Zellstabilisatoren zur Perfusionslösung (Chlorpromazin 12 mg/l, Ultracorten 50 mg/l and Mg++ 4 mval); Gruppe III: Initialperfusion wie bei II and anschließend 3stündlich intermittierende Kurzzeitperfusion mit der gleichen Lösung während der hypothermen Lagerung; Gruppe IV wie III, jedoch mit 1 stündlich intermittierender Perfusion. - Schwerwiegende ultrastrukturelle Veränderungen wie Desorganisation and Vesikulation des endo. plasmatischen Reticulums, Mitochondrienschwellung, Margination des Chromatins und Rupturen des Plasmalemms mit Austritt von Cytoplasmabestandteilen in die Disseschen Räume traten bei Gruppe I and II bereits nach 6 Std, bei Gruppe III und IV erst nach 12 Std auf. Bei Gruppe IV wirkten sich jedoch wasserhaltige Einschlußvacuolen and ein interstitielles Ödem nachteilig aus. Der Einflua der Zellstabilisatoren and des Spüleffektes bei intermittierender Perfusion auf die Erhaltung der Ultrastruktur wurden diskutiert.
    Notes: Summary Morphological changes in pig liver were examined electronmicroscopically for 24 hrs during the following methods of preservation. Group I: singular gravity perfusion in situ with cooled, modified Ringer lactate solution and subsequent simple storage at 4° C; group II: as I but with the addition of so-called cell stabilizers to the perfusion solution (chlorpromazine 12 mg/l, Ultracorten 50 mg/l and Mg++ 4 mval); group III: initial perfusion as in II and subsequent 3-hourly intermittent shortterm perfusion with the same solution during hypothermic storage; group IV: as III but with 1-hourly intermittent perfusion.-Severe ultrastructural changes, such as disorganisation and vesiculation of the endoplasmic reticulum, mitochondrial swelling, margination of chromatin and ruptures of the plasmalemma with escape of cytoplasma constituents into the spaces of Disse, occurred in groups I and II after 6 h, in groups III and IV only after 12 h. In group IV, however, water-containing inclusion vacuoles and interstitial oedema had an unfavourable effect. The effects of cell stabilizers and washing out by intermittent perfusion on the preservation of the ultrastructure are discussed.

    Description / Table of Contents: Zusammenfassung 6 Schweinelebern wurden durch einfache Unterkühlung 24 Std konserviert und die Strukturerhaltung in verschiedenen Zeitintervallen elektronenmikroskopisch und enzymcytochemisch untersucht. Für die initiale hypotherme Kurzzeitperfusion diente eine mit Kalium und Glucose angereicherte modifizierte Ringer-Lösung, der bei 3 Lebern zusätzlich sog. Zellstabilisatoren (Chlorpromazin und Prednisolon) hinzugefügt worden waren. Schwerwiegende Schädigungen der Ultrastruktur in den Hepatocyten, wie Desorganisation, Fragmentation, Vesikulation und Degranulation des endoplasmatischen Reticulums, Mitochondrienschwellung vom Matrix-Typ, Verklumpung und Margination des Chromatins sowie Rupturen der Zellmembran mit Austritt von Cytoplasmabestandteilen und Kernen in die Disséschen Räume und Sinusoidlichtungen traten vereinzelt nach 6–9stündiger, in der Mehrzahl der Zellen jedoch erst nach 12stündiger Konservierung auf. Die Lysosomen blieben über 18 Std unverändert. Bemerkenswert waren früh einsetzende regressive Veränderungen in den Kupfferschen Sternzellen, die schon nach 6 Std vielfach zu Cytolysen führten. Der Zusatz von Chlorpromazin und Prednisolon zur Perfusionslösung blieb ohne morphologisch erfaßbaren Effekt auf die Erhaltung der Ultrastruktur.
    Notes: Summary 6 porcine livers were preserved by simple hypothermia for 24 hrs. Ultrastructural alterations were studied at various intervals with electron microscopic and enzyme cytochemical methods. A modified Ringer's solution with increased concentrations of potassium and glucose was used for initial hypothermic shorttime perfusion. In 3 of 6 cases cell stabilizers (chlorpromazine and prednisolone) were added to the perfusate. Severe ultrastructural changes such as disorganization, fragmentation, vesiculation, and degranulation of endoplasmic reticulum, swelling of the matrix in mitochondria, aggregation, and margination of chromatin as well as rupture of the plasma membrane followed by the release of cellular constituents into the Dissé spaces and the sinusoids appeared in several liver cells after 6–9 hrs of preservation. In a significant number of cells, however, they were observed only after 12 hrs of preservation. In lysosomes there were no changes for more than 18 hrs of preservation. Especially interesting were early regressive alterations of Kupffer cells leading frequently to cytolysis already after 6 hrs of preservation. The addition of chlorpromazine and prednisolone to the perfusate had no effect on the preservation of ultrastructure.

    Notes: Summary The morphological and biochemical characters of twenty nine sporulation mutants were compared. Some of the predictions made on the basis of unidirectional pleïotropic interactions were confirmed, namely that the latest proteolytic enzymes, like elastase, are related to late morphological stages. From the cytological point of view, mutants blocked at various stages were described. Among the late mutants, both coatless mutants with normal but incomplete cortex and cortexless mutants with flexible spore coats were found. Particularly interesting is the class of abnormal late sporulation mutants which form normal mature heat-resistant spores at high frequencies, but, in addition, present various anomalies in the structure of the spore coats and various sporangial inclusions such as a spongy fibrous material, resembling the cortex, and either onion-like or rod-shaped inclusions, probably formed by spore coat components. The presence of these structures is related to the derepression of elastase activity and may reflect overproduction of spore components. Several mutants also contain abnormal, large, dark, membrane-bound mesosomes, either compact or loose, whose presence is related to the lack of oxidation of tetrazolium dyes. The morphological heterogeneity of mutant populations is also noted. These findings are discussed in relation to the theory of sequential gene activation.

    Notes: Abstract Transparent and highly oriented samples of high dimensional stability were obtained by extruding crystallised linear polyethylene through a tapered die at temperatures higher than 80° C and below the melting point. The crystal orientation of the extrudate was examined by X-ray techniques, and it was found that there was a preferential a-axes orientation parallel to the radial direction of the rod-shaped extrudate. Electron micrographs of the fractured surface of the extrudate revealed closely packed thin fibrils of diameters of about 300 Å. Small angle X-ray scattering showed a long period of 200 to 300 Å along the longitudinal direction of fibrils, depending on the extruding conditions. This fact suggests the existence of the lamellar structure in the fibrils.
