    Notes: Summary The Ca2+-activated nonselective cation channel in mouse pancreatic acini has been studied with the help of patch-clamp single-channel current recording in both the cell-attached conformation and in excised inside-out membrane patches. In intact resting mouse pancreatic acinar cells no unitary activity was observed. Adding saponin to the bath solution to disrupt the plasma membrane (apart from the isolated patch membrane from which current recording was made) evoked unitary inward current steps when the free ionized Ca2+ concentration in the bath ([Ca2+] i ) was 5×10−8 m or above. When an electrically isolated patch membrane was excised and the internal aspects of the plasma membrane were exposed to the bath solution, channel activation could be obtained when [Ca2+] i was 10−7 m or above. However, with the passage of time the total inward current declined and about 1 min after excision no unitary current steps could be observed. At this stage Ca2+ in micromolar concentration was needed to open the channels and several hundred micromoles of Ca2+ per liter were required for maximal channel activation. Our results indicate that the Ca2+-activated nonselective cation channel is more sensitive to internal Ca2+ than hitherto understood and that it may therefore play a role under physiological conditions in intact cells.

    Notes: Abstract A lithium isotope separation was performed using a laser isotope separation method. It was found that the lithium atoms with a natural isotopic abundance enhanced its6Li concentration up to over 90% by tuning the laser wavelength to the2 P 1/2 of6Li. Too high power, however, leads to a loss of enrichment due to the power broadening effect which was analysed by the equation of motion of density matrices.

    Notes: Summary The anticancer agent 1,3 Bis(2-chloroethyl)-1-Nitrosourea (BCNU) cures the advanced syngeneic LSA lymphoma of C57BL mice with high efficiency. The cured animals resist further tumor challenge by large numbers of viable syngeneic tumor cells. Growth assays of spleen proliferation of the intravenously inoculated tumor revealed a progressive-regressive pattern of spleen growth after LSA-tumor injection. Lymphoma colony forming units (LCFU) in the spleen initially increased then regressed. In vitro assays of serum showed a alack of cytotoxic activity in mice cured by BCNU. Added spleen, thymus, or bone-marrow cells were similarly ineffective. Spleen and bone-marrow cells from immune mice passively transferred to normal mice showed weak cytotoxic activity against the LSA tumor. BCNU-cured mouse cells were more effective in protection than those cured with Chlorozotocin (CLZ).

    Notes: Summary Cationic antigens have been observed to bind with the negatively charged glomerular basement membrane (GBM). Using the cationic reagent polyethyleneimine (PEI), the distribution of glomerular anionic sites was evaluated ultrastructurally in the early stage (2 h-day 7) of cationic antigen mediated in-situ immune complex formation type glomerulonephritis (GN) in the rat. — Renal perfusion via the renal artery with 100 μg of cationized human IgG (pl〉9.5), followed by the i.v. injection of specific antibodies, led to an initial increase in urinary albumin excretion, subsequent massive globulinuria and the formation of numerous subepithelial deposits on day 7. — The most striking alteration in glomerular anionic sites was observed on the epithelial cell surface coat; the PEI deposition on the epithelial cell surface was almost identical to that in control glomeruli at 2 and 4 h after the induction of GN; thereafter, on day 7, a broad loss of anionic sites was obseryed on flattened epithelial foot processes. In contrast, fixed anionic sites of the laminae rarae of the GBM showed no apparent alterations in the distribution and number from 2 h to day 7 and did not disappear even in the lamina rara externa adjacent to subepithelial deposits. — These findings not only show that fixed anionic sites of the GBM, in contrast to the rapid decrease in those of the epithelial cell surface, are not completely neutralized or destroyed even in GN, in which cationic antigen participates in the in-situ formation of GBM-deposits. This also indicates that initial impairment of the charge-selective barrier of the GBM by the in-situ interaction between cationic antigen and antibody, is followed by the disfunction of the size-selective permselectivity of the GBM, ultimately causing massive proteinuria.

    Notes: Summary Health examinations for lead poisoning were made on 62 family members from 15 families of homes carrying on lead work, such as quench-hardening in a molten lead bath and type-printing, as work at home. The most interesting findings concern the occurrence of cases with an unduly high lead absorption among children, but not among adult family members other than home lead-workers. The home environments of the children with an unduly high lead absorption represented contamination with housedust high in lead contents. The ingestion of the contaminated housedust by hand-to-mouth is probably responsible for the excessive lead exposure of the affected children. The results of the present study suggest that contanimation of housedust with lead due to home lead-work constitutes a possible hazardous source of lead exposure for children.

    Notes: Summary The effects of tetraethylammonium (TEA) on the action potential in cultured chick embryonic heart muscle cells were investigated. The onset of prolongation of the action potential occurred within 10 min following intracellular iontophoretic application of TEA, but after more than 50 min following extracellular application. These facts suggest that the major site of action of TEA is on the inner surface of the membrane in these cells.
