    Notizen: Summary Crude 3 M KCl extracts of the methylcholanthrene-induced fibrosarcoma of C3H/HeJ mice, MCA-F, were demonstrated to contain two fractions, one inducing tumor resistance and the other facilitating the outgrowth of neoplastic cell challenge. In immunoprotection tests in syngeneic C3H/HeJ mice, optimal doses of crude solubilized tumor antigen afforded only a 28% reduction in growth compared with saline-treated controls. When crude extracts were fractionated by preparative isoelectric focusing (pIEF) in a slab of superfine Sephadex G-75, significant biologic activity was demonstrated in two fractions. Fraction (Fr) 1, pI 2.5–3.6, induced potent tumor facilitation, increasing the tumor size by more than 100%, while Fr 15, pI 5.8–6.0, engendered resistance that reduced their respective biological effects to MCA-F, but not the antigenically unrelated MCA-D tumor. Thus 3 M KCl extracts contain at least two biologically active components, one immunoprotective and one tumor-facilitating. Since the weak immunoprotective activity of crude materials may represent the vectorial effect of these antagonistic components, subsequent molecular characterization of both moieties may afford insight into the complex response of hosts toward tumors. Furthermore, TSTA purified by the rapid method of isoelectric focusing may be a more suitable reagent for immunotherapy than the parent crude 3 M KCl extracts by virtue of the absence of facilitating antigens.

    Notizen: Summary Rabbits imunized with the immunoprotective TSTA fraction partially purified by preparative isoelectric focusing of 3 M KCl extracts from a chemically induced murine sarcoma, MCA-F, produced specific xenoantisera as assessed by an indirect membrane immunofluorescence assay. Only the immunizing tumor, MCA-F, and not the antigenically distinct MCA-D or MCA-T target cells were stained by the xenoantiserum. Absorption of anti-MCA-F antiserum with the antigenically distinct MCA-D or MCA-T cells did not reduce its capacity to bind to MCA-F cells. The immunofluorescence reaction was competitively inhibited by MCA-F fractions that induced specific immunoprotection: crude 3 M KCl extract, isoelectrically focused TSTA (fraction 15), and intact irradiated MCA-F cells. The TSTA specificity of these xenoantisera suggests that they may provide useful reagents for rapid isolation and characterization of the immunoprotective moiety.

    Notizen: Summary Tumor-specific transplantation antigens (TSTA) were purified from 3 M KCl and butanol extracts of C3H/HeJ 3-methylcholanthrene-induced fibrosarcomas by preparative isotachophoresis (pITP). Fractions from pITP which reacted with antisera to TSTA preparations in an enzyme-linked immunospecific assay were tested in vivo for induction of resistance to the growth of transplanted tumor cells. Isotachophoresis of crude 3 M KCl extracts yielded TSTA that was immunogenic at doses between 17 and 124 μg. Isotachophoresis of TSTA partially purified from crude 3 M KCl or butanol extracts by preparative isoelectric focusing (pIEF) of 3 M KCl and butanol extracts yielded TSTA that was immunogenic over a two-fold log dose range. As little as 10 ng purified TSTA reduced tumor growth by 50%. Tumor growth reduction was specific, and immunized animals survived longer than non-immunized controls. A purification of 10,000-fold over crude 3 M KCl extracts and 2,500-fold over crude butanol extracts was obtained. These results suggest that TSTA from murine tumor cells is preparatively purified by following extraction with pIEF and then pITP.

    Notizen: Summary Adjuvant immunotherapy with hemolytic streptococci abates the growth of a syngeneic methylcholanthrene (MCA)-induced fibrosarcoma propagated in C3H/HeJ mice. Interperitoneal (IP) administration of streptococci either prior to or after tumor inoculation reduced neoplastic outgrowth. While subcutaneous (SC) administration of streptococci prior to tumor inoculation did not influence tumor outgrowth, SC treatment afterwards was effective. Thus, therapeutic effects of streptococcal vaccine depend upon the route and/or timing of administration. Surgery performed shortly before tumor inoculation abrogated host response to streptococci. On the other hand, surgery performed 3 days after tumor inoculation did not alter the adjuvant action of streptococcal vaccines. The failure of attempts to achieve an immunotherapeutic effect in splenectomized hosts suggests that the spleen was essential for the action of the streptococcal vaccine.

    Notizen: Abstract Nonspecific immune monitoring of the percentage active T (A-T) rosette-forming cells (RFC) and spontaneous blastogenesis (SB) appear to be useful indexes of host reaction toward allografts. In order to assess the significance of the observed changes, renal failure patients were serially evaluated before and after hemodialysis for the percentage of total T (T-T) and A-T RFC and for peripheral blood leukocyte metabolic activity measured by a whole blood spontaneous blastogenesis (SB) assay. Renal failure patients had a significantly lower (P〈0.05) percentage of T-T RFC than normal persons, while the percentage A-T RFC and SB remained stable. A hemodialysis treatment did not change the mean values of any of the three parameters. On the other hand, serial patient evaluation over 3 months revealed significant fluctuations in the percentage T-T RFC, but not the percentage A-T RFC and SB. Since the percentage A-T RFC and SB are relatively constant measures of immune status in ungrafted patients, significant changes in these nonspecific immune probes may reflect allograft rejection.

    Notizen: Abstract Morphologic changes of the spleen and dynamic changes of biologic activity of spleen cells from C3H/HeJ mice bearing methylcholanthrene induced fibrosarcoma (MCA-F) were investigated in different stages of tumor growth, using local adoptive transfer assay (LATA). The spleen size, weight and the number of spleen cells increased with the tumor growth. In early and late stages of tumor growth, tumor-bearing mice possessed non-specific tumor-enhancing cells which were radioresistant (700 rads), phagocytic and adherent, suggesting that they were macrophages. On the other hand, in middle stages of tumorgenesis, tumor-bearing mice possessed specific cytotosic cells which were radiosensitive (700 rads) and anti-Thy 1.2 serum positive, suggesting that they were T-cells. Thus, the appearance of tumor-enhancing cells was earlier than cytotoxic cells in MCA-F bearing host, and the balance of tumor-enhancing and cytotoxic cells may influence the tumor outgrowth in different stages of tumorgenesis. The spleen serves as a reservoir of factors which either abrogate cell mediated resistance or stimulate neoplastic growth, or as the source of cytotoxic cells, at certain times of tumor burden.

    Notizen: Abstract The effect of splenectomy on neoplastic outgrowth was examined prior to and after implantation of methylcholanthrene-induced C3H/He murine tumors. Splenectomy performed 12 days before tumor inoculation did not affect the tumor outgrowth, however, both splenectomy and sham operation performed shortly before tumor inoculation resulted in significant tumor facilitation compared with the non-operated group, suggesting that this accelerated tumor was not related to the presence or absence of splenic tissue, but rather to systemically-induced immunosuppression. While splenectomy performed 6, 9, 12 days after tumor inoculation did not alter the tumor growth, splenectomy performed early (3 days) or late stage (20 days) after tumor cell challenge revealed a retarded neoplastic outgrowth, compared with the sham operated group. These results suggest that splenectomy in very early and late stages of tumor-bearing host may be effective for tumor treatment.
