/kg, and the plasma protein binding of TZU-0460 or its active metabolite, desacetyl TZU-0460 was less than 10% in normal subjects. These variables were not altered with renal impairment. Sixty percent of TZU-0460 given orally was excreted via the kidney, mainly by tubular secretion. The half-time of elimination was 3.94 h in normal subjects, and was prolonged to 12.13 h in severe renal failure (creatinine clearance below 30 ml/min/1.48 m2). Dosage adjustment of TZU-0460 is necessary in renal failure.

Notes: Thirty-eight California and French wines were analyzed for γ-nonalactone by Freon extraction and gas chromatography. The California white wines had from 0 to 16 pg/L (average for 6 samples was 7 μg/L and the red wines from 12 to 43 μg/L (average for 20 samples was 24 μgL). French red wines obtained similar amounts, 14 to 41 μgL (average for 12 samples was 21 μgL). The sensory threshold for γ-nonalaconte was determined to be 30 μg/L.
