    Notes: Abstract The feasibility of direct radioimmunoassay of unconjugated estradiol in dried serum extracts in the assessment of ovarian function in the regulation of the menstrual cycle has been investigated. Using an antiserum to an estradiol-6-conjugate, assay reliability was evaluated and the procedure standardized through a series of tests aimed at assessing accuracy, sensitivity, and precision. These included multiple titration with competing steroids, the definition of blank effects from solvent and sample, the assay both of samples to which known amounts of estradiol had been added and of serially diluted samples, and clinical validation using as a reference samples related to well-defined physiological situations. The variability of replicate estimates and the repeatability of the calibration curve were evaluated to obtain information on assay precision and sensitivity. The analytical performances proved to be perfectly adequate for the clinical purposes for which the assay was intended, in terms both of reliability of results and of methodologic practicability.

    Notes: Abstract Antisera to H4-lactate dehydrogenase (LDH) were elicited in rabbits, against both human (h) and porcine (p) isoenzymes. 125I-labelled H4-LDH was prepared by electrolytic iodination. A simple and fast procedure (1-h incubation for clinical assays) was set up by using polyethylene glycol for the bound-free separation. The results obtained in the antiserum characterization indicated that the heterologous homotetramer, M4 was completely discriminated in the porcine system while a weak cross-reaction with human antisera resulted. In both cases, for the hybrid forms, a crossreactivity level related to the stoichiometric contents of the H-subunit in the tetramers was observed. The H4-LDH from other species was found to be much more effectively disinguished in the porcine than in the human system. The assay for human LDH was further validated in terms of analytical suitability and clinical response. For healthy subjects the mean concentration was 0.46±0.19μg/ml (mean±SD). Patients with acute myocardial infarction had levels ranging from 1.2 to 5.9μg/ml.

    Notes: Abstract Two progesterone-tyraminyl (125I) derivatives were prepared from progesterone-11-hemisuccinate and progesterone-3-0(carboxymethyl)oxime, respectively. The analytical performances of these tracers in the progesterone-anti progesterone-11-hemisuccinyl-BSA system were compared with those of tritiated radioligand assumed as a reference. For all of the 11 antisera tested much higher affinity and titers were observed in the case of ‘homologous’ 125I tracer, which however was found to involve drastic sensitivity losses. On the contrary, quite acceptable dose-response curves and affinity and titers close to those related to tritiated progesterone resulted with the heterologous tracer. These results, interpreted in terms of native ligand-tracer competitivity for the antibody sites, led to the choice of the C3-tyraminyl derivative for the development of a radioimmunoassay method to be compared with the reference tritiated progesterone systems. The two procedures, including minor differences in incubation conditions and B/F separation, were validated for the direct assay of unpurified dried serum extracts. In both cases, satifactory levels of sensitivity, precision and accuracy were proved, the estimates obtained being in a good agreement with data from the literature. On the basis of the present results, some general consideration on the optimization of steroid radioimmunoassay with radioiodinated tracers are tentatively drawn.

    Notes: Abstract For lubricated slider bearing the elementary theory of lubrication does not allow to describe the pressure build up at the entrance section. In this work the whole flow field is subdivided into three regions: a first one, upstream of the entrance, where the fluid behaves like a perfect fluid; a second region, immediately downstream, where the two boundary layers gradually grow, extending up to a distance determined by means of a model based on analytic functions. The third region is described by Navier–Stokes and solved by adopting a shooting method. Numerical computations were carried out to determine the pressure distribution and the position of the center of pressure for different values of Reynolds number and pad angle. Sommario. La teoria elementare della lubrificazione nel caso del pattino piano lubrificato non è in grado di evidenziare il fenomeno della sovrappressione nella sezione di ingresso. Nel presente lavoro viene proposto un modello che suddivide il campo di moto in tre regioni: una prima a monte della sezione di ingresso, in cui il fluido si comporta come un fluido perfetto; una seconda regione immediatamente a valle, in cui si tiene conto del graduale sviluppo dello strato limite e la cui estensione è determinata tramite un modello basato sulle funzioni analitiche. La terza regione è studiata risolvendo l'equazione di Navier–Stokes con tecnica numerica basata su un metodo di shooting. In questo lavoro viene riportato l'andamento della pressione all'interno del cuscinetto e la posizione del centro di pressione per differenti valori del numero di Reynolds e per varie inclinazioni del pattino.
