Notes: Summary Activity of the enzyme glutathione reductase (EC 1.6.4.2) in erythrocytes and fibroblasts of a patient with karyotype 46, XY, del(8) (pterp212:) was found to be in the normal range. With results from other laboratories, this allowed a more precise mapping of the gene for this enzyme in the region 8p2100–8p212.

Notes: Abstract A mentally retarded 8-year-old boy with a de novo partial monosomy for the short arm of the No. 8 chromosome is described. Based on G-banding analysis, the patient's karyotype was identified in lymphocytes and skin fibroblasts as 46,XY,del(8) (pter→p21:). No chromosomal abnormalities were found in the phenotypically normal mother, father and sister of the propositus. Four further cases described in the literature indicate that partial monosomy of the short arm of the No. 8 chromosome might be associated with a syndrome characterized by the following stigmata: mental retardation, slow growth, high forehead, broad chest, wide-set nipples, pulmonary stenosis with atrial and/or ventricular septal defect, hypoplasia of the genitalia, dermatoglyphic stigmata.

Notes: Summary The effect of low doses of heparin on the binding of phenytoin and prazosin to plasma proteins was evaluated in four normal subjects. Heparin activates the hydrolysis of triglycerides in plasma. The ensuing increase in non-esterified fatty acids (NEFA) was more marked in vitro than in vivo and increased the free fraction (FF) of phenytoin and prazosin in plasma. The higher FF caused a change in the plasma to whole blood ratio (P/B ratio) of both drugs. The changes in FF and P/B ratio after heparin were small, but could be of significance in pharmacokinetic studies.

Notes: Summary We have measured the metabolites (demethylated and hydroxylated) of amitriptyline in a group of seven normal volunteers. They were phenotyped as extensive or poor metabolizers using debrisoquine and bufuralol. The results demonstrate that the oxidative metabolism (aliphatic hydroxylation) of amitriptyline is under the same genetic control as that of debrisoquine and bufuralol. However, phenotypic polymorphism cannot be used to predict amitriptyline blood concentration after a single oral dose, since the principal metabolic pathway of amitriptyline is demethylation and not aliphatic hydroxylation.

Notes: Summary Prescribing patterns were studied in a psychiatric outpatient clinic. Psychiatrists tended to prescribe large quantities of antidepressants or other drugs. In contrast, the daily doses for antidepressants and neuroleptics were often too low. Significant variations were noted between three outpatient clinics in the choices among a various psychotropic drugs from a given therapeutic group.

Notes: Summary In a companion paper we described the disposition of a 75 mg single dose of amitriptyline in normal volunteers who were phenotyped as extensive or poor metabolizers of debrisoquine and bufuralol, and had a four-fold range in the oral clearance of the antidepressant, 50 mg of amitriptyline was also administered to the same volunteers. This paper compares the results after both doses and suggests that the disposition of amitriptyline is linear even in subjects with a low oral clearance. There was no relation between the pharmacokinetic data and the intensity of sedation or of psychomotor impairment.

Description / Table of Contents: Preparation, Crystal Structure, and Temperature Dependence of the Homogeneity Range of the Phase Ca2-xAs1-xBr1+x and Thermal Behaviour of Ca3AsBr3The phase (Ca2-x□x)(As1-xBr1+x) (yellow, NaCl type lattice, x ≙ degree of substitution) was prepared from “Ca3As2” and CaBr2 in different molar ratios in steel ampoules under argon at 900 and 950°C resp. The lattice constant as a function of the composition, the homogeneity range, and dependence of the bromine rich phase boundary on the temperature were determined. The structure was deduced from single crystal X-ray investigation and density measurements at different compositions. The thermal behaviour of Ca3AsBr3 (colourless, isotypic to Mg3NF3, prepared at 850°C) was studied by annealing samples in molybdenum ampoules under argon in the temperature range 900-1250°C and by differential thermal analysis. From the experimental results a section of the phase diagram Ca3As2-CaBr2 was constructed.

Description / Table of Contents: Chalcogenolates and their Derivatives. I. Syntheses and Properties of Ionic ChalcogenophenolatesThe syntheses and properties of ionic chalcogenophenolates are described. Using liquid ammonia as solvent the alkali chalcogenophenolates M[EPh] (M = Na, K; E = Se, Te; Ph = C6H5) have been synthesized via reduction of the diphenyl dichalcogenides with alkali metals. Similarly, the tetraphenylphosphonium chalcogenophenolates [Ph4P][EPh] (E = S, Se, Te) have been obtained by reacting alkali chalcogenophenolates with tetraphenylphosphonium chloride.
