    Notes: Abstract Six species of benthic diatoms and a natural benthic diatom community were cultured in flasks on a variety of sediments. Diatom species which secreted large quantities of mucilage were effective sediment stabilizers. These mucilage-secreting species significantly reduced resuspension and retarded laminar flow of the sediments when the culture flasks were agitated. Diatom species which secreted little or no mucilage were not effective sediment stabilizers. These non-mucilage-secreting species did not significantly effect resuspension or laminar flow of the sediments when the culture flasks were agitated. A sediment stabilizing mechanism based on the secretion of mucilage by pennate benthic diatoms is proposed. The effect such a process may have on distributional patterns of benthic invertebrates in areas where extensive diatom or other microalgal films occur is discussed.

    Notes: Summary 1. Various fixation and embedding procedures were tested on the retention of immunofluorescent staining. Intact hyphae of Ophiobolus graminis Sacc. were fixed in various aldehyde solution both before and after indirect immunofluorescent staining. Paraformaldehyde and glutaraldehyde solutions did not affect the intensity of fluorescence while acrolein quenched fluorescence slightly. 2. Immunofluorescent stained hyphae fixed in 3% glutaraldehyde, dehydrated, embedded in glycol methacrylate and sectioned at 1–2 μm retained fluorescence allowing more precise location of the antigen.

    Notes: Abstract Antigens from four cultures ofO. graminis were compared immunoelectro-phoretically. Each culture produced a characteristic immunogram. More common antigens were found between the two cultures isolated from wheat or the two cultures isolated from oat than between a wheat and an oat isolate. Cell-wall antigens were the best reference antigens for serologic analysis of strain relationship. O. graminis antisera were cross-reacted with antigens from a number of other species of fungi. Relatively few of these cross-reacted with antisera to cell-wall antigens whereas more cross-reacted with antisera to whole-cell antigens. Immunoelectrophoretic analysis of antigens from a range of isolates ofO. graminis indicates specific immunograms which can be determined and separated from the immunograms developed by all other fungi when tested againstO. graminis antiserum. Immunoelectrophoresis can therefore be used as an aid in determiningO. graminis.

    Notes: Abstract Fluorescein isothiocyanate (FITC) conjugates were prepared to whole-cell and cell-wall fractions of four cultures ofOphiobolus graminis W1, W2, O1 and O2. Homologous and heterologous direct and indirect FA staining of the four cultures ofO. graminis gave positive staining in all reactions. This indicated that the four cultures could not be differentiated by fluorescent antibody (FA) staining. Species specificity of the conjugates was shown by the staining ofO. graminis hyphae in the rhizosphere of wheat and oat roots. Plant tissues were not stained. Furthermore out of 52 rhizosphere isolates stained with whole-cell and cell-wall conjugates of the four cultures ofO. graminis, only 7 cross-reacted with the whole-cell conjugates whereas none cross-reacted with the cell-wall conjugates. These results indicate the potentiality of the FA staining technique as a serological tool in localizing, and identifyingO. graminis amongst mixed fungal populations in the rhizosphere of roots.

    Notes: Abstract Immunological methods for distinguishing rod from coccus forms of anArthrobacter species are presented. Diffusion precipitation tests indicate that the rod form is antigenically more complex than the coccus form with respect to both soluble and cell-wall fractions. Antisera prepared against fractions of coccus and rod forms are able, after cross absorption, to specifically label rod and coccus forms.
