    Description / Table of Contents: Summary The kinetics of erythropoietic and granulocytopoietic cell proliferation have been investigated in the same patient at two distinct stages: firstly in preleukemia presenting as pancytopenia with ineffective erythropoiesis, and secondly 2 years later in acute myelogenous leukemia. The method of investigation is based on determining the DNA synthesis rate of individual cells by means of quantitative14C-autoradiography after short-term incubation with14C-thymidine and fluorodeoxyuridine. Erythropoiesis was equally ineffective in the two stages, the rate of proliferation, however, slowed down towards the leukemic state. The production rate of myeloblasts was markedly reduced in preleukemia accompanied by a very low labelling index. In leukemia on the other hand the production rate was increased to such a degree that the leukemic myeoblast compartment is to be considered as prevailingly self-reproductive. The proliferation rate of myeloblasts was reduced already in preleukemia, and there was a further decrease in leukemia. From the point of view of cell kinetics the manifestation of leukemia in this patient is explained best by a change in the mode of proliferation: the myeloblasts change from steady state growth to behaving like an exponentially expanding population.
    Notes: Zusammenfassung Die Proliferationskinetik von Erythropoese und Granulopoese wurde bei einer Patientin in zwei verschiedenen Krankheitsstadien untersucht: zuerst im präleukämischen Stadium, das sich mit Panzytopenie mit ineffektiver Erythropoese darbot, und 2 Jahre später bei manifester akuter myeloischer Leukämie (AML). Die Untersuchungstechnik basiert auf der Bestimmung der DNS-Synthesegeschwindigkeit einzelner Zellen durch quantitative14C-Autoradiographie nach Kurzinkubation mit14C-Thymidin und Fluordesoxyuridin. In beiden Krankheitsstadien ergab sich eine in gleicher Stärke ineffektive Erythropoese, doch kam bei der AML noch eine verminderte proliferative Aktivität der einzelnen Erythroblasten hinzu. In der präleukämischen Phase waren der Umfang der Neubildung von Myeloblasten stark vermindert und der Markierungsindex erniedrigt. Demgegenüber war die Neubildungsrate von Myeloblasten im leukämischen Stadium so stark erhöht, daß dieses Zellkompartiment als überwiegend selbst-reproduktiv anzusehen ist. Dabei fand sich bereits bei der Präleukämie eine verminderte Proliferationsgeschwindigkeit der einzelnen Myeloblasten, ein Verhalten, das sich zur Leukämie hin verstärkte. Aus dem Blickwinkel der Proliferationskinetik ist die klinische Manifestation der Leukämie dieser Patientin am besten durch eine Änderung des Proliferationsmodus erklärbar: Aus einem Gleichgewicht zwischen Neubildung und Differenzierung (steady state-Wachstum) gehen die Myeloblasten zu einem teilweise exponentiellen Wachstum über.

    Description / Table of Contents: Zusammenfassung Das nach der Energie seiner β-Zerfälle zwischen Tritium und Kohlenstoff-14 liegende Isotop 125J wurde auf seine Eignung zur quantitativen Autoradiographie geprüft. Absorption und Geometrie-Faktoren der radioaktiven Strahlung wurden untersucht. Hieraus ließen sich geeignete Meßbedingungen entwickeln. Durch gleichzeitige Exposition von radioaktiven Referenzquellen können absolute Radioaktivitätsmengen ermittelt werden. Als Referenzquellen sind membranmarkierte Standardzellen geeignet, die den physikalischen Eigenschaften des Isotopes Rechnung tragen. Hierzu wurden enzymatisch radiojodinierte Schaferythrozyten auf ihre Eignung geprüft. Die absolute Zahl von antigenen Stoffen auf den Oberflächen einzelner Zellen erhält man, wenn man die spezifische Aktivität der markierten Antikörpermoleküle bestimmt und die Silberkorndichte der zu untersuchenden Zellen und der Standardzellen mißt. Die Radioaktivität pro Standardzelle wird in herkömmlicher Weise ermittelt. Die neue Methode wurde zur Quantifizierung membrangebundener Immunglobulinmoleküle vom IgG-Typ auf einzelnen menschlichen Lymphozyten angewendet. Hierbei ist die Ermittlung einer immunologischen Sättigung des markierten Antikörpers wesentlich. Auf Lymphozyten von Normalpersonen und von chronisch-lymphatischen Leukämiepatienten konnten sehr unterschiedliche absolute Immunglobulinmengen bestimmt werden.
    Notes: Summary Iodine 125, an emitter of β-radiation with an energy lying between that of tritium and carbon-14, is investigated for its applicability in quantitative autoradiography. Absorption and geometric factors of radiation are elucidated. From this, appropriate measuring conditions are derived. The simultaneous exposure of radioactive standard sources permits the evaluation of absolute amounts of radioactivity. Standard cells with labelled membranes are a suitable source of reference taking into account the physical properties of the isotope. Sheep red blood cells are examined for their suitability as standard cells after enzymatic radioiodination. The absolute number of antigenic substances on the surface of single cells is obtained by determining the specific activity of the labelled antibody molecules, and by measuring the silver grain densities of the cells under investigation and of the standard cells. The radioactivity per standard cell can be assessed by conventional procedures. The new method is applied to the quantification of membrane-bound immunoglobulin molecules of the IgG-type on single human lymphocytes. The determination of an immunologic saturation of the labelled antibody is essential for this purpose. On the lymphocytes of a normal person and of a patient with chronic lymphatic leukaemia quite different amounts of immunoglobulins have been evaluated.

    Notes: Summary An automated microphotometric system has been designed for the successive assessment of a maximum number of parameters from individual cells. The hardware and software elements are described as well as the various photometric functions that can be performed. The aim is to obtain a multiparameter analysis of morphology, cytochemistry, cell surface characteristics, and metabolic activity including cell cycle characteristics for any set of cells selected from a sample. The combination of DNA synthesis rate determination in a really quantitative approach with the analysis of features of nuclear chromatin texture opens up a new field for associating morphology with functional properties. Examples of application are presented to demonstrate the applicability of the system in a heterogeneous cell population of a very limited sample size. For this purpose, human peripheral blood cells grown in diffusion chambers in preirradiated mice were investigated. It is shown that, depending on their functions, different cell types may have quite different nuclear-to-cell area ratios. Further, a crude morphological parameter, such as the degree of basophilia of Giemsa-stained cells, may be associated with the rate of DNA synthesis, thus bearing information on the replicative activity of a cell. Cell surface properties related to the leukemia-associated cALL antigen are investigated in a human-derived cell line and correlated with cell-cycle characteristics. It is concluded that different antigenic sites rather than cell-cycle dependent differences of antigen density account for the outcome of a bimodal distribution.
