Summary Semi-quantitative cytophotometric determinations were carried out on excised specimens from the human bronchial tree and on the epithelial changes induced in the golden hamster bronchus with di-ethylnitrosamine. Questionable precancerous stages of the human bronchial epithelium (transitional epithelium, squamous epithelial metaplasia, transitional epithelium with atypia) revealed a generally normal DNA distribution that differed greatly from that of the carcinomas in the same cases. These epithelial changes even in the presence of atypia, therefore, cannot be accepted as obligate and direct precursors of the invasive carcinoma. The criterium of an abnormal DNA distribution cannot be used in the experimentally induced metaplasia of the bronchial epithelium of the golden hamster, since the fully developed carcinomas have a normal DNA distribution with diploid stem-lines.

Zusammenfassung An Excisionsmaterial aus dem menschlichen Bronchialbaum und an experimentell mit Diäthylnitrosamin erzeugten Bronchialepithelveränderungen beim Goldhamster wurden cytophotometrische halbquantitative DNS-Bestimmungen durchgeführt: Fragliche Carcinomvorstufen des menschlichen Bronchialepithels (Übergangsepithel, Plattenepithelmetaplasie, Übergangsepithel mit Atypien) zeigen eine im wesentlichen normale DNS-Verteilung, die von der der Carcinome in den gleichen Fällen erheblich abweicht. Diese Epithelveränderungen können auch beim Vorliegen von Atypien nicht als obligate, direkte Vorstadien des invasiven Carcinoms gelten. Das Kriterium einer abartigen DNS-Verteilung konnte auf die experimentell erzeugten Bronchialepithelmetaplasien beim Goldhamster nicht angewandt werden, da die voll ausgebildeten Carcinome eine normale DNS-Verteilung mit diploider Stammlinie aufweisen.
