    Notes: Direct measurements of the methylation of newly-synthesized DNA were made in cultures of a clonal mouse adrenal cortex cell line, Y129OS3, by (1) following the incorporation of radioactivity from methionine-(methyl)-C14 into a segment of DNA which had been density-labeled with bromouracil and (2) labeling DNA cytosine with C14-deoxycytidine and then following the appearance of radioactivity in DNA 5-methylcytosine. The results establish that during exponential growth the DNA of this cell line is methylated entirely within a few minutes of its synthesis. Using the second technique described above accurate, sensitive measurements of DNA methylation levels can be made by comparing radioactivity in 5-methylcytosine to radioactivity in cytosine plus 5-methylcytosine. In this cell line 5-methylcytosine accounts for 4.3 ± 0.2% of the DNA cytosine. Some apparent contradictions between these results and those of other workers are discussed.

    Notes: The calcium uptake and ATPase activities of isolated sarcoplasmic reticulum were studied during the first six days of chick skeletal muscle maturation in tissue culture. Statistically significant increases in these activities were observed between the second and the sixth day of maturation. Increases in oxalate-dependent calcium uptake were demonstrated at concentrations of 2.5 × 10-5 M calcium and 10-4 M calcium. Calcium-binding determinations conducted in the absence of oxalate displayed changes manifested by an increase at day 5 followed by a significant decrease at day 6. Increases in total ATPase activity during maturation paralleled the sequential increases in calcium uptake. Calcium-stimulated ATPase activity, however, did not change significantly during periods of marked increase in calcium uptake, suggesting that these activities are dissociated during development of the sarcoplasmic reticulum. These data demonstrate that calcium uptake and total ATPase activity increase during muscle maturation in tissue culture and that these activities are present prior to spontaneous contractions.

    Notes: If the aging of diploid human fibroblasts reflects stable genetic or epigenetic changes which are few in number and different in different cells, then complementation could occur in hybrids between individual senescent cells. However neither pairs of aged fibroblasts nor even pairs of young and senescent fibroblasts produce viable hybrids under conditions known to promote cell fusion.

    Notes: Using a new technique for the determination of the extent of DNA methylation by comparing the incorporation of radioactivity from deoxycytidine-2-C14 into DNA cytosine and 5-methylcytosine (5MC), the 5MC content of DNA was found to be a tissue specific property in two systems: (1) mouse tissue culture cell lines, in which the level of 5MC varied from 2.97 ± 0.03% conversion of cytosine to 5MC in the DNA of a mouse melanoma line to 4.58 ± 0.18% in a primary teratoma line, and (2) the organs of the developing chick embryo, in which the level of 5MC varied from 3.60 ± 0.04% conversion of cytosine to 5MC in the DNA of the chorioallantoic membrane to 4.19 ± 0.02% in the brain.

    Notes: In muskellunge blastulae the yolk sac syncytium originally contains nuclei comparable in size to the blastomere nuclei from which the syncytial nuclei arose. By mitosis the originally diploid syncytial nuclei become numerous and crowded together. Continued synchronous mitosis of neighboring syncytial nuclei and the resultant crowding together of their spindles culminates in overlapping spindles, multipolar spindles, disorganized spindles, and the crowding together of large numbers of condensed chromosomes from contigous spindles. When such an aggregation of condensed chromosomes becomes enclosed within one nuclear membrane, a giant nucleus appears in the following interphase. It soon becomes postmitotic.
