    Notes: Abstract Self-transmissible plasmids carryinghis andnif genes fromKlebsiella pneumoniae have been introduced into threehis mutants ofProteus mirabilis: strains 5006-1, WR19 and WR20. Expression ofhis by the transconjugants was unequivocal, if slightly temperature-sensitive, but none was Nif+ when tested for acetylene reduction in anaerobic glucose medium using inocula from rich or glucose-minimal aerobic agar cultures. Succinate or pyruvate in place of glucose, low glucose, lower temperature or elevated Na2MoO4 did not allownif expression and no nitrogenase MoFe-protein peptide was detected immunologically after exposure to conditions in which diazotrophic enterobacteria, normal or genetically constructed, derepressnif. One strain,P. mirabilis WR19, carrying thehis nif Kmr plasmid pMF250 was examined in detail. Thenif activator genenifA was introduced on the plasmid pCK1. Such derivatives remained Nif- when tested, after aerobic growth on rich agar media, with normal or low glucose, with succinate or with elevated Mo. However, pre-conditioning by aerobic growth on glucose-minimal agar led to subsequent anaerobic expression ofnif in glucose medium from pMF250 in WR19 carrying pCK1. NH 4 + or proline could serve as N-source in the glucose-minimal agar. Maximum activity was about 5% of that ofK. pneumoniae in our assay conditions. Material cross-reacting with anti-serum to the nitrogenase MoFe protein was formed. Nitrogenase activity was not ‘switched off’ by NH 4 + .P. mirabilis WR19 (pCK1) showed NH 4 + -constitutive temperature-sensitive kanamycin resistance (anif-related phenotype of this plasmid) in aerobic glucose minimal medium. Expression ofnif inP. mirabilis WR19 (pCK1, pMF250) was NH 4 + -repressible despite the constitutivenifA character of pCK1 and introduction of thentrA + plasmid pMM17 did not alter this phenotype. However, pCK1 did not give rise to NH 4 + -constitutive diazotrophy in the wild-typeK. pneumoniae M5al. A construct of WR19 carrying pMF250 and constitutiventrC plasmid (pMD45) remained Nif- even after pre-growth on glucose-minimal media. We conclude (a) thatP. mirabilis forms a gene product functionally equivalent to that ofntrA inK. pneumoniae, (b) that it forms no functional equivalent of thentrC product in our growth conditions. The need for pre-conditioning on aerobic glucose media remains perplexing.

    Notes: The effects of five kinds of questioning, two interpersonal atmospheres of interviewing, and five levels of item difficulty on the accuracy and completeness of testimony about a short film were tested in a legal interrogation setting. Subjects enjoyed the supportive style of interviewing more than the challenging style, but atmosphere had no important effect on recall performance.The type of questioning produced almost no differences in affective or cognitive reactions. However, as the specificity of questions increased, so did the completeness of testimony. Accuracy of testimony showed slight decreases for more specific questions. The trade-off between accuracy and completeness was mediated by item difficulty. It was very pronounced for items of high difficulty and not apparent for items of low difficulty.Leading questions by themselves or in interaction with atmosphere did not produce special distortions in accuracy.

    Notes: Summary The relationship of diet and phylogeny to the intrinsic rate of population increase (r max) was examined in a sample of 39 mammalian species that live in Neotropical forests. Diets of species did not predict their r max, contrary to published predictions based on associations between basal metabolic rate and diet and between basal metabolic rate and r max. Phylogeny did however, apparently because life history characteristics and susceptibility to predation vary predictably with phylogeny and with one another.

    Notes: Abstract Rats and mice were exposed for periods of up to six months and two successive generations of mice were raised in a ground-level chamber system filled with 80% helium −20% oxygen, at 24°C. A duplicate chamber for controls contained a comparable nitrogen-oxygen mixture, and in both the other environmental parameters were well-controlled and nearly identical. Animals adapted to helium showed no greater increase in oxygen consumption (P〉0.05) when placed in helium-oxygen than did those raised in air. Growth rates were identical, but the helium mice consumed more food and water. Selected biochemical analyses were made on the parent and two successive generations of mice. These included blood indices; electrophoretically separated tissue protein patterns from liver, skeletal muscle, and cardiac muscle; quantitative determinations of LDH, MDH, and G6PDH from the same tissues; serum insulin; and semi-quantitative histochemical estimates of liver glycogen. No cases of statistically significant difference or consistent trends were seen between the experimental environmental groups. Additional analyses of liver nucleotides and redox-coenzymes also failed to show a significant difference. The relative weights of liver, heart, kidney, and diaphragm (wet and dry) were the same in both groups. Histopathological examination of kidney and adrenal tissue produced unremarkable findings and none that were attributable to the nature of the gaseous environment. It must be concluded that prolonged exposure to helium-oxygen, relative to air, does not produce detectable changes in several key subcellular factors which might be altered by serious metabolic disturbances, and therefore the helium exposure is well tolerated.
