    Notizen: SYNOPSIS. Of several methods developed for cryopreservation of Trypanosoma vivax, Trypanosoma congolense, and Trypanosoma brucei metacyclic forms in tsetse fly organs, as well as bloodstream forms in host blood, one proved the most satisfactory. In this method, infected fly proboscises and salivary glands were placed in glass capillary tubes containing fetal calf serum with 8% (v/v) glycerol as the cryoprotectant. The method for bloodstream forms involved the addition of glycerol directly to infected blood, which was then dispensed into capillary tubes. Next the tubes were placed in paper containers inside a glass test tube with a 5 mm thick plasticine jacket. The insulated assembly was suspended in the liquid nitrogen vapor phase in an LR-35 (Union Carbide) refrigerator for 45 min. Under these conditions, the cooling rate was 2 C/min. The frozen samples were transferred to permanent storage. The viability and infectivity of the preserved organisms were found to be satisfactory upon testing, and no antigenic changes were observed. Laboratory and field applications of the method are discussed.

    Notizen: The value of the residual R2 = ((I1 - I2)2)/(I21), where I1 and I2 are the intensities of reflexion for two unrelated structures having the same symmetry and containing the same atoms, has the value 1 - Σ4/(2Σ2 - Σ4) for non-centrosymmetric structures and 4/3 - 2Σ4/3(Σ2 - Σ4) for centrosymmetric; the corresponding values for R1 = ((F1 - F2)2)/(F21) are approximately 2(1 - π/4) ∼ 0.43 and 2(1 - 2/π) ∼ 0.73. More complex expressions are derived for hyper- and sesquisymmetric structures. If a residual with a scaling factor, such as S2 = ((I1 - EI2)2)/(I21), is used, and the scaling factor E is refined by least-squares, the value of E obtained is about ½ or ½, instead of the true value unity.

The slope apparent particle size is half the harmonic mean of h\midsec φ\mid and b\midcsc φ\mid, where h is the height of the cylinder or prism, b is the breadth of a rectangle having an area equal to that of its top face and a width equal to that of its radiation shadow, and φ is the angle between its axis and the diffraction vector. For cylinders, but not in general for prisms, the intercept apparent particle size is 1/√2 times the geometric mean of the same quantities. For a hemisphere of diameter d the slope apparent particle size is given by 2\over 3d/(1 + \midcos φ\mid), where φ is the angle between the diffraction vector and the normal to the plane face, and the intercept apparent particle size is the square root of 1\over 6πd2\midcsc 2φ\mid.

    Notizen: Reduction of random experimental errors and accounting for systematic errors require good experimental techniques and systematic handling of data. For the Straumanis–Ievinš, van Arkel, and Bradley–Jay methods, in the absence of error, the sum of the readings of the measuring instrument for corresponding lines on either side of the exit (or entrance) hole would be constant. Drift of this sum with &thgr; reveals certain types of systematic error resulting from geometrical aberrations having low symmetry; random scatter about the drift line gives an immediate measure of the random error in setting on a line. Unfortunately no such internal assessment is possible with the usual Guinier or diffractometer arrangements. The standard deviation of a measurement of line position is found to be show little variation with &thgr;, though there is some indication that it is dependent on line intensity, being greater for very weak and very strong lines. The standard deviations of Bragg angles and lattice spacings or parameters are easily determined from that of the position measurements.
