Summary 1. The multiplication of Mengo virus in L- and FL-cells is almost completely inhibited by thienylalanine (4.38 mM/l) if the substance is added during the first three hours after infection. Adding it only five hours after infection no inhibition can be detected. 2. Staining with fluorescent antibodies revealed viral antigen in a small percentage of cells (20–30% of the untreated virus controls) treated with thienylalanine. However, the distribution of the fluorescence differs distinctly from that in untreated cells. 3. From the results obtained it can be concluded that a) thienylalanine inhibits the formation of mature Mengo virus by disturbing the synthesis of the RNA-polymerase(s). The inhibition-sensitive phase continues at least to the third hour after infection. b) in the majority of the cells thienylalanine also inhibits the synthesis of capsid-proteins. However, in this case the inhibitor-sensitive phase is finished earlier. c) thienylalanine interferes with at least two different early proteins.

Zusammenfassung 1. Die Vermehrung von Mengovirus in L- und FL-Zellen wird durch Thienylalanin (4,38 mmol/l) nahezu vollständig gehemmt, wobei es gleichgültig ist, ob der Inhibitor gleichzeitig mit dem Virus oder nach beendeter Adsorption (bis zur 3. Stundep.i.) zugesetzt wird. In einer späteren Phase des Multiplikationszyklus (5. Stundep.i.) wird die Synthese von infektiösem Virus durch den Zusatz von Thienylalanin nicht mehr gehemmt. 2. Fluoreszenzserologisch wurde nachgewiesen, daß virales Kapsidantigen auch in Gegenwart von Thienylalanin in einem geringen Prozentsatz (20 bis 30% der Viruskontrollen) der Zellen gebildet wird, wobei jedoch die Verteilung des fluoreszierenden Proteins — im Vergleich zu unbehandelten Zellen — unterschiedlich ist. 3. Aus den Versuchsergebnissen werden die folgenden Schlüsse gezogen: a) Thienylalanin hemmt die Bildung von reifem Mengovirus durch eine gestörte Synthese der RNS-Polymerase(n), wobei die Inhibitorempfindliche Phase mindestens bis zur 3. Stundep.i. dauert; b) durch Thienylalanin wird auch die Synthese von Kapsidprotein in der Mehrzahl der Zellen gehemmt, wobei jedoch die Inhibitor-empfindliche Phase kürzer ist; c) Thienylalanin interferiert daher mit mindestens zwei verschiedenen „early proteins“.
