    Description / Table of Contents: Zusammenfassung Der Vorteil der vorliegenden Perfusionsstudie des Pankreas in vitro ist der, daß er eine Vorhersage der funktionellen Lebensfähigkeit des perfundierten Pankreas in direkter Beziehung zur spezifischen Organfunktion erlaubt. Das Pankreas mit Duodenum wurde Hunden entnommen und für 3 Std mit einer Blut-Ringer-Lactatlösung mit einem Membranoxygenator, einer Pulspumpe und einem Wärmeaustauscher bei 37°C perfundiert. Aufgrund der vorliegenden Untersuchungen erweisen sich der Insulinausstoß nach Glucosegabe und die Pankreassekretion nach Sekretin-Reiz als besonders zuverlässige Kriterien in der Beurteilung der Lebensfähigkeit des Organs. Der durchschnittliche Sauerstoffverbrauch des Pankreas liegt bei 0,77 ml 02/100 g. Ein durchschnittlicher Flow von 80 ml/min bei einem Druck von 75 mm Hg erweist sich in der vorliegenden Versuchsanordnung als nützlich. Makroskopische und histologische Untersuchungen bestätigen die Ergebnisse.
    Notes: Summary An advantage of the current perfusion-test in vitro is that it allows prediction of the functional viability of the perfused pancreas in direct relation to the specific organ function. The canine pancreas including the duodenum has been perfused for 3 h with a membrane oxygenator, a pulsatile pump and a heat exchanger at a temperature of 37°C. Among the criteria used in this study the insulin-output into the venous blood in response to glucose administration as well as pancreassecretion after secretin stimulation were found to be the most reliable indications as to the viability of the pancreas. This is further supported by an average oxygen consumption of 0.77 ml 02/100 g at a constant flow of 80 ml/min and a pressure of 75 mm Hg, and the macroscopical and histological findings.

    Notes: Summary The buccal absorption of procainamide was studied in 4 normal, male volunteers over a pH range of 5–11. In another investigation the same 4 volunteers collected their urine for 24 h after taking 250 mg of procainamide hydrochloride orally on 3 separate occasions, during which urine pH was uncontrolled, acidified by ammonium chloride ingestion or alkalinised by sodium bicarbonate ingestion. The procainamide remaining after buccal absorption and present in the urine samples was determined spectrophotometrically. It was found that neither the buccal absorption nor the renal excretion of procainamide was substantially altered by changes in pH.

    Description / Table of Contents: Zusammenfassung Vier Gruppen mit insgesamt 124 Wistar-Ratten (Körpergewicht 100–120 g) erhielten im Bereich der Schwanzwurzel eine intracutane Injektion von 0,08 — 0,1 ml kompletten Freundschen Adjuvans. Eine zweite gleichartige Injektion wurde bei je einer Gruppe 25, 39, 52 bzw. 101 Tage nach der ersten Applikation vorgenommen. Alle Tiere wurden vom Tage der Erstinjektion bis mindestens 50 Tage post reinjectionem regelmäßig mikroskopisch auf extraoculare und spaltlampenmikroskopisch auf Augenveränderungen untersucht. Folgende Ergebnisse wurden erzielt: 1. Nach Reinjektion war die Latenzzeit arthritischer Veränderungen verkürzt. 2. Frühe Reinjektion führte neben einem unterhaltenden Effekt auf die Arthritis zu einer Vermehrung der Uveitis-Fälle und größerer Rezidiv-Häufigkeit. Der Anteil der Primär-Uveitiden war höher als der Anteil der Zweiterkrankungen. 3. Späte Reinjektion (52. bzw. 101. Tag p.i.) führte zu einem milderen Arthritis-Verlauf, wobei die Anzahl der erneut betroffenen Gelenke die Zahl erstmalig reagierender Gelenke erreichte oder deutlich übertraf. Uveitiden waren weniger häufig und weniger stark remittierend aber nicht milder als nach Erstinjektion. Die relative Häufigkeit von Zweiterkrankungen war indessen wesentlich höher als die der Primär-Uveitiden. Der Gedanke an einen lokal disponierenden Einfluß des Primäreffekts liegt nahe.
    Notes: Summary Four groups of 124 Wistar rats altogether (average body weight 100–120g) received intracutaneous injections of 0.08–0.1 ml Freund's complete adjuvant in the region of the tail head. A second injection of the same kind was given 25, 39, 52 or 101 days after the first one. From administration of the first injection until at least 50 days post reinjectionem all animals were examined macroscopically for extra-ocular lesions and biomicroscopically for eye lesions at frequent regular intervals. Finally serial sections of 44 eyes were examined by light microscopy. The following results were obtained: 1. After reinjection the latent period of arthritic changes was reduced. 2. Early reinjection, besides maintaining arthritis caused a higher incidence of uveitis and relapse. There was a higher proportion of primary uveitis than of recurrence. 3. Late reinjection (days 52 and 101 post injectionem) was followed by a milder course of arthritis, while the number of joints reacting for the second time equalled or distinctly exceeded that of joints inflamed for the first time. Cases of uveitis were less frequent and less prone to temporary remission but were no milder than those following the first injection. The frequency of recurrence of lesions was, however, significantly higher than that of primary uveitis. It may be that the primary effect predisposes the area to recurrence.

    Description / Table of Contents: Zusammenfassung Groß-Chinchilla-Kaninchen (1,5–2,5 kg KG) erhielten interscapulär eine einzige Injektion von respektive: a) Einer Emulsion von 50 mg (Naßgewicht) frischer homologer Retina in 0,4 ml kompletten Freundschen Adjuvans (110 Tiere); b) einer Suspension von 50 mg homogenisierter, frischer homologer Retina in 0,4 ml physiologischer Kochsalzlösung (10 Tiere); c) einer Emulsion von 50 mg frischer homologer Retina in 0,4 ml inkompletten Freundschen Adjuvans (10 Tiere); d) 0,4 ml kompletten Freundschen Adjuvans ohne Retina (20 Tiere). Vor Beginn des Versuchs wurden alle Kaninchen und post injectionem 90 Versuchs-sowie sämtliche Kontrolltiere über 40 Tage täglich auf Augenveränderungen (Ophthalmoskop, Spaltleuchte) und neurologische Ausfälle untersucht. 63 von 90 über die volle Dauer des Experiments beobachteten Tiere reagierten nach einer Latenzzeit von 12–39 Tagen mit einer Chorioretinitis. Die schwersten Entzündungen traten in der Regel während der 3. Woche post injectionem auf und erreichten binnen 60 Std ihr Maximum. Die klinischen Bilder der Erkrankung werden beschrieben und in drei Erscheinungsformen gruppiert. Klinische Zeichen einer Beteiligung der vorderen Augenabschnitte oder des ZNS wurden nicht gefunden. Die beschriebenen Veränderungen waren ausschließlich nach Injektion homologer Retina in komplettem Freundschen Adjuvans, jedoch nicht nach Injektion der Kontrollpräparate b, c, und d zu beobachten.
    Notes: Summary Great Chinchilla rabbits (body weight 1.5–2.5 kg) were given interscapularly a single immunizing dose of, respectively: a) 50 mg (wet weight) of freshly dissected homologous retina emulsified in 0.4 ml of Freund's complete adjuvant (110 animals); b) 50 mg homologous retina as above incorporated in 0.4 ml saline (10 animals); c) 50 mg homologous retina as above incorporated in 0.4 ml of Freund's incomplete adjuvant (10 animals); d) 0.4 ml Freund's adjuvant (20 animals). Before starting the experiment, and at daily intervals thereafter, all animals were examined for eye lesions (ophthalmoscope, slit lamp) and for neurological deficits. In 90 experimental and in all control animals these examinations were carried out over a period of 40 days. 63 of 93 experimental animals developed ocular lesions characterized by chorioretinic patches of varying size in all parts of the fundus. The time of disease onset varied from 12 to 39 days after immunization. The most violent inflammations appeared as a rule in the third week after inoculation and reached maximum severity within 60 hours. Three types of clinical pictures are described. No involvement of the anterior segment or of the central nervous system was noted clinically in any of the animals. In contrast to the control preparations tested (b, c, d), only homologous retina in Freund's complete adjuvant was effective.

    Description / Table of Contents: Zusammenfassung Die vorliegende Arbeit beschreibt die Histopathologie der experimentell allergischen Chorioretinitis des Kaninchens (Groß-Chinchilla-Kaninchen: 51 Versuchs- und 10 Kontrolltiere). Chorioretinitische Veränderungen wurden durch einmalige interscapuläre i.c.-Injektion einer Emulsion von 50 mg frischer homologer Retina in 0,4 ml kompletten Freundschen Adjuvans erzeugt. Den Kontrolltieren applizierten wir analog 0,4 ml kompletten Freundschen Adjuvans ohne Retina. Die klinischen Erscheinungsformen und Phasen der Erkrankung wurden anhand von Serien- und Stufenschnitten (Fixierung: Formol 1∶9, Zenker Paraffinschnitte 8–10 μm Färbungen: H.-E., PAS, van Gieson, Panchrom-Kardos, Toluidinblau) lichtmikroskopisch untersucht. Das histologische Bild war durch überwiegend histiomonocytäre Zellinfiltration Von Chorioidea und Retina gekennzeichnet. Die Entzündung führte lokal zur teilweisen oder völligen Destruktion dieser Schichten. Unspezifischen Riesenzellen zwischen Aderhaut und Retina, blasige Auftreibung von Zellen des Pigmentepithels, „subretinale Zellen“ sowie keulenförmige Bildungen anstelle der Stäbchen- und Zapfenschicht werden u.a. beschrieben. Mononucleäre Zellen innerhalb und außerhalb der Netzhautgefäße (Vasculitis), im Bereich der vitreoretinalen Grenzschicht und im rindennahen Glaskörper waren häufig. Die histopathologischen Verhältnisse weisen auf vorwiegend zellgebundene immunologische Prozesse hin. Sie geben Anlaß zur Diskussion der Lokalisation von Antigen und Primärschaden.
    Notes: Summary This paper describes the histopathology of experimental allergic chorioretinitis in rabbits of the Great Chinchilla strain (51 experimental animals, 10 controls). Chorioretinitic lesions were induced by a single intracutaneous, interscapularly placed injection of 50 mg (wet weight) freshly dissected homologous retina, emulsified in 0.4 ml of Freund's complete adjuvant. In the same way control animals got 0.4 ml of Freund's complete adjuvant only. The types of clinical pictures as well as various phases of the disease described previously were examined microscopically by means of serial sections (Fixatives: Formalin 1∶9, Zenker's fluid. Paraffin sections 8–10 μm. Staining: H.-E., van Gieson, Panchrom-Kardos, Toluidine blue). The histologic pictures were characterized by predominant histio-monocytoid cellular infiltration of choroid and retina. Inflammation locally caused partial or complete destruction of these layers. Other findings were: unspecific giant cells between choroid and retina, blister-like swelling of pigmentary epithelial cells, “subretinal cells”, and club-like formations in lieu of the rod and cone layers. Mononuclear cells within and around the papillary vessels (vasculitis), in the vitreoretinal boundary, and within the outer zones of the vitreous were seen frequently. The histopathologic conditions point to predominantly cellularly linked immunological processes. They give rise to discussion of the localizations of antigen and primary lesion.

    Description / Table of Contents: Zusammenfassung An einem experimentell erzeugten Hornhautödem bzw. an normalen Hornhäuten von Kaninchen wird die Wirkung von Glucose- und Glycerinlösungen unterschiedlicher Konzentration verglichen. Als Maß des Quellungsgrades dient die Hornhautdicke, die mit einem Meßgerät eigener Anfertigung in Anlehnung an das Gerät von Jäger registriert wird. Es wurden folgende Ergebnisse erzielt: 1. Die normale Kaninchenhornhaut konnte sowohl durch Glucose- als auch durch Glycerinlösung bis etwa auf 50% der Normaldicke entquollen werden. 2. Glucoselösungen wirkten auf das experimentell erzeugte Hornhautödem stärker entquellend als glycerinlösungen gleicher Osmolarität. 3. Eine geringe regenerationsfördernde Wirkung auf das Corneaendothel konnte bei Glycerinbehandlung deutlicher nachgewiesen werden als bei Glucoselösung vergleichbarer Konzentration. Aus den Ergebnissen folgt, daß sich Glycerin ebenso wie Glucose zur Entquellung eines Hornhautödems eignet. Eine Entscheidung über die Verwendbarkeit von Glycerinlösung beim Hornhautödem des Menschen erfordert weitere Untersuchungen.
    Notes: Summary The effect of glucose and glycerine solutions of different concentrations was examined on a corneal oedema produced experimentally and on normal corneae of rabbits, respectively. The swollen cornea was measured by means of a home-made device similar to that described by Jäger! The following results were obtained: 1. By instillation of glucose as well as glycerine solutions it was possible to reduce the thickness of the normal rabbit cornea to 50% of the original value. 2. Glucose solutions showed a more dehydrating effect on the experimentally produced corneal oedema than glycerine solutions of the same osmolarity. 3. A slight effect favouring the regeneration of the corneal endothelium could be seen more distinctly after treatment with glycerine than after the instillation of glucose solutions of comparable concentrations. It is concluded that glycerine and glucose are equally suitable for dehydrating a corneal oedema. Further investigations, however, are necessary before glycerine solutions can be used in human corneal oedemata.

Mitteilung an und führt nach statistisch orientierter Auswertung der klinischen Befunde zu folgenden Ergebnissen: 1. Junge Wistar-Ratten (mittleres Körpergewicht 119 g) reagierten auf einmalige intracutane Injektion von 0,08–0,1 ml kompletten Freundschen Adjuvans zu 76,1% mit Arthritis, zu 79,5% mit Augenentzündungen, zu 51,6% mit Ohrknötchen und zu 58,7% mit Schwanzknoten. 2. Nach Latenzzeiten zwischen 1 und 80 Tagen trat bei 67,7% der untersuchten Augen eine Uveitis anterior auf. Bei 21 Tieren blieb sie auf ein Auge beschränkt. In 37 von 49 Fällen beidseitiger Affektion lagen zwischen dem Entzündungsbeginn von rechtem und linkem Auge Intervalle von 1–37 Tagen. 3. Bei 58 von 119 Augen (48,7%) mit einer Uveitis anterior kam es im Verlauf von 101–102 Tagen zu klinisch sicheren Remissionen. 35 Fälle mit einem, 14 mit zwei und 9 Fälle mit drei Rezidiven wurden beobachtet. Beginn. Dauer und Schweregrad der Schübe gleicher Ordnung sowie die Dauer entzündungsfreier Intervalle werden, abgestuft nach der Schubzahl der Verläufe, in Form arithmetischer Mittel angegeben. Remissionslose Entzündungen traten meist später auf und hielten länger an als die ersten Schübe rezidivierender Entzündungen. Die Entzündungsschübe I–III begannen jeweils in Abständen von etwa 2 Wochen, der Schub IV bereits nach einem Intervall von 8–9 Tagen. Eine Tendenz zu rascherem Abklingen in der Reihe aufeinanderfolgender Entzündungsschübe war nicht generell von einer Milderung der Symptome begleitet. 4. Mit der Arthritis als augenfälligstem Symptom der Adjuvans-Krankheit zeigten sich die Augenveränderungen eng, aber nicht zwingend, Ohrknötchen indessen als nahezu obligatorisch assoziiert. Als einziges Zeichen einer Adjuvans-Krankheit wurden Augenentzündungen dreimal häufiger beobachtet als Arthritis.
    Notes: Summary The present publication is a continuation of our first communication. A statistically orientated valuation of clinical findings leads to the following results: 1. Young Wistar rats (average body weight 119 g) responded to a single intracutaneous injection of 0.08–0.1 ml of Freund's complete adjuvant by developing several symptoms of adjuvant disease. 76.1% of the animals developed arthritis, 79.5% inflammations of the eyes and 51.6% and 58.7% respectively showed nodular alterations in external ears and tail. 2. After a latent period from 1 to 80 days 67.7% of the eyes examined developed iridocyclitic changes. In 21 animals inflammation was limited to one eyeball. In 37 out of 49 cases of bilateral affection the interval, between onset of inflammation in the right and the left eyes varied between 1 and 37 days. 3. During a period of 101–102 days clinically assured recidivations were demonstrated in 58 out of 119 eyes (48.7%) showing iridocyclitis. 35 cases showed one, 14 two and 9 cases three recidivations. Onset, duration and severity of batches of the same rank as well as the duration of intervals without inflammatory signs —graduated according to the number of batches per course—are stated as arith-metical means. Non-relapsing inflammations mostly began later and lasted longer than the first batches of relapsing inflammations. The inflammatory batches II and III started after an interval of 2 weeks, and the batch IV already after an interval of 8–9 days. A tendency to quicker improvement in the series of successive batches was not generally accompanied by a moderation of symtoms. 4. Eye lesions showed a close but no obligatory association with the arthritis as the most obvious symptom of adjuvant disease. The development of nodular changes in the external ears, however, seemed bound to the presence of arthritis. As the only sign of an adjuvant disease eye lesions were observed three times more frequently than arthritis.

    Description / Table of Contents: Zusammenfassung An 10 gesunden Studenten wird das Verhalten von Blutdruck, Herzfrequenz, EKG, Gewebedurchblutung, Pupillenweite und oculärer Tension nach lokaler Applikation von 3 Tropfen 10%igem Neosynephrin-POS in den Bindehautsack eines Auges untersucht. Eine signifikante Änderung der Kreislaufgrößen konnte nicht beobachtet werden. Die Frage der allgemeinen Nebenwirkungen wird diskutiert und zusammenfassend verneint.
    Notes: Summary In 10 healthy students the response of blood pressure, heart rate, ECG, blood-supply, diameter of the pupil and ocular tension have been studied after application of 3 drops of neosynephrine (10%) into the cul-de-sac of one eye. The results showed no significant changes of circulation parameters. The question of side-effects is discussed and in general the answers are negative.
