    Notes: The magnetoresistance (MR) ratio of a ferromagnetic tunnel junction with a spin-valvelike structure of NiFe/CoFex/Al–O/CoFex/NiFe/IrMn/Al is dependent upon CoFe composition. MR ratio increases with increasing Fe content, and shows a maximum of 42% for Fe content 26 at. % after annealing at 225 °C. Before annealing, the bias voltage dependence depend on Fe content, however, the bias voltage dependence did not depend on Fe content after annealing. We think that the increasing defect states in the barrier layer with the increasing Fe content in CoFex layer cause the degradation of the bias voltage dependence. After annealing, the defect states of samples decrease to same level, and the bias voltage dependencies of samples are improved and become same. © 2000 American Institute of Physics.

    Notes: InGaAs layers with In composition of 0.57 were grown with metalorganic vapor phase epitaxy (MOVPE) using graded buffer layers on exactly oriented and misoriented GaAs(100) substrates. No mirror-like surface was obtained at growth temperatures between 570 °C and 630 °C. A mirror-like surface was achieved at a growth temperature of 450 °C. The threading dislocation density in the layer grown at 450 °C on the GaAs substrate misoriented toward (111)A was determined to be 1×107 cm−2 using transmission electron microscopy. Photoluminescence results also confirmed that the density of recombination centers in layers grown at 450 °C was low. Low temperature growth with MOVPE was found to be effective in InGaAs layers on GaAs substrates with high In composition above 0.4. © 2002 American Institute of Physics.

    Notes: We used Northern blot analysis in order to investigate the ontogeny of the murine joining (J)-chain gene. No J-chain expression was detected in embryonic tissues, including liver, spleen and intestine, but an expression of µ heavy chain was detected in foetal liver at day 17. J-chain expression was detected in the spleen at day 9 and in the intestine at day 15 after birth. Western blot analysis was carried out in order to compare the protein levels of J and µ heavy chains in serum from day 8 to day 24 after birth, using antihuman J chain and antimouse µ chain antibodies. Although µ chain protein could be detected in serum from day 8, J-chain protein was detectable only at day 24. These results suggest that the expression of J chain is a later event than the µ chain in the mouse, which thus differs in embryogenesis from humans.

    Notes: For most studies involving the response of plants to future concentrations of atmospheric carbon dioxide (CO2), a current concentration of 360–370 μatm is assumed, based on recent data obtained from the Mauna Loa observatory. In the present study, average seasonal diurnal values of ambient CO2 obtained at ground level from three global locations (Australia, Japan and the USA) indicated that the average CO2 (at canopy height) can vary from over 500 μatm at night to 350 μatm during the day with average 24-h values ranging from 390 to 465 μatm. At all sites sampled, ambient CO2 rose to a maximum value during the pre-dawn period (03.00–06.00 hours); at sunrise, CO2 remained elevated for several hours before declining to a steady-state concentration between 350 and 400 μatm by mid-morning (08.00–10.00 hours). Responses of plant growth to simulations of the observed variation of in situ CO2 were compared to growth at a constant CO2 concentration in controlled environment chambers. Three diurnal patterns were used (constant 370 μatm CO2, constant 370 during the day (07.00–19.00 hours), high CO2 (500 μatm) at night; or, high CO2 (500 μatm) at night and during the early morning (07.00–09.00 hours) decreasing to 370 μatm by 10.00 hours). Three plant species − soybean (Glycine max, L (Merr.), velvetleaf (Abutilon theophrasti L.) and tomato (Lycopersicon esculentum L.) − were grown in each of these environments. For soybean, high night-time CO2 resulted in a significant increase in net assimilation rate (NAR), plant growth, leaf area and biomass relative to a constant ambient value of CO2 by 29 days after sowing. Significant increases in NAR for all three species, and significant increases in leaf area, growth and total biomass for two of the three C3 species tested (velvetleaf and soybean) were also observed after 29 days post sowing for the high night/early morning diurnal pattern of CO2. Data from these experiments suggest that the ambient CO2 concentration experienced by some plants is higher than the Mauna Loa average, and that growth of some agricultural species at in situ CO2 levels can differ significantly from the constant CO2 value used as a control in many CO2 experiments. This suggests that a reassessment of control conditions used to quantify the response of plants to future, elevated CO2 may be required.

    Notes: Background Prostaglandin D2 (PGD2), a major cyclo-oxygenase metabolite of arachidonic acid in mast cells, induces bronchoconstriction in the human lung. It has been reported that mice lacking PGD receptor fail to develop the bronchial hyper-responsiveness upon ovalbumin challenge, suggesting that PGD2 functions as a mediator of allergic asthma.Objective To determine if there are any mutations associated with the development of asthma in the haematopoietic prostaglandin D synthase (H-PGDS) gene and the human prostanoid DP receptor (PTGDR) gene.Methods and results We screened the 5′flanking and coding regions of the H-PGDS gene and the PTGDR gene by direct sequence. We identified one variant in intron 2 (IVS2 + 11 A 〉 C) and one variant in intron 3 (IVS3 + 13T 〉 C) of the H-PGDS gene, and two variants in the 5′flanking region of the PTGDR gene (−197T 〉 C and −2C 〉 T). The IVS3 + 13T 〉 C and −197T 〉 C variants were rare, appearing only once in 48 subjects. transmission disequilibrium test (TDT) analysis of 144 asthmatic families revealed that the IVS2 + 11 A allele of the H-PGDS gene was significantly transmitted preferentially to asthma-affected children (P = 0.0056), but no association was observed between −2C/T polymorphism of the PTGDR gene and asthma (P 〉 0.05).Conclusion Our results suggest that the IVS2 + 11 A/C allele may be involved in the development of asthma in the Japanese population.

    Notes: IL-4 gene cluster on chromosome 5 contains several candidate genes for atopy and asthma. Several independent studies have shown evidence for linkage between the markers flanking IL-4 gene cluster and asthma and/or asthma-related traits. Interferon regulatory factor 1 (IRF-1) is located approximately 300 kb telomeric to IL-4 and recent study reveals that IRF-1 deficiency results in an elevated production of Th2-related cytokines and a compensatory decrease in the expression of native cell- and Th1-related cytokines.〈section xml:id="abs1-2"〉〈title type="main"〉ObjectiveTo determine if there are any mutations associated with the development of atopy and asthma present in the coding exons and 5′ flanking region of the IRF-1 gene.〈section xml:id="abs1-3"〉〈title type="main"〉Methods and resultsWe have screened the promoter and coding regions of the IRF-1 gene in atopic asthmatics and controls by SSCP method. We found three novel nuclear variants (the −300G/T and 4396 A/G polymorphisms and the 6355G 〉 A rare variant) in the IRF-1 gene. No variants causing amino acid alterations of IRF-1 were detected. The −300G/T polymorphism was in nearly complete linkage disequilibrium with the 4396 A/G polymorphism. An association between the 4396 A 〉 G polymorphism and atopy/asthma was examined by transmission disequilibrium test in 81 asthmatic families. Either of 4396 A or 4396G alleles was not significantly preferentially transmitted to atopy- or asthma-affected children.〈section xml:id="abs1-4"〉〈title type="main"〉ConclusionThe IRF-1 gene is less likely to play a substantial role in the development of atopy and asthma in the Japanese population.

    Notes: Abstract:  Cultured rat keratinocyte sheets form hair follicles in combination with rat vibrissa dermal papillae when they are transplanted subcutaneously in syngeneic rats and athymic mice. In the present study, the histologic details of these induced follicles were analyzed by preparing cultured sheets mixed with normal rat keratinocytes and green fluorescent protein (GFP)-transgenic rat keratinocytes. Histologic examination demonstrated that some induced follicles maintained their size and morphology for at least 18 weeks, whereas others decreased in size and others totally differentiated into cornified structures between 3 and 6 weeks. The percentage of the grafts with GFP-positive cells decreased during the same period. This finding suggests that some GFP-positive cells were transient-amplifying cells that turned into terminally differentiated cells and were lost during this period. Some large follicles and some small follicles maintained their hair-producing ability and the proliferative activity in their hair matrix for 18 weeks. In addition, one 6-week-old follicle contained label-retaining cells in the outer root sheath. Seven of 25 follicles induced from chimera epithelium contained both GFP-positive cells and GFP-negative cells. These results suggest that stem cells are present in the induced follicle and the induced follicle consists of polyclonally derived cells. The presence of early anagen-like large follicles at week 6 and 9 and a telogen-like small follicle at week 18 also suggests that hair-growth cycle phases proceeded in the induced follicles. In conclusion, the follicles induced in the cultured keratinocyte sheets maintained hair-producing ability and proliferative activity for at least 18 weeks. This and the presence of label-retaining cells suggest that there are stem cells in the induced follicles, which seem to have a hair-growth cycle.

    Notes: Abstract: Germinative cells, small cell masses attached to the stalks of dermal papillae that are able to differentiate into the hair shaft and inner root sheath, form follicular bulb-like structures when co-cultured with dermal papilla cells. We studied the growth characteristics of germinative cells to determine the cell types in the vibrissa germinative tissue. Germinative tissues, attaching to dermal papillae, were cultured on 3T3 feeder layers. The cultured keratinocytes were harvested and transferred, equally and for two passages, onto lined dermal papilla cells (LDPC) and/or 3T3 feeder layers. The resulting germinative cells were classified into three types in the present experimental condition. Type 1 cells grow very well on either feeder layer, whereas Type 3 cells scarcely grow on either feeder layer. Type 2 cells are very conspicuous and are reversible. They grow well on 3T3 but growth is suppressed on LDPC feeder layers. The Type 2 cells that grow well on 3T3 feeder layers, however, are suppressed when transferred onto LDPC and the Type 2 cells that are suppressed on LDPC begin to grow again on 3T3. The transition of one cell type to another in vitro and the cell types that these germinative cell types correspond to in vivo is discussed. It was concluded that stem cells or their close progenitors reside in the germinative tissues of the vibrissa bulb except at late anagen–early catagen.

    Notes: The dento-epithelial junctions (DEJ) of Rhesus monkeys were studied by electron microscopy for their histochemical responses to colloidal thorium and ruthenium red. Both of these cationic dyues are useful markers for the detection of acidic carbohydrates. With colloidal thorium, deposits occurred on the intercellular cell surfaces but not on the surfaces facing the tooth. The basal lamina, dental cuticle, desmosomes and hemidesmosomes reacted negatively. With ruthenium red, cell surfaces, desmosomal junctions, the peripheral densities and the spaces between the peripheral densities and the cell membranes of hemidesmosomes stained intensely. The lamina densa was also stained, but the lamina lucida and dental cuticle were negatively reacting. The negative reactions of the basal lamina and dental cuticle after colloidal thorium demonstrate the lack of acidic carbohydrates in both layers. With ruthenium red and periodic acid-oxidation, the dental cuticle was denoted again to be negative for acidic carbohydrates. As for the reaction positive basal lamina with ruthenium red, we interpreted that the reaction was not due to acidic carbohydrates but potentially attributed to a protien moiety.
