    Notes: A 19F NMR multipulse measurement of the chemical shift in a single crystal of cubic RbCaF3 gave −47.0±3 ppm as the isotropic value with respect to C6F6, and 128.7±6 ppm as the anisotropy. The shielding is accounted for by the usual diamagnetic and paramagnetic ionic overlap and covalent terms. As a result, the Ca++ −F− bond is estimated to be 98% ionic. No significant spectral changes were found when the crystal was cooled below the cubic to tetragonal phase transition temperature. Reasons for this are given.

    Notes: Using multipulse techniques, principal values of the proton nuclear magnetic resonance (NMR) chemical shift tensors were measured at room temperature for the monoclinic form of oxalic acid, potassium hydrogen oxalate, and potassium hydroxide. The isotropic and perpendicular shifts of the first two compounds, which are of medium hydrogen bond strength, were found to fit the linear correlation with O⋅⋅⋅O distance, RO⋅O, established by Rohlfing, Allen, and Ditchfield for medium and strongly O–H⋅⋅⋅O hydrogen bonded solids. Although KOH is very weakly hydrogen bonded, the shifts were also found to conform to the correlation, at least as well as does the other data, thus extending this to weakly hydrogen bonded solids. An empirical correlation of the isotropic and perpendicular shifts with exp(−RO⋅O/ρ), where RO⋅O is in A(ring) and ρ is 0.94 A(ring), is given here which has better agreement with the data and has an interpretation in terms of a simple ionic model of an O–H⋅⋅⋅O bond.

    Notes: The proton NMR shielding tensor was measured at room temperature in the hexagonal layered compound magnesium hydroxide. The isotropic shielding is 29.9±2 and the anisotropy is 12.7±2 ppm, on the 1H absolute scale. From a Gierke and Flygare (GF) analysis of the data the OH− spin–rotation constant was found to be 51±14 kHz. The GF contributions from the neighbors of a given OH− group were subtracted from the experimental shielding and corrections were made for the effects of vibrational motion to give an isotropic shielding of 29.9±2, with an anisotropy of 13.7±2 ppm, for an isolated OH−. The OH− shielding anisotropy is in good agreement with the ab initio calculations of Ratcliffe et al., but the isotropic shielding is not. Proton spin–lattice relaxation measurements at 300 and 77 K gave evidence of proton conductivity in Mg(OH)2.

    Notes: Summary The ability of immature embryos of wheat (Triticum aestivum L.) to respond in cell culture was examined in crosses between the ‘Wichita’ monosomic series and a highly regenerable line, ‘ND7532’. Segregation in disomic controls and 13 monosomic families showed a good fit to a monogenic ratio indicating a qualitative mode of inheritance. Segregation in the cross involving monosomic 2D showed a high frequency of regeneration (93.6%) and high callus growth rate (1.87 g/90 days) indicating that 2D is a critical chromosome. Modifying genes may be located on other chromosomes. Substitution of chromosomes from a low regenerable cultivar ‘Vona’ further indicated that the group 2 chromosomes, in particular chromosome 2D, possess genetic factors promoting callus growth and regeneration.

    Notes: Summary Previous C-banding analysis of wheat (Triticum aestivum)X rye (Secale cereale) hybrids regenerated from tissue culture revealed enlarged C-bands in some rye chromosomes, but the molecular nature of the change was not determined. In situ hybridization using two DNA probes containing repeated sequences from rye telomeric heterochromatin was conducted on these wheatX rye hybrids and their progeny to investigate the occurrence of amplification in repeated sequences. Clones pSC 74 and pSC 119, which contain sequences from the 480-bp and 120-bp repeated DNA families of rye, respectively, were used as probes. Amplification of 480-bp repeated sequences in the short arm telomere of chromosome 7R was detected in three wheatxrye hybrids and their progeny. The amplified 480-bp sequences were detected by an enlarged hybridization site for pSC 74 at the 7RS telomere, and by the appearance at this same telomeric site of an unlabeled, blue chromosome segment in an otherwise completely brown chromosome hybridizing entirely to the biotin-labeled pSC 119 probe. This variant form of chromosome 7R was not observed in several ‘Chaupon’ plants, or in the other hybrids derived from the same embryos, indicating the origin of the change in tissue culture. The amplified sequences were inherited up to at least three generations. Deletions and translocations were also observed.

    Notes: Summary The ability of immature embryos of wheat (Triticum aestivum L.) to respond to tissue culture has been shown to involve the group 2 chromosomes. The available group 2 ditelosomic and nullisomic-tetrasomic lines of ‘Chinese Spring’ wheat were used to determine the chromosome arm location and chromosome dosage effect associated with the expression of tissue culture response (TCR). Significant differences were found between the aneuploid lines and the euploid control for the expression of both regenerable callus formation and callus growth rate. A model is proposed suggesting that a major TCR gene is located on 2DL and that 2AL and 2BS possess minor TCR genes. Furthermore, a major regulatory gene controlling the expression of TCR genes may be located on chromosome 2BL.

    Notes: Summary Fertile r0 plants of the winter wheat line ND7532 (Triticum aestivum L.) were regenerated from callus tissue after 60–190 days in culture. Seeds produced from these self-pollinated plants were planted in the field. Of the 5586 R1 plants, 32 differed for one or more agronomic traits from plants not passed through tissue culture process. Gliadin electrophoregrams were prepared from bulk samples of R2 seed from these 32 plants. Four of the 32 produced gliadin patterns different from controls, so 12 seeds of each of these four lines were examined individually. Three of the four mutant lines were fixed for the presence of a mutant protein of 50 relative mobility units (RMU) and the corresponding loss of a parental protein of 26 RMU. The remaining line segregated for the presence/absence of band 50 and the corresponding loss/retention of band 26. The mutant protein of 50 RMU was never seen in control plants. This indicated that either band 50 was coded for by a mutant gene allelic to the gene that coded for band 26 or that bands 26 and 50 were coded for by two different structural alleles under the control of a common regulatory locus. Each of the 12 seeds from the four mutant lines contained a prominent protein band at 30 (RMU), which was only observed as a faint band in one control seed. The types of variation in gliadin patterns observed in somaclones of ND7532 were similar to those reported for the line ‘Yaqui 50E’, except that, gliadin changes occurred less frequently in ND7532.
