Zusammenfassung Die Blutbasophilengranula von Mensch und Kaninchen erweisen sich als extrem wasserlöslich, sind jedoch nicht alkohollöslich. Der granulolytische Effekt des Wassers ist weitgehend unabhängig von den osmotischen Verhältnissen und der Wasserstoffionenkonzentration der Fixierungslösung. Eine ausreichende Fixation der Blutbasophilengranula ist dann gegeben, wenn ihre vollkommene Wasserresistenz erreicht ist. Hierfür eignen sich folgende Fixierungslösungen: 4%ige basische Bleiacetatlösung, Formol-Äthanol (1∶9) nach Schubert, frisch absolutiertes Methanol und Äthanol, Bleiacetat-Äthanol-Eisessig nach Mota. Die beste und rascheste Fixierung erzielten wir mit einer eigenen Modifikation der Motaschen Lösung, nämlich mit einem Gemisch von Bleiacetat, Formolalkohol und Eisessig. Die eigenen Versuchsergebnisse werden den Befunden anderer Autoren gegenübergestellt. Dabei wird auf die Möglichkeit artifizieller Degranulation und ihrer Fehldeutung nach ungeeigneter Fixierung hingewiesen.

Summary The granules of blood basophils of men and rabbits proved to be extremely soluble in water. However, they are not soluble in alcohol. The granulolytic effect of water is largely independent of the osmotic conditions and the hydrogen ion concentration of the fixative solution. A complete water resistance of basophilic granules manifests their sufficient fixation. The following fixatives are suitable: four per cent basic lead acetate, formol-ethanol (1∶9), recently distillated methanol and ethanol as well as lead acetate — ethanol — acetic acid after Mota. We found our own modification of Motas fixative solution, a mixture of basic lead acetate, formol alcohol and acetic acid to give the best and most time-saving preservation. Our own results are compared with the reports of other authors. The possibility of artificial degranulation and its misleading interpretation after inappropriate fixation is mentioned.
