    Notes: Summary Repetitive DNA sequences, derived from the human β-globin gene cluster, were mapped within a series of human genomic DNA segments containing core (H2A, H2B, H3 and H4) and H1 histone genes. Cloned recombinant λCH4A phage with human histone gene inserts were analyzed by Southern blot analysis using the following32P-labeled (nick translated) repetitive sequences as probes:Alu I,Kpn I and LTR-like. A cloned DNA designated RS002-5′C6 containing (i)a (TG)16 simple repeat, (ii) an (ATTTT)n repeat and (iii)a 52 base pair alternating purine and pyrimidine sequence was also used as a radiolabelled hybridization probe. Analysis of 12 recombinant phage, containing 6 arrangements of core histone genes, indicated the presence ofAlu I,Kpn and RS002-5′C6 repetitive sequences. In contrast, analysis of 4 human genomic DNA segments, containing both core and H1 histone genes, indicated the presence of onlyAlu I family sequences. LTR-like sequences were not detected in association with any of the core or H1 histone genes examined. These results suggest that human histone and β-globin genes share certain aspects of sequence organization in flanking regions despite marked differences in their overall structure and pattern of expression.

    Description / Table of Contents: Zusammenfassung Bei 50Escherichia coli-Stämmen, die aus dem Blasenpunktionsurin von Patienten mit chronischer Pyelonephritis gewonnen wurden, erfolgten Bestimmungen ihrer Virulenzeigenschaften. In den 26 Fällen mit einer akuten Erkrankung fanden sich ausschließlich S-Formen der Bakterien, K 1 Antigen wurde in 30% nachgewiesen, 77% verfügten über die Fähigkeit zur Hämolysinbildung und 30% über die zur Colicin V (Aerobactin)-Produktion. In 70% fanden sich Plasmidspezies der Größe zwischen 50 und 70 Md. Demgegenüber wurden bei den 24 Patienten mit einem inaktiven Stadium der Pyelonephritis nur in 70% derE. coli-Stämme S-Formen gefunden. Bei ihnen konnten in 10% K 1 Antigen, in 20% Hämolysin und in 10% Colicin V bestimmt werden. Plasmide zwischen 50 und 70 Md. lagen in 30% der Bakterienstämme vor. Nach der multivariaten Varianz- und Diskriminanzanalyse bestätigt sich, daß uropathogene Bakterienstämme über mehrere Virulenzeigenschaften verfügen müssen, dabei kommt der Fähigkeit zur mannoseresistenten Hämagglutination eine besondere Bedeutung zu.
    Notes: Summary In 50Escherichia coli strains obtained from the bladder puncture urine of patients with chronic pyelonephritis, determinations of virulence properties were performed. All of theE. coli strains isolated from 26 acute episodes of pyelonephritis were found in the smooth form. 30% possessed K 1 antigen, 77% showed the ability to form hemolysin and 30% produced colicin V (aerobactin). Fimbriae (detected by mannose-resistant hemagglutination) were registered in 81%, and plasmids ranging between 50 and 70 Md were demonstrated in 70% of the bacteria. In contrast to this, only 70% of theE. coli strains isolated from 24 patients at an inactive stage of pyelonephritis were found in the smooth form; 10% of these encoded K 1 antigen, 20% hemolysin and 10% colicin V. Plasmids in the range 50 to 70 Md could be found in 30%. On the basis of multivariate analysis of variance and discriminant analysis, it was confirmed that uropathogenic strains possess several virulence properties, mannose-resistant hemagglutination being of particular importance.

    Notes: Abstract To address the relationship between chromatin structure and histone gene expression, the nucleosomal organization of a cell cycle-dependent human H4 histone gene in a bovine papilloma virus (BPV) minichromosome was examined. The nucleosome repeat length of the human H4 histone gene, maintained as a stable episome in a C127 mouse cell line designated 1–8 [1], was compared with that of the chromosomal copy of the H4 gene in human (HeLa) cells. In both cell lines, the H4 histone gene is predominantly expressed during the S phase of the cell cycle. The nucleosome repeat length of total HeLa cell and C127 mouse cell chromatin was similarly examined. Nuclei were digested with micrococcal nuclease and the DNA was fractionated electrophoretically, transferred to nitrocellulose filters and hybridized with radiolabelled (32P) cloned DNA probes. The nucleosome repeat length of the H4 gene, as an episome in the C127 mouse cell (153 ± 8) and as an integrated copy in a HeLa cell (163 ± 10) was considerably shorter than total genomic host cell (C127) (190 ± 5) or HeLa cell chromatin (183 ± 7). Our results indicate that the episomal H4 histone gene is packaged as chromatin. Moreover, the shortened nucleosome repeat length of the H4 gene, both as an episome or integrated chromosome sequence, suggests that the repeat length is characteristic of the gene and may be functionally related to its cell cycle regulated expression.

    Description / Table of Contents: Investigation of Non-Magnetic Manganese-Silicon-Steels for Retaining RingsThe need for higher strength of retaining rings on electric generator shafts is met today by stress corrosion resistant austenitic manganesechromium-nitrogen-steels like X 5 MnCr N 18 18. The aim of this paper is the investigation of less expensive manganese-silicon-carbon-steels for perfectly dry conditions. By cold working and a recrystallising annealing treatment, as well as by the addition of 0.1 % niobium a small grain size is achieved. Thereby the flow stress is raised and furthermore by silicon which increases the solution strengthening and the work hardening rate during the final cold working. Compared to the steel X 55 MnCr 184 the alloy X 55 MnSi 18 3 shows a higher strength but is more sensitive to stress corrosion cracking.
    Notes: Dem Wunsch nach hochfesten Kappenringen für Wechselstromgeneratoren wird heute durch spannungsrißkorrosionsbeständige austenitische Stähle wie X 5 MnCrN 18 18 entsprochen. Das Ziel der vorliegenden Arbeit war die Untersuchung kostengünstigerer Mangan-Silizium-Kohlenstoff-Stähle für völlig trockene Betriebsbedingungen. Durch Kaltverformung und Rekristallisation sowie durch Zugabe von 0,1% Niob läßt sich eine feine Korngröße einstellen. Dadurch steigt die Fließgrenze wie auch durch die Zugabe von Silizium, das die Mischkristallhärte und den Verfestigungskoeffizient bei der abschließenden Kaltverformung erhöht. Im Vergleich zum bisher gebräuchlichen kostengünstigen Stahl X 55 MnCr 184 zeigt die Legierung X 55 MnSi 18 3 eine höhere Festigkeit, aber auch eine stärkere Neigung zur Spannungsrißkorrosion.

    Notes: Protein-DNA interactions within the promoter of a cell cycle-regulated human H4 histone gene were examined by binding of 5′-end-labeled DNA segments to Western blots of nuclear protein fractions. Specific protein interactions were observed with DNA segments located between -500 bp and -1,070 bp upstream of the ATG initiation codon and included a histone H1 binding segment flanked on both sides by binding sites for a 45 kD nuclear protein. This region of the gene contains a DNase I-sensitive site in the center (-720 to -820 bp), and sequence analysis revealed the presence of scaffold attachment sequences in the two flanking segments. Topoisomerase II consensus sequences and in vitro topoisomerase II cleavage sites were also detected in the two flanking segments. Our results suggest that the 45 kd nuclear protein may preferentially interact with these two segments of the H4 histone gene to mediate association with the nuclear matrix. The presence of negative regulatory elements in this putative matrix attachment region provides a basis for the speculation that such nuclear proteins are associated with alterations in gene-matrix interaction that are functionally related to gene expression.

    Notes: We have identified a segment of DNA in the region 6,500 nucleotides upstream from a cell-cycle-dependent human H4 histone gene (pF0108A) which exhibits properties of an enhancer element. This distal element is not required for cap site initiation from the F0108A H4 histone gene. When the enhancer element is present in the genome as a stable integrated sequence, either in its natural upstream location or in a construct where the element is moved just upstream from the proximal promoter sequences, a 25-fold increase in the level of human H4 histone RNAs is observed. This increased level of mRNA reflects an increase in the rate of transcription. The enhancer effect is also observed when the distal element is inserted in inverse orientation with respect to this gene. In addition, the far upstream element can increase expression of a prokaryotic chloramphenicol acetyl transferase (CAT) gene under control of the simian virus 40 (SV40) early promotor, indicating that the ability to influence transcription is not confined to the gene with which it is normally associated. The ability of the histone gene distal enhancer element to function in both mouse and human cells indicates that transacting regulatory factors encoded by either the human or murine genome are capable of mediating the functional properties of this element, further supporting the cross-species compatibility of regulatory sequences and molecules that influence transcription of human histone genes.

    Notes: A Hopf bifurcation where a stable fixed pointer bifurcates into a stable periodic orbit as a parameter passes through a critical value, occurs frequently in nonlinear problems. Here, the Maynard Smith nonlinear map in population dynamics involving a parameter k is analysed with the help of numerical experiments in order to determine the periodic structure of the map beyond the primary Hopf bifurcation of period 6 which occurs at k = 1. The interesting result obtained is that in the parameter range from k = 1 to the value for blow-up of all initial conditions, there are successive windows of periods 7, 8 and 9, the last containing a secondary Hopf bifurcation of period 4.
