    Notes: Summary The retinal microcirculation of anaesthetised normal cats was studied during hyperglycaemia (15 to 55 mmol/l) induced by intravenous infusion of glucose, using high speed cine fluorescence angiography. Saline (0.150 mmol/l) was infused as a control for the volume effect of glucose solution and equiosmolar mannitol was infused as a control for the osmotic effect. The mean retinal arteriolar inflow rate increased from 34±1 mm/sec to 41±4 mm/sec during glucose infusion, and from 46±1 mm/sec to 56±3 mm/sec during mannitol infusion. The blood pressure similarly increased from 105±5 mmHg to 125±2 mmHg during glucose infusion and from 110 ±7 mmHg to 129±1 mmHg during mannitol infusion. During mannitol infusion the increased inflow was accompanied by a reduction in the arteriolar width so that the volume flow remained unchanged. During glucose infusion this constriction did not occur, resulting in a significantly increased volume of retinal blood flow (9±1 μl/min to 12±1 μl/min).

    Notes: Summary The effect of insulin injection in fetal rabbits on plasma somatomedin activity and cartilage metabolism was investigated. One fetus in each of 12 litters was injected with 1 unit of insulin zinc suspension subcutaneously on day 27 of gestation and a control fetus was injected with the same volume of 0.154 mol/l saline. The litter was delivered by caesarean section on day 29 and each fetus identified. Plasma somatomedin activity was determined by fetal rabbit cartilage bioassay. Costal cartilage from individual fetuses was incubated in medium containing [3H]thymidine or [35S]sulphate as indicators of cell replication and matrix synthesis respectively. Individual values for somatomedin activity or cartilage isotope uptake were ranked within a litter. In each case the rank in the litter of the insulin-injected fetus, but not the saline-injected fetus, was significantly higher than the mean rank of the litter. Insulin did not stimulate cartilage metabolism in vitro.

    Notes: Summary A mildly diabetic state was induced in pregnant rats following treatment with streptozotocin the day after mating. On day 21 of gestation, these rats had a lower plasma insulin (55±9 versus 107±23 mU/l for control rats; p〈0.05, mean ±SEM) and a reduced pancreatic area occupied by insulincontaining cells compared with control animals (0.40±0.04 versus 1.03±0.08%; p〈0.001), but hyperglycaemia was not apparent. Fetuses from mildly diabetic animals were longer but not heavier than those from control rats. Plasma somatomedin activity measured by fetal rat cartilage bioassay was higher in fetuses from mildly diabetic rats (1.12±0.07 versus: 0.74±0.05 U/ml for control fetuses; p〈0.001) as was cartilage metabolic activity in basal culture medium (35S sulphate uptake) (1 883±141 versus 1473±104 c.p.m./mg for control rats; p〈0.05), but plasma insulin levels and the pancreatic area occupied by insulin-containing cells did not differ between the two groups of fetuses. Fetal plasma somatomedin activity, measured by fetal cartilage assay, showed a significant positive correlation with both body weight and length. It is concluded that by day 21 of gestation a small body overgrowth had occurred in the fetus of the mildly diabetic rat and this was associated with an increase in plasma somatomedin activity, but not with any abnormality of circulating insulin levels or volume density of B cells in the pancreatic islets.

    Notes: Abstract: GTP and GDP decreased the saturable binding of [3H]baclofen or [3H]γ-aminobutyric acid ([3H]GABA) to GABAB but not GABAA receptors whereas GMP displayed negligible activity. This effect was specific to guanyl nucleotides and was not mimicked by high concentrations of ATP. The inhibition of ligand binding was the result of a diminished receptor affinity with no change in receptor number. The use of a complete physiological saline solution rather than Tris buffer plus Ca2+ or Mg2+ increased the potency of GTP at the GABAB receptor. The results are discussed in relation to the effects of GABA and GTP on adenylate cyclase activity in the brain.

    Notes: Immunoglobulin E (IgE), directed against components of Acacia (wattle) pollen, has been detected by radioallergosorbent tests (RAST) in the sera of some children and adults who develop allergic symptoms in the presence of flowering Acacia trees in Australia. All these subjects also had high levels of IgE directed against Lolium perenne (rye grass) pollen. Inhibition by RAST showed that most of the IgE molecules which bound to Acacia pollen components also bound to L. perenne pollen extracts, and to Glycoprotein I, the major allergen of L. perenne pollen. In these assays, the allergens have been immobilized on polyvinyl chloride microtitre trays: the sensitivity of this approach is compared to that of commercial RAST kits.

    Notes: In this study, the ability to predict allergenic bronchial reactivity from non-specific bronchial reactivity, measured by free corridor-running, and the level of skin reactivity to whole rye grass pollen extract (WRGPE–Lolium perene) and house-dust mite extract (HDM–Dermatophagoides pteronyssinus) was examined in asthmatic children. Discriminant analysis of the data showed that, for the concentration of extracts used and the conditions of bronchial challenge for both allergens, the level of skin sensitivity to undiluted extract and the percentage fall in peak flow-rate on exercise best-predicted the outcome of bronchial challenge for WRGPE (100% of cases) and for HDM (89% of cases).The results suggest that it may be possible to predict allergenic bronchial-reactivity to different allergens in children by simple clinical methods, if conditions of provocation and potency of allergen extracts can be standardized.

    Notes: The pattern of the humoral-immune response to cow's milk was examined in children with eczema and in children who had acute urticarial and/or angioedematous reactions to cow's milk. Each patient group was compared to a group of age-matched controls.Whereas the eczematous patients had significantly elevated IgG, IgA and IgE milk-antibodies, patients, allergic to cow's milk, had elevated IgE milk-antibodies. These differences in the pattern of antibody response to cow's milk suggests that these two patient groups constitute separate allergic populations, and that different pathogenic mechanisms may be operative in these two skin diseases associated with elevated levels of IgE antibodies to cow's milk.
