Summary Transfection with marker rescue techniques provides a sensitive assay for the DNA of phage SPP1. The method was used in a study of the process of replication of this phage. The data show that early during the eclipse, SPP1 DNA increases exponentially, suggesting a geometric mode of replication. DNA replication ceases at the time of the appearance of the first intracellular phage particles.
